Antigen-specific mRNA lipid nanoparticle platforms for the prevention and treatment of allergy and autoimmune diseases

While most nanomedicines primarily aim to stimulate the immune system against infections or tumors,there is a growing demand for inducing immune tolerance under certain conditions,such as allergic and autoimmune *** have explored nanotechnology-based strategies to induce immune tolerance in a targeted and specific *** approach involves the use of nanoparticles(NPs)to encapsulate immunosuppressive drugs and/or antigens to educate naïve T cells and promote the generation of antigen-specific regulatory T cells that inhibit immune ***,this approach has certain *** hydrophobicity of proteins or peptides restricts the degree to which they can be encapsulated in NPs,which in turn,affects their loading efficiency and treatment *** the emergence of mRNA lipid nanoparticle(LNP)platforms,there is the possibility of overcoming the limitations of protein and peptide *** date,mRNA LNP systems have been shown to provide organ,cellular,and subcellular targeting for the induction of immune *** method of drug delivery is flexible and scalable that can be customized for a specific patient,resulting in an effective means of administering relevant proteins or epitopes to induce antigen-specific immune *** continued research and development,this technology could offer a safer and more effective alternative to current therapies,ultimately improving the quality of life of patients worldwide.

Shifts in the Holstein dairy cow milk fat globule membrane proteome that occur during the first week of lactation are affected by parity

Background:The milk fat globule membrane(MFGM)proteomes of colostrum and transition milk are rich sources of proteins that are likely important for neonatal calf *** addition,characterization of these proteomes could also yield valuable information regarding mammary gland physiology of the early postpartum lactating *** objectives of this research were to characterize the MFGM proteomes of colostrum and transition milk through sample collections at four timepoints postpartum,including the first milking(M1,colostrum),second milking(M2,transition milk),fourth milking(M4,transition milk),and fourteenth milking(M14,mature milk),and compare these proteomes between multiparous(MP;n=10)and primiparous(PP;n=10)Holstein dairy *** MFGM proteins were labeled using Tandem Mass tagging and analyzed using liquid chromatography-tandem mass spectrometry(Lc-MS/MS).Protein identification was completed using MAScOT and Sequest in Proteome Discoverer *** scaled abundance values were analyzed using PROc MIXED in SAS to determine the effects of milking(MIL),parity(PAR),and MIL×*** adaptive false-discovery rate(FDR)-adjusted P values were determined using PROc *** characterization and bioinformatic analysis were completed using a combination of PANTHER,Blast,and ***:A total of 104 common proteins were identified in each of the MFGM *** analysis revealed that 70.2%of identified proteins were affected by *** these,78.1%were lower in M14 compared with M1,including immune-related proteins lactotransferrin,lactadherin and *** affected 44.2%of *** the proteins affected by PAR,84.8%were higher in MP cows compared with PP cows,including apolipoprotein E and histones 2A,2B,3,and 4 *** subfamily 1 member 1A and annexin 5 were higher in samples from PP ***×parity affected 32.7%of identified proteins,including lactotransferrin,gelsolin,vitamin D binding protein,and S100 ***

Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FDA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.

Acute-on-chronic liver failure(AcLF)is a syndrome that occurs in patients with chronic liver disease and is characterized by acute decompensation,organ failure and high short-term *** due to the lack of universal diagnostic criteria,the actual AcLF prevalence remains unclear;nevertheless,it is expected to be a highly prevalent condition *** transplantation is an effective protective measure for selected AcLF *** liver transplantation,diagnosing and treating precipitant events and providing supportive treatment for organ failures are currently the cornerstone of AcLF *** new clinical specific therapies have been researched,more studies are necessary to assess safety and ***,future AcLF management strategies must consider measures to improve access to liver transplantation because the time window for this life-saving therapy is frequently ***,an urgent and global discussion about allocation and prioritization for transplantation in critically ill AcLF patients is needed because there is evidence suggesting that the current model may not portray their waitlist *** addition,while donor organ quality is meant to be a prognostic factor in the AcLF setting,recent evidence suggests that machine perfusion of the liver may be a safe tool to improve the donor organ pool and expedite liver transplantation in this scenario.

Dietary glutamine,glutamic acid and nucleotide supplementation accelerate carbon turnover(δ^(13)c)on stomach of weaned piglets

The use of stable isotope analysis as a tool for characterization of carbon turnover(δ^(13)c) in piglet's tissues by tracing its feeding system has drawn attention. Thus, this study aimed at evaluating the influence of dietary glutamine, glutamic acid and nucleotides supplementation on carbon turnover in fundic-stomach region of weaned piglets at an average age of 21 days. The diets consisted of additive-free diet — control(c); 1% glutamine(G); 1% glutamic acid(GA) and 1% nucleotides(Nu). At weaning day(day 0: baseline), 3 piglets were slaughtered to quantify the δ^(13)c of stomach. The remaining 120 piglets were blocked by weight and sex, randomly assigned to pens with 3 piglets slaughtered per treatment at days 1,2,4, 5,7,9,13.20.27 and 49 after weaning in order to verify the fundic-stomach isotopic composition by *** were analyzed in terms of ^(13)c/^(12)c ratio by mass spectrometry and converted to relative isotopic enrichment values(δ^(13)c ‰) used to plot the first order exponential curves over time using OriginPro 8.0 software. The inclusion of glutamine, glutamate and nucleotides in piglet's diets has accelerated the carbon turnover in stomach during the post-weaning period, demonstrating also that glutamate has guaranteed fastest 13 c incorporation rate on fundic-stomach region and pH-lowering. Besides that, stable isotopes technique(δ^(13)c) has proved to be an important methodology to determine the time-scales at which piglets shift among diets with different isotopic values, characterizing the trophic effects of additives and the phenotypic flexibility of stomach.

Extended criteria brain-dead organ donors:Prevalence and impact on the utilisation of livers for transplantation in Brazil

BAcKGROUND Despite its association with higher postoperative morbidity and mortality,the use of extended criteria donor(EcD)livers for transplantation has increased globally due to the high demand for the *** To investigate the prevalence of EcD in donation after brain death(DBD)and its impact on organ acceptance for *** Retrospective analysis of DBD organ offers for liver transplantation between 2017 and 2020 in a high-volume transplant *** incidence of the Eurotransplant risk factors to define an EcD(ET-EcD)among DBD donors and the likelihood of organ acceptance over the years were *** relationship between organ refusal for transplantation,the occurrence,and the number of ET-EcD was assessed by simple and multiple logistic regression *** A total of 1619 organ donors were *** these,78.31%(n=1268)had at least one ET-EcD *** was an increase in the acceptance of EcD DBD organs for transplantation(1 criterion:from 23.40%to 31.60%;2 criteria:from 13.10%to 27.70%;3 criteria:From 6.30%to 13.60%).For each addition of one ETEcD variable,the estimated chance of organ refusal was 64.4%higher(OR 1.644,95%cI 1.469-1.839,Pcept for the donor serum sodium>165 mmol/L(P=0.310),all ET-EcD criteria increased the estimated chance of organ refusal for *** A high prevalence of EcD DBD was *** the increase in their utilisation,the presence and the number of extended donor criteria were associated with an increased likelihood of their refusal for transplantation.

In this paper,the assessment of the sensitivity of the soil in the rural area of (c)ukarica municipality to the processes of degradation is *** areas,especially in the vicinity of large cities,are exposed to numerous processes of degradation:soil erosion,urban and industrial zone expansion at the expense of fertile agricultural soils,activation of landslides and a number of other significant ecological *** on the characteristics of the research area,the MEDALUS (Mediterranean Desertification and Land Use) model was applied,and for the assessment of sensitivity to the processes of degradation the main quality indicators were considered:soil,climate,vegetation and *** each of the analyzed quality indicators,parameter groups were *** parameter is quantified according to the defined method by giving them a sensitivity coefficient between 1.0 and *** 10.0 has been applied to analyze and prepare layers of quality ***,the geometric mean for all four quality indicators was used to generate the map of environmental sensitivity to *** results obtained show that 41.54% of the study area is classified as critical;22.34% of the surface as fragile;8.47% of the areas are potentially endangered and 9.58% not threatened to degradation *** results have also shown that MEDALUS model is a functional tool for simulations which support sustainable land management in the areas prone to degradation.

Dear Editor,Abscisic acid （ABA） induces turgot loss and hence stomatal closure by promoting rapid net K^＋ efflux from guard cells （Gcs） through outward-rectifying K^＋（K^＋out） channels （Schroeder et al., 1987; Blatt, 1990）. The mechanisms of ABA signaling in Gcs are detailed elsewhere （see Munemasa et al., 2015; Weiner et al., 2010; Pandey et al., 2007）. Briefly, ABA binds to the PYR/ PYL/RcARs, a family of soluble steroidogenic acute regulatory- related lipid transfer （START） proteins, and, in turn, inactivates the downstream PP2c （type 2c protein phosphatase）, leading to the activation of SnRK2.6 （SNF1 [sucrose non-fermenting-1- related protein kinase]/OST1 [open stomata 1]） protein kinases.
