Inositol Polyphosphate Phosphatidylinositol 5-Phosphatase9 （At5PTase9） Controls Plant Salt Tolerance by Regulating Endocytosis

Phosphatidylinositol 5-phosphatases （5PTases） components of membrane trafficking system. Recently, we that hydrolyze the 5＇ position of the inositol ring are key reported that mutation in AtSPTase7 gene reduced produc- tion of reactive oxygen species （ROS） and decreased expression of stress-responsive genes, resulting in increased salt sensitivity. Here, we describe an even more salt-sensitive 5ptase mutant, At5ptase9, which also hydrolyzes the 5＇ phos- phate groups specifically from membrane-bound phosphatidylinositides. Interestingly, the mutants were more tolerant to osmotic stress. We analyzed the main cellular processes that may be affected by the mutation, such as production of ROS, influx of calcium, and induction of salt-response genes. The At5ptase9 mutants showed reduced ROS produc- tion and Ca2~ influx, as well as decreased fluid-phase endocytosis. Inhibition of endocytosis by phenylarsine oxide or Tyrphostin A23 in wild-type plants blocked these responses. Induction of salt-responsive genes in wild-type plants was also suppressed by the endocytosis inhibitors. Thus, inhibition of endocytosis in wild-type plants mimicked the salt stress responses, observed in the AtSptase9 mutants. In summary, our results show a key non-redundant role of At5PTase7 and 9 isozymes, and underscore the localization of membrane-bound Ptdlns in regulating plant salt tolerance by coordinating the endocytosis, ROS production, Ca2＋ influx, and induction of stress-responsive genes.

Emergency department management of acute agitation in the reproductive age female and pregnancy

BACKGROUND:Agitation is a common presentation within emergent departments(EDs).Agitation during pregnancy should be treated as an obstetric emergency,as the distress may jeopardize both the patient and fetus.The safety of psychotropic medications in the reproductive age female has not been well established.This review aimed to explore a summary of general agitation recommendations with an emphasis on ED management of agitation during pregnancy.METHODS:A literature review was conducted to explore the pathophysiology of acute agitation and devise a preferred treatment plan for ED management of acute agitation in the reproductive age or pregnant female.RESULTS:While nonpharmacological management is preferred,ED visits for agitation often require medical management.Medication should be selected based on the etiology of agitation and the clinical setting to avoid major adverse effects.Adverse effects are common in pregnant females.For mild to moderate agitation in pregnancy,diphenhydramine is an effective sedating agent with minimal adverse effects.In moderate to severe agitation,high-potency typical psychotropics are preferred due to their neutral effects on hemodynamics.Haloperidol has become the most frequently utilized psychotropic for agitation during pregnancy.Second generation psychotropics are often utilized as second-line therapy,including risperidone.Benzodiazepines and ketamine have demonstrated adverse fetal outcomes.CONCLUSION:While randomized control studies cannot be ethically conducted on pregnant patients requiring sedation,animal models and epidemiologic studies have demonstrated the effects of psychotropic medication exposure in utero.As the fetal risk associated with multiple doses of psychotropic medications remains unknown,weighing the risks and benefits of each agent,while utilizing the lowest effective dose remains critical in the treatment of acute agitation within the EDs.

Development of a building information model-guided post-earthquake building inspection framework using 3D synthetic environments

Computer vision-based inspection methods show promise for automating post-earthquake building inspections.These methods survey a building with unmanned aerial vehicles and automatically detect damage in the collected images.Nevertheless,assessing the damage′s impact on structural safety requires localizing damage to specific building components with known design and function.This paper proposes a BIM-based automated inspection framework to provide context for visual surveys.A deep learning-based semantic segmentation algorithm is trained to automatically identify damage in images.The BIM automatically associates any identified damage with specific building components.Then,components are classified into damage states consistent with component fragility models for integration with a structural analysis.To demonstrate the framework,methods are developed to photorealistically simulate severe structural damage in a synthetic computer graphics environment.A graphics model of a real building in Urbana,Illinois,is generated to test the framework;the model is integrated with a structural analysis to apply earthquake damage in a physically realistic manner.A simulated UAV survey is flown of the graphics model and the framework is applied.The method achieves high accuracy in assigning damage states to visible structural components.This assignment enables integration with a performance-based earthquake assessment to classify building safety.

In the world between 1 and 2 billion people have obesity, In the United States, which has the highest obesity rate in the world, one in three people have obesity. Obesity rates are increasing in every country in which it has taken hold.^1 Obesity rates also affect all ages from the growing elderly-obese population to the dramatic increase in pediatric obesity, for example in the United States it is predicted that by the end of the decade one in two US children will have obesity. Obesity similarly affects all races and both sexes. As a consequence of the co-morbidities of obesity and of the associated costs such as days off work, obesity costs the US economy about 100-200 billion dollars per year.

The many faces of p53:something for everyone

The p53 gene,from discovery to classification:the first 10 years Forty years ago four research laboratories in London,Paris,NewYork/Bethesda,and Princeton uncovered the existence of the p53 protein (Deleo et al.,1979;Lane and Crawford,1979;Linzer and levine,1979;Kress et al.,1979).Each laboratory came upon this protein for a different reason and with a different experimental approach that uncovered this unanticipated result.Together,the four papers permitted one to con elude the following:(i) in SV40-infected and transformed cells the SV40-e ncoded on cogene protein,the large T-antigen,formed a protein complex with a cellular-encoded protein of -53000 daltons in size,(ii) This p53 protein was detected at high levels in a variety of transformed cells derived from viral,chemical,or inherited (teratocarcinomas) transformation events,(iii) Nontransformed cells expressed lower levels of the p53 protein,(iv) Animals bearing tumors produced antibodies directed against the p53 protein.

An overview of the management of post-vasectomy pain syndrome

Post-vasectomy pain syndrome remains one of the more challenging urological problems to manage. This can be a frustrating process for both the patient and clinician as there is no well-recognized diagnostic regimen or reliable effective treatment. Many of these patients will end up seeing physicians across many disciplines, further frustrating them. The etiology of post-vasectomy pain syndrome is not clearly delineated. Postulations include damage to the scrotal and spermatic cord nerve structures via inflammatory effects of the immune system, back pressure effects in the obstructed vas and epididymis, vascular stasis, nerve impingement, or perineural fibrosis. Post-vasectomy pain syndrome is defined as at least 3 months of chronic or intermittent scrotal content pain. This article reviews the current understanding of post-vasectomy pain syndrome, theories behind its pathophysiology, evaluation pathways, and treatment options.
