Objective: To examine the effect of combination of Andrographis paniculata herb fraction(AHF) and Centella asiatica herb fraction(CHF) on PPAR γ and GLUT4 mRNA expression in 3 T3-L1 adipocyte, and its effect on insulin-stimulated glucose uptake. Methods: 3 T3 L1 adipocyte cells were used to investigate gene expression of PPARy reverse transcription-polymerase chain reaction method. The γ and GLUT4 protein b adipocyte cells were differentiated by using insulin, dexamethasone and 3-isobutyl-1-methylxanthine from 3 T3-L1 cells. Pioglitazone, AHF, CHF and the combination of both herbs were evaluated on glucose uptake activity, PPAR γ and GLUT4 mRNA expressions in 3 T3-L1 adipocyte. Results: The results showed that combination of AHF at 30 μg/mL and CHF at 10 μg/mL could enhance insulin-stimulated glucose uptake. The combination also increased PPAR γ and GLUT4 mR NA expressions significantly in comparison to those of negative control(DMSO). These effects were equal in comparison to those of pioglitazone(0.02 μM) and its single extracts Conclusions: The combination of AHF and CHF can increase glucose uptake and insulin sensitivity through up-regulation of PPAR γ and GLUT4 mRNA expressions in 3 T3-L1 adipocyte.

One of the impacts of the Fukushima disaster was the shutdown of all nuclear power plants in Japan,reaching zero production in 2015.In response,the country started importing more fossil energy including coal,oil,and natural gas to fill the energy gap.However,this led to a significant increase in carbon emissions,hindering the efforts to reduce its carbon footprint.In the current situation,Japan is actively working to balance its energy requirements with environmental considerations,including the utilization of hydrogen fuel.Therefore,this paper aims to explore the feasibility and implications of using hydrogen power plants as a means to reduce emissions,and this analysis will be conducted using the energy modeling of the MARKAL-TIMES Japan framework.The hydrogen scenario(HS)is assumed with the extensive integration of hydrogen into the power generation sector,supported by a hydrogen import scheme.Additionally,this scenario will be compared with the Business as Usual(BAU)scenario.The results showed that the generation capacities of the BAU and HS scenarios have significantly different primary energy supplies.The BAU scenario is highly dependent on fossil fuels,while the HS scenario integrates hydrogen contribution along with an increase in renewable energy,reaching a peak contribution of 2,160 PJ in 2050.In the HS scenario,the target of reducing CO_(2) emissions by 80%is achieved through significant hydrogen penetration.By 2050,the total CO_(2) emissions are estimated to be 939 million tons for the BAU scenario and 261 million tons for the Hydrogen scenario.In addition,the contribution of hydrogen to electricity generation is expected to be 153 TWh,smaller than PV and wind power.

Assessment of corrected time-step method for nominal ionospheric gradient calculation: A comparative analysis with spatial approaches

The effect of ionospheric delay on the ground-based augmentation system under normal conditions can be mitigated by determining the value of the nominal ionospheric gradient(σvig).The nominal ionospheric gradient is generally obtained from Continuously Operating Reference Stations data by using the spatial single-difference method(mixed-pair,station-pair,or satellite-pair)or the temporal single-difference method(time-step).The time-step method uses only a single receiver,but it still contains ionospheric temporal variations.We introduce a corrected time-step method using a fixed-ionospheric pierce point from the geostationary equatorial orbit satellite and test it through simulations based on the global ionospheric model.We also investigate the effect of satellite paths on the corrected time-step method in the region of the equator,which tends to be in a more north–south direction and to have less coverage for the east–west ionospheric gradient.This study also addresses the limitations of temporal variation correction coverage and recommends using only the correction from self-observations.All processes are developed under simulations because observational data are still difficult to obtain.Our findings demonstrate that the corrected time-step method yieldsσvig values consistent with other approaches.

The Mutis-Timau Forest Complex,located on Timor Island,Indonesia,is a mountainous tropical forest area that gradually decreases due to deforestation and forest degradation.Previous modelling studies based on patterns indicate that deforestation primarily occurs at lower elevations and near the boundaries of forests and settlements,often associated with shifting cultivation by local farmers.This study adopts a process-based modelling approach,specifically the agent-based model,to simulate land changes,particularly farmers'expansion of agricultural land around the Mutis mountain forest.The underlying concept of this agent-based approach is the interaction between the human and environmental systems.Farmers,representing the human system,interact with the land,which represents the environmental system,through land use decision-making mechanisms.The research was conducted in the Community Forest of the Timor Tengah Utara District,one of the sites within the Mutis-Timau Forest Complex with the highest deforestation rate.Land use change simulations were performed using agent-based modelling from 1999 to 2030,considering the socio-economic conditions of farmers,spatial preferences,land use decisions,and natural transitions.The results revealed that the agricultural area increased by 14%under the Business as Usual scenario and 5%under the Reducing Emission from Deforestation and Forest Degradation scenario,compared to the initial agricultural area of 245 hectares.The probability of farmers deciding to extend agricultural activities was positively associated with the number of livestock maintained by farmers and the size of the village area.Conversely,the likelihood of farmers opting for agricultural extensification decreased with an increase in the area of private land and the farmer's age.These findings are crucial for the managers of the Mutis-Timau Forest Complex and other relevant stakeholders,as they aid in arranging actions to combat deforestation,designing proper forest-related policies,and providing support for in

Background:Transmembrane 4 L six family member 5(TM4SF5)translocates subcellularly and functions metabolically,although it is unclear how intracellu-lar TM4SF5 translocation is linked to metabolic contexts.It is thus of interests to understand how the traffic dynamics of TM4SF5 to subcellular endosomal membranes are correlated to regulatory roles of metabolisms.Methods:Here,we explored the metabolic significance of TM4SF5 localization at mitochondria-lysosome contact sites(MLCSs),using in vitro cells and in vivo animal systems,via approaches by immunofluorescence,proximity labelling based proteomics analysis,organelle reconstitution etc.Results:Upon extracellular glucose repletion following depletion,TM4SF5 became enriched at MLCSs via an interaction between mitochondrial FK506-binding protein 8(FKBP8)and lysosomal TM4SF5.Proximity labeling showed molecular clustering of phospho-dynamic-related protein I(DRP1)and certain mitophagy receptors at TM4SF5-enriched MLCSs,leading to mitochondrial fis-sion and autophagy.TM4SF5 bound NPC intracellular cholesterol transporter 1(NPC1)and free cholesterol,and mediated export of lysosomal cholesterol to mitochondria,leading to impaired oxidative phosphorylation but intact tri-carboxylic acid(TCA)cycle andβ-oxidation.In mouse models,hepatocyte Tm4sf5 promoted mitophagy and cholesterol transport to mitochondria,both with positive relations to liver malignancy.Conclusions:Our findings suggested that TM4SF5-enriched MLCSs regu-late glucose catabolism by facilitating cholesterol export for mitochondrial reprogramming,presumably while hepatocellular carcinogenesis,recapitulating aspects for hepatocellular carcinoma metabolism with mitochondrial repro-gramming to support biomolecule synthesis in addition to glycolytic energetics.

Macronutrient deficiency inhibits the growth and development of chili plants.One of the non-destructive methods that plays a role in processing plant image data based on specific characteristics is computer vision.This study uses 5166 image data after augmentation process for six plant health conditions.But the analysis of one feature cannot represent plant health condition.Therefore,a careful combination of features is required.This study combines three types of features with HSV and RGB for color,GLCM and LBP for texture,and Hu moments and centroid distance for shapes.Each feature and its combination are trained and tested using the same MLP architecture.The combination of RGB,GLCM,Hu moments,and Distance of centroid features results the best performance.In addition,this study compares the MLP architecture used with previous studies such as SVM,Random Forest Technique,Naive Bayes,and CNN.CNN produced the best performance,followed by SVM and MLP,with accuracy reaching 97.76%,90.55%and 89.70%,respectively.Although MLP has lower accuracy than CNN,the model for identifying plant health conditions has a reasonably good success rate to be applied in a simple agricultural environment.

The Use of Single-Phase Immersion Cooling by Using Two Types of Dielectric Fluid for Data Center Energy Savings

Data centers are recognized as one of the most important aspects of the fourth industrial revolution since conventional data centers are inefficient and have dependency on high energy consumption,in which the cooling is responsible for 40%of the usage.Therefore,this research proposes the immersion cooling method to solving the high energy consumption of data centers by cooling its component using two types of dielectric fluids.Four stages of experimentalmethods are used,such as fluid types,cooling effectiveness,optimization,and durability.Furthermore,benchmark software is used to measure the CPU maximum work with the temperature data performed for 24 h.The results of this study show that the immersion cooling reduces 13℃ lower temperature than the conventional cooling method which means it saves more energy consumption in the data center.The most optimum variable used to decrease the temperature is 1.5 lpm of flow rate and 800 rpm of fan rotation.Furthermore,the cooling performance of the dielectric fluids shows that the mineral oil(MO)is better than the virgin coconut oil(VCO).In durability experiment,there are no components damage after five months immersed in the fluid.

Soil health depletion due to intensive tillage operations is a global issue in the agricultural sector.Conservation tillage(CT)which involves non-inversion tillage and leaving~30%of the soil surface covered with crop residues,is a strategy designed to enhance soil health.However,no comprehensive study to investigate the long-term effect of CT on soil biological activity and the soil nutrient supply has yet been widely carried out.Biological and chemical soil properties were assessed at depths 0-5,10-15,and 20-25 cm depths after 18 years of CT and conventional tillage practice(PT).Various stages in the vegetative growth of maize were investigated in 2021 in Hungary.The findings indicated that tillage intensity,soil depth,and growth stages all significantly influenced soil enzyme activities and the concentration of soil nutrients.Less soil disturbance resulted in a significantly larger concentration of soil carbon parameters(total organic carbon and labile carbon)in CT plots,where the activity ofβ-glucosidase and dehydrogenase(DHA)in the upper soil layer increased significantly(0.7-2.6 and 2.6-4.7 times,respectively)compared to PT.The high amount of organic matter and the greater resistance to erosion observed in CT also contributed to the higher concentration of available nutrients(NH_(4),NO_(3),Ca,K)and total P in the surface soil layer.Phosphatase activity was highest in the mid-stage of vegetative growth and was positively correlated to the total P concentration.The alterations in soil water content were clearly negatively corelated with the change in DHA and phosphatase activity.Overall,due to the more balanced environmental conditions,the decomposition of organic substances was more balanced and slower in CT than in PT.This implied that the mobilization of nutrients in the soil was more balanced as well,and that the nutrients were released gradually.The enhancement of the soil nutrient-supplying capacity achieved by means of long-term conservation tillage provides a promising strategy for sustainable nutrient

A cruciate ligament avulsion is a much less common form of injury than a cruciate ligament tear.Simultaneous tibial avulsion fractures of both cruciate ligaments occur even more rarely.Over the last decades,many studies have described arthroscopic fixation of acute cruciate tibial avulsion fractures,but arthroscopic treatment in a late presenting patient has not been reported in the literature.This case report presents a 32-year-old female with a chronic tibial avulsion fracture of both anterior cruciate ligament and posterior cruciate ligament.Simultaneous fixation of both fractures was performed arthroscopically at week four post-injury.At one year of follow-up,the patient had demonstrated full knee range of motion and stable knee with no complaints,and achieved excellent clinical outcomes.Radiographs showed union of both fractures,and the patient had resumed high-impact exercises.

Prospects and Challenges of Utilizing Solar Energy for the COVID-19 Vaccine Cold Storage in Remote Clinics of Tropical Countries:Review

The rapid spread of COVID-19 pandemic has forced several countries in the world to store vaccines in cold storage towards ensuring their protection from being damaged and to maintain their stability.However,most remote clinics,especially those in the equator and islands,are faced with the challenges of hot climates and the inability to afford electricity resources needed to power the cold storage facility.Meanwhile,the hot equatorial region has abundant solar energy to power the vaccine cold storage but previous studies showed that several field workers do not have the ability to maintain the vaccine storage temperature as indicated by the manufacturer’s recommendations.Therefore,this literature review study examines the prospects and challenges of implementing solar-powered cold storage to provide cooling space for remote clinics.This is expected to contribute significantly to cold chain vaccine management technology.The findings showed that four technology integration schemes including Cold Storage Ice Maker,Cold Storage Ice Maker-PCM,Refrigerator-Ice Maker and,Absorbing Cooling-PV-Ice Maker have the potential to be applied in clinics situated in certain tropical regions.
