The beijing genomics institute (BGI) of Chinese Academy of Sciences (CAS) wasofficially founded in December 2003. Its predecessor, beijing Huada genomics Research Center, haspresented significant contributions to the development of life sciences in china by its excellentscientific innovations and achievements in the last five years. From the participation ofInternational Human Genome Project (HGP) to the independent completion of the whole-genome draft andfine maps of Chinese super hybrid rice genome, from the porcine genome research cooperated withDenmark to the international chicken genome research, and from the silkworm genome projectaccomplished together with The Southwest Agriculture University of china to the great contributionsto the national campaign against SARS, all these facts illustrate that BGI not only has afirst-class scientific research power in the world but also is full of devotion to the flourish ofscience and nation. During these years, we have upheld the strategy of ''Advance through projects,strive for innovation, grow through cooperation'', in the aim of making BGI as one of the pioneers ofCAS climbing the peak of life sciences in the world.

A Putative NES Mediates Cytoplasmic Localization of Apoptin in Normal Cells

Apoptin, a protein expressed by chicken anemia virus, is found predominantly in the cytoplasm in normal cells, whereas it localizes in the nucleus in transformed and malignant cells. However, the mecha- nisms that regulate the different subcellular localization of Apoptin in normal and tumor cells have not been fully clarified. In this work, a putative nuclear export signal (NES) in Apoptin was predicted. It was testified that the putative NES (pNES) of Apoptin was not a functional NES, but actually acted as a cytoplasmic retention signal. Deletion of the pNES led to the nuclear accumulation of Apoptin in normal cells. In addition, when a strong nuclear localization signal was introduced into Apoptin, it exclusively translocated to the nucleus in normal cells. These observations indicated that the cytoplasmic localization of Apoptin in normal cells results from the balance between cytoplasmic retention and nuclear import. On the other hand, the pNES was also proved to be necessary for Apoptin multimerization. Mutants lacking the pNES did not form obviously visible globular aggregates in normal or tumor cells.

The capacity for self-renewal and differentiation of human embryonic stem(ES)cells makes them a potential source forgeneration of pancreatic beta cells for treating type I diabetes ***,we report a newly developed and effectivemethod,carried out in a serum-free system,which induced human ES cells to differentiate into insulin-producing *** A was used in the initial stage to induce definitive endoderm differentiation from human ES cells,as detectedby the expression of the definitive endoderm markers Sox 17 and ***,all-trans retinoic acid(RA)wasused to promote pancreatic differentiation,as indicated by the expression of the early pancreatic transcription factorspdr1 and *** maturation in DMEM/F12 serum-free medium with bFGF and nicotinamide,the differentiatedcells expressed islet specific markers such as C-peptide,insulin,glucagon and *** percentage of C-peptide-posi-tive cells exceeded 15%.The secretion of insulin and C-peptide by these cells corresponded to the variations in *** transplanted into renal capsules of Streptozotocin(STZ)-treated nude mice,these differentiated human EScells survived and maintained the expression of beta cell marker genes,including C-peptide,pdx1,glucokinase,nkx6.1,IAPP,pax6 and *** percent of the transplanted nude mice exhibited apparent restoration of stable euglycemia;and the corrected phenotype was sustained for more than six *** new method provides a promising in vitro dif-ferentiation model for studying the mechanisms of human pancreas development and illustrates the potential of usinghuman ES cells for the treatment of type I diabetes mellitus.

The number of Hepatitis B virus (HBV) whole genomic sequences in public nucleotide databases (GenBank, EMBL, and DDBJ) had reached 866 by January 1, 2007. Coming from 46 countries and regions, these sequences were categorized as eight genotypes (A–H). With the statistical and phylogenetic analysis on all available complete genomic data of HBV, we here present an overview of HBV sequences in public databases. From all registered 229 HBV genomes in Chinese regions as well as 59 sequencing data from our research group, we report the establishment of reference sequences of HBV strains prevailing in china. These analyses provide clues for the effects of HBV genotypes in host clinical progressions, geographic distribution of the infection, and the viral evolutionary history. Moreover, the viral sequence reference would be helpful in the identiffcation of various HBV mutations. Based on the analysis of various public databases, we suggest that the Chinese HBV database with the clinical information should be constructed.

The NiAl based materials including NiAl-TiC-Al2O3 composite,NiAl-Cr(Mo)-Hf-Ho eutectic alloy and NiAl-Cr(Mo)-CrxSy in situ composite were fabricated and their wear properties were tested at different *** results revealed that the NiAl-TiC-Al2O3 composite,NiAl-Cr(Mo)-Hf-Ho eutectic alloy and NiAl-Cr(Mo)-CrxSy in situ composite exhibited the excellent wear properties between 700℃ and 900℃.The microstructure observations exhibited that the self-lubricant films formed on the worn surfaces during the dry sliding test at high temperature,which decreased the wear rate and friction coefficient *** observation on the self-lubricant film revealed that it was mainly comprised by ceramic amorphous and *** with the NiAl-TiC-Al2O3 composite,the NiAl-Cr(Mo)-CrxSy in situ composite has lower friction coefficient at low *** phenomena may be ascribed to the addition of sulfide which contributes much to the formation of self-lubricant,and moreover the TiC addition increase the strength of NiAl based material and its wear resistance.

Non-recurring-engineering(NRE)and time-to-market play an increasingly important role in the field of IC ***,with the continuous development of IC manufacturing technology,it is necessary to propose a novel design methodology to shorten design cycle and enhance design *** this paper,operator design methodology(ODM)is presented and an H.264 encoder is implemented as a *** to the flow of ODM,the register transfer level(RTL)design of H.264 encoder has been accomplished with 15 man-months,which is lower than the average 19 man-months in the field of traditional application specific integrated circuit(ASIC)***,with the advantage of operator design library,the design has a comparable performance with other ASIC *** obtained design can support a real-time video encoding of 720p at 60 frames per second or 1080p at 30 frames per second,working at 167 MHz with SMIC 0.13 μm CMOS *** results provide good evidence for the practicability and efficiency of ODM.

Induction of chromosomal inversion by integration of T-DNA in the rice genome

Transfer DNA （T-DNA） of Agrobacterium tumefaciens integration in the plant genome may lead to rearrangements of host plant chromosomal fragments, including inversions. However, there is very little information concerning the inversion. The present study re- ports a transgenic rice line selected from a T-DNA tagged population, which displays a semi-dwarf phenotype. Molecular analysis of this mutant indicated an insertion of two tandem copies of T-DNA into a locus on the rice genome in a head to tail mode. This insertion of T-DNA resulted in the inversion of a 4.9 Mb chromosomal segment. Results of sequence analysis suggest that the chromosomal inversion resulted from the insertion of T-DNA with the help of sequence microhomology between insertion region of T-DNA and target sequence of the host plant.
