Effect of Xiaofenghuoxue decotion on Th1 and Th2 cytokine production in BALF in allergen-induced asthma guinea pigs

Objective To investigate the effect of Xiaofenghuoxue decotion(XFHX)on the production of Th1 and Th2 cytokine in bronchoalveolar lavage fluid(BALF)in asthma guinea pigs sensitized and challenged with *** The animals were randomly divided into 5 groups:the normal control group,the asthma group,the XFHX high dose group,the XFHX low dose group and the budesonide group.2 weeks after sensitization with a mixture of 10% OVA and 10% Al(OH)3,the drugs were given to the animals for 7 days,and then the animals were challenged with aerosol of 1% ovalbumin,the latency time for the onset of respiratory abnormalities was examined,BALF was collected 16 h *** in BALF was *** level of IFN-γ reduced by Th1 cells and IL-4 and IL-5 produced by Th2 cells were determined by *** contents were also *** The latency time for the onset of respiratory abnormalities and leukocyte infiltration in both XFHX high dose group and low dose group significantly decreased compared with the asthma *** level of IFN-γ in both XFHX high dose group and low dose group significantly increased compared with the asthma group,the level of IL-4 and IL-5 in both XFHX high dose group and low dose group significantly decreased compared with the asthma *** production of NO was inhibited in both XFHX high dose group and low dose group compared with the asthma *** These results suggest that the anti-asthma effect of Xiaofenghuoxue decotion is associated with NO regulated reversal of Th1/Th2 cytokine balance in allergen-induced asthma guinea pigs.

To investigate the effect of signal transducers and activators of transcription 1 （STAT1） antisense oligonucleotides （ASON） on concentrations of TNF-α IL-8, NO secreted by alveolar macrophages （AMs） in bleomycin-induced rat pulmonary fibrosis, five adult female Wistar rats were intratracheally instilled with bleomycin. After 7 days, the rats were killed by right ventricle of heart exsanguinations under ketamine anaesthesia and bronchoalveolar lavage （BAL） was performed to obtain AMs. AMs were divided into four groups, treated with STAT1 ASON, STAT1 sense oligonucleotides （SON）, dexamethasone （DEX） and medium alone （control）, respectively. AMs and media were collected after culture for 36 h. The mRNA and protein expressions of STAT1 and ICAM-1 in AMs were detected by RT-PCR and ELISA, respectively. The concentrations of TNF-α IL-8, NO in cultured medium were detected. The STAT1 mRNA expression by AMs in the STAT1 ASON group was lower than those of AMs in the STAT1 SON group, the DEX group and the control group （p 〈 0.05）. Moreover, the STAT1 mRNA expression by AMs in the DEX group was also lower than those of AMs in the STAT1 SON group and the control group （p 〈 0.05）, but the STAT1 mRNA expression by AMs in the STAT1 SON group was not different from that of the control group （p 〉 0.05）. The protein expressions of STAT1 and ICAM-1 and the mRNA expression of ICAM-1 showed similar changes to the STAT1 mRNA expression by AMs. The concentrations of TNF-α IL-8, NO in cultured medium from STAT1 ASON group were lower than those from STAT1 SON, DEX and the control groups （p 〈 0.05）. Moreover, the concentrations of TNF-α IL-8, NO in cultured medium from DEX group were also lower than those from the control and STAT1 SON group （p 〈 0.05）, but no difference between STAT1 SON group and the control （p 〉 0.05）. The results suggest that STAT1 ASON could inhibit the secretion of TNF-α IL-8, NO in AMs, and STAT1 could become a target of treating pulmonary fibrosis. Cellular ＆ Molecul

I read with great interest the article by Collaborating Research Group for Noninvasive Mechanical Ventilation of Chinese respiratory Society.~1 Based on the concept mentioned in this paper, I have found that it is really an encouraging new finding in the field of clinical application of mechanical ventilation and treatment of acute respiratory failure (ARF) of chronic obstructive pulmonary disease (COPD).

CCR3 monoclonal antibody inhibits airway eosinophilic inflammation and mucus overproduction in a mouse model of asthma

Aim:To explore the effect of a rat anti-mouse CC-chemokine receptor-3(CCR3)monoclonal antibody(CCR3 mAb)on airway eosinophilia and mucus overpro-duction in asthmatic ***:An asthma model was sensitized and chal-lenged by ovalbumin(OVA)in male C57BL/6 *** mice were givendual administration(intraperitoneal injection and aerosol inhalation)of CCR3 mAbor nonspecific rat IgG(ns-IgG).The number of total and differential inflammatorycells in the bronchial alveolar lavage fluid(BALF)was *** number,the goblet cell percentage(GCP)and airway mucus index(AMI)were measured inthe lung ***(IL)-5 levels in the BALF were *** expres-sion of MUC5AC and the epidermal growth factor receptor(EGFR)mRNA in thelung tissues was detected by semi-quantitative *** results were com-pared among the ***:CCR3 mAb significantly suppressed theincreased eosinophils in the BALF and lung tissues in OVA-challenged mice com-pared with ns-IgG-treated ***-5 levels in the BALF in CCR3 mAb and ns-IgGadministration mice exhibited no obvious changes relative to OVA-challengedasthmatic ***3 mAb reduced the increased GCP and AMI after OVA chal-lenge and decreased the enhanced expression of MUC5AC and EGFR mRNA inlung tissues in asthmatic ***:CCR3 mAb can significantlyinhibit airway eosinophilia and mucus overproduction in asthmatic ***-age of CCR3 may represent a new strategy to asthma therapy.

Inhibitory effect of acetamide-45 on airway inflammation and phosphodiesterase 4 in allergic rats

Aim:To determine the effects of acetamide-45 on respiratory function,airwayinflammation,and the activity of phosphodiesterase 4（PDE4）in allergic ***:Rats were sensitized by a single intramuscular injection with ovalbumin（OVA）and were challenged with ovalbumin applied by using an aerosol repeat-edly for 7 d after 2 ***-45 at concentrations of 5,10,or 30 mg/kg wasthen administered by intraperitoneal *** in dynamic lung compli-ance and lung resistance,the accumulation of inflammatory cells in bronchoalveolarlavage,PDE4 activity,and the concentration of interleukin-4 in rat lung tissuewere ***:Seven days of treatment with acetamide-45 preventedeosinophil accumulation in allergic *** doses of 5,10,and 30 mg/kg,aceta-mide-45 decreased lung resistance to 0.20±0.04,0.25±0.07,and 0.22±0.05cmH2O·s-1·mL-1,respectively（P41±0.07,0.39±0.06,and 0.42±0.09 mL/cmH20（P45,the PDE4 activities inlung tissue were 281±55,273±57,and 238±36 nmol·g-1·min-1（P4 in lung tissue were 6.22±1.13,5.95±1.20,and 5.68±2.20 μtg/g protein（P45 wasfound to improve respiratory function and inhibit airway inflammation in thisanimal model,and the PDE4 activity of lung tissue was obviously ***-45 was an effective anti-inflammatory agent in respiratory inflammation,and the mechanism of its action might depend on inhibition of PDE4.

Aim:HMGB 1 （high-mobility group box-1） is a nuclear protein containing a con-sensus RB （retinoblastoma）-binding LXCXE *** this study,we studied thepotential association of HMGB 1 and RB and the in vitro and in vivo activities ofHMGB 1 in human breast cancer ***:The protein-protein interactionwas determined by immunoprecipitation-Western blotting and glutathione-S-trans-ferase capture assays;cell growth and radiosensitivity were examined by cellcounts,MTT assay,and clonogenic assay;cell cycle progression and apoptosiswere evaluated using flow cytometry;and the antitumor activity of HMGB 1 wasexamined with tumor xenografts in nude ***:HMGB 1 was associatedwith RB via a LXCXE motif-dependent *** 1 enhanced the abilityof RB for E2F and cyclin A transcription *** increased expression ofHMGB 1 conferred an altered phenotypes characterized by the suppression of cellgrowth;G1arrest and apoptosis was induced in MCF-7 cells containing the wild-type retinoblastoma （Rb） gene,but showed no activities in BT-549 cells contain-ing the Rb gene *** HMGB 1-induced apoptosis accompanied by caspase3 activation and PARP (poly（ADP-ribose）polymerase) *** 1 elevatedthe radiosensitivity of breast cancer cells in both the MCF-7 and BT-549 cell *** enhanced expression of HMGB 1 caused a suppression of growth of MCF-7tumor xenografts in nude mice,while LXCXE-defective HMGB 1 completely lostantitumor growth ***:HMGB 1 functions as a tumor suppressorand radiosensitizer in breast cancer.A HMGB 1-RB interaction is critical for theHMGB 1-mediated transcriptional repression,cell growth inhibition,G1cell cyclearrest,apoptosis induction,and tumor growth suppression,but is not required ***,it may be possible to design new therapies for thetreatment of breast cancer that exert their effects by modulating the HMGB 1 andRB regulatory pathway and HMGB 1-related gene therapy.
