Optical coherence tomography (OCT) noninvasively quantifies the thickness of the retinal nerve fiber layer (RNFL). OCT has been studied in several neuro-ophthalmic conditions, including Parkinson’s disease (PD). Recent studies suggest that the quantitative analysis of RNFL can be precisely and noninvasively done by OCT scans and the results suggest that the thickness of RNFL is significantly decreased in patients with PD compared with age-matched controls and the foveal retinal thickness correlates with disease severity in PD. In this article, the application of OCT imaging of the retina in PD was reviewed. Literature survey of PubMed was carried out using the search terms of "Optical Coherence Tomography" combined with "Parkinson’s Disease" and "retinal nerve fiber layer" (without restriction to the year of publication). Some related articles were also included. The search was completed in Jul. 2.11 and revised and updated as necessary. The aim of this article is to review the current literatures on the use of optical coherence tomography in patients affected by PD and to enhance its use in clinical practice in neuro-ophthalmology.

Phosphorylation/dephosphorylation is probably the most common and important reversible post-translational modification of proteins. Analyzing the functional effects of phosphorylation is helpful for understanding the biological functions of proteins. Identification of the phosphorylation sites of phosphorylated protein is a prerequisite for research on phosphorylation. In this work, an effective and simple method of identification of protein phosphorylation sites has been developed. Phosphopeptides were selectively enriched with immobilized metal affinity chromatography (IMAC) and subsequently chemically modified by 4-sulfophenyI isothiocyanate, and then the chemically modified phosphopeptides were sequenced with post-source decay (PSD) matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry for detecting phosphorylation sites. The charge of derivatization by 4-sulfophenyl isothiocyanate introduces a negative sulfonic acid group at the N-terminus of a peptide, and enables the selective detection of only a single series of C-terminal y-type ions. This chemically assisted method greatly simplifies the extremely complex pattern of PSD fragment ions and makes the PSD spectra more easier to be interpreted. The phosphorylation sites of a synthesized model phosphopeptide and human c-myc protein have been successfully identified by this method.

AIM: To investigate the effect of protein kinase C (PKC) on transforming growth factor-β2.(TGFβ2. and dopamine in retinal Müller cells of guinea pig myopic eye. METHODS: Myopia was induced by translucent goggles in guinea pig, whose retinal Müller cells were cultured using the enzyme-digesting method. Retinal Müller cells were divided into 5 groups: normal control, myopia, myopia plus GF1092.3X, myopia plus PMA, myopia plus DMSO. PKC activities were detected by the non-radioactive methods. TGFβ2.and tyrosine hydroxylase (TH) proteins were analyzed by Western Blotting in retinal Müller cells. Dopamine was determined by the high-performance liquid chromatography- electrochemical detection in suspensions. RESULTS: After 14 days deprived, the occluded eyes became myopic with ocular axle elongating. Müller cells of guinea pigs were obtained using enzyme digestion. Compared with normal control group, the increase in PKC activity and the up-regulation in TGFβ2.expression were found in retinal Müller cells of myopic eyes, with the decrease of TH and dopamine content (P 2.and TH content were up-regulated with the increase of dopamine content (P 2.3X, proteins of TGFβ2.and TH were down-regulated in the myopic eyes, with the decrease of dopamine content (P 2.and dopamine are modulated by PKC in Müller cells of the myopic eyes in guinea pig.

To investigate the effect of signal transducers and activators of transcription 1 （STAT1） antisense oligonucleotides （ASON） on concentrations of TNF-α IL-8, NO secreted by alveolar macrophages （AMs） in bleomycin-induced rat pulmonary fibrosis, five adult female Wistar rats were intratracheally instilled with bleomycin. After 7 days, the rats were killed by right ventricle of heart exsanguinations under ketamine anaesthesia and bronchoalveolar lavage （BAL） was performed to obtain AMs. AMs were divided into four groups, treated with STAT1 ASON, STAT1 sense oligonucleotides （SON）, dexamethasone （DEX） and medium alone （control）, respectively. AMs and media were collected after culture for 36 h. The mRNA and protein expressions of STAT1 and ICAM-1 in AMs were detected by RT-PCR and ELISA, respectively. The concentrations of TNF-α IL-8, NO in cultured medium were detected. The STAT1 mRNA expression by AMs in the STAT1 ASON group was lower than those of AMs in the STAT1 SON group, the DEX group and the control group （p 〈 0.05）. Moreover, the STAT1 mRNA expression by AMs in the DEX group was also lower than those of AMs in the STAT1 SON group and the control group （p 〈 0.05）, but the STAT1 mRNA expression by AMs in the STAT1 SON group was not different from that of the control group （p 〉 0.05）. The protein expressions of STAT1 and ICAM-1 and the mRNA expression of ICAM-1 showed similar changes to the STAT1 mRNA expression by AMs. The concentrations of TNF-α IL-8, NO in cultured medium from STAT1 ASON group were lower than those from STAT1 SON, DEX and the control groups （p 〈 0.05）. Moreover, the concentrations of TNF-α IL-8, NO in cultured medium from DEX group were also lower than those from the control and STAT1 SON group （p 〈 0.05）, but no difference between STAT1 SON group and the control （p 〉 0.05）. The results suggest that STAT1 ASON could inhibit the secretion of TNF-α IL-8, NO in AMs, and STAT1 could become a target of treating pulmonary fibrosis. Cellular ＆ Molecul

Three logarithmic linear equations between D_(Me)^(V/L) and [m_(NaCl)], and the relationship of D_(Me)^(V/L) versus F/Cl or K/Na mole ratios have been established by the experiments of the partitioning of Pb and Zn between granitic silicate melt and aqueous fluid. These results have been used to quantitatively study some essential problems, such as the possibility and degree of Pb-Zn mineralization in the system of granitic magma and hydrothermal fluid, and the influence of the relative contents of alkali and volatiles on the Pb-Zn mineralization in the same system. Some new points have been put forward in this paper.

The kinetics on the growth of *** in the presence of Cu(Ⅱ) was studied using of series piezoelectric quartz crystal (SPQC). Arsenic ion inhibits the growth of *** , which is consistent with the previous results by other measuring methods. This further demonstrates that the SPQC can monitor the chemical activity of *** growth. Cupric ion accelerates the growth of *** . The mechanism was discussed, i. e., copper promotes the protein enzyme of *** , rusticyanin, to form over the range of cupric ion concentration studied. The reaction order of cupric ion in accelerating the bacterial growth is 0.067. The growth of *** is dependent on temperature, the apparent reaction activation energy decreases from 2..56 kJ/mol to 18.32.kJ/mol because of the addition of 10 mg/L Cu(Ⅱ) to the bacterial growth solution of pH 2.0 at initial inoculum of 10%.

In an attempt to isolate and identify the antigenic epitopes on ferritin of Schistosoma japonicum (SjFer) and to test their protective potentiality against Schistosoma japonicum (S.j), polyclonal antisera against SjFer was prepared to screen a 12.mer random peptide library. Three rounds of biopan- ning were performed and resulted in an enrichment. Six peptides selected randomly from the third elute were all found to be positive by evaluating the binding to anti-SjFer sera by ELISA and Western blot- ting. Three amino acid sequences were deduced from the six phage clones by sequencing. When they were used to vaccinate mice, the three peptides could induce significant reduction in adult worms (2..7%, 2..4%, and 2..9%) as well as in liver eggs per gram (LEPG) (40.0%, 38.2., and 40.8%). This result showed that three mimotopes on SjFer were obtained and they could induce significant protective efficacy against S.j.

BACKGROUND:Chronic hepatitis B virus(HBV)infection is a major cause of liver fibrosis,but the mechanisms underlying HBV-related fibrogenesis are still unknown. Although the roles of HBV X protein(HBx)remain poorly understood,it is thought to play an important role in the regulation of cellular growth and hepatocarcinogenesis. The aim of this study was to determine the role of HBx in liver fibrogenesis by studying the effect of HBx on the proliferation and expression of fibrosis-related molecules in the human hepatic stellate cell line,LX-2. METHODS:We established an in vitro co-culture system with LX-2.cells and a stable QSG7701-HBx cell line which had been transfected with the HBx gene. 3 H-TdR incorporation and flow cytometry were used to determine the effects of HBx on the proliferation of LX-2.cells. α-smooth muscle actin(α-SMA),transforming growth factor-β1(TGF-β1),transforming growth factor-βreceptor Ⅱ(TGF-βRⅡ),and connective tissue growth factor (CTGF)in LX-2.cells were analyzed by Western *** addition,the expression levels of collagen typeⅠ(ColⅠ) from the co-cultured media were measured by ELISA. RESULTS: 3 H-TdR incorporation increased significantly in LX-2.cells co-cultured with QSG7701-HBx cells compared to those cultured with QSG7701-pcDNA3 and QSG7701 (non-tumorigenic human liver cell line).Cell cycle results revealed that HBx accelerated the progression of G1 to S in LX-2.*** expressions ofα-SMA,TGF-β1,TGF-βR Ⅱ,CTGF and ColⅠwere significantly increased in the co- cultures of LX-2.cells with stable QSG7701-HBx cells. CONCLUSION:These results suggest that HBx may facilitate liver fibrosis by promoting hepatic stellate cell proliferation and upregulating the expression of fibrosis- related molecules.
