A new Paphiopedilum entity found in southern guangdong was studied taxonomically by means of internal transcribed spacer of nuclear ribosomal DNA analysis in addition to morphological and phytogeographical *** entity is similar to Paphiopedilum gratrixianum(Masters)Rolfe morphologically,from which it differs by having narrower leaves not wider than 2.cm and lacking purple spots toward abaxial base,pale green dorsal sepal veined with pale purplish brown and not heavily spotted with purple,and staminode subtruncate and mucronate at *** analysis of internal transcribed spacer sequences of this entity and its allies came to the same conclusion that the entity is a member of Paphiopedilum section Paphiopedilum and closely related to but distinct from *** a result,it is treated here as a new species,Paphiopedilum guangdongense ***&***.

Simulation of 1991-2.05 Meiyu seasons in the Yangtze-Huaihe region using BCC_RegCM 1.0

By using BCC_RegCM 1.0 (RegCM for short) from Beijing Climate center (BCC), China Meteorological Administration (CMA), the Meiyu season characters over the Yangtze-Huaihe region during 1991-2.05 are simulated. The major conclusions of this study may be summarized as the following: (1) RegCM can reproduce the interannual variation and the spatial distribution of the summertime precipitation and temperature in the Yangtze-Huaihe region. (2. By use of a generalized Meiyu criterion and in accordance with model-calculated precipitation and temperature, the Meiyu onset and ending date have been determined. Compared with the observation, RegCM can simulate the interannual variation of the Yangtze-Huaihe Meiyu with preferable capability for most of the normal Meiyu years (such as 1995, 1997, 2.00, 2.01, 2.02.and 2.04., especially for the rich Meiyu years of 1996, 1998 and 1999. (3) In terms of the average simulation for the recent 15 years, the timings of onset and ending of Meiyu occur on June 1 and July 13, respectively, which are earlier than the climatological observation. For duration, Meiyu persists for 32.d, 3 d shorter than the observation. The index of Meiyu intensity is 2.4., while the climatological one is 3.00. Therefore, RegCM is capable of simulating the climatological Meiyu duration and intensity, while the capability of simulating the onset and ending date of Meiyu still needs to be improved.

Tanshinone prevents cancellous bone loss induced by ovariectomy in rats

AIM: To investigate the skeletal effects of total tanshinone in ovariectomized rats by analyzing cancellous bonehistomorphometry of fourth lumbar vertebrae (LV4. and proximal tibial metaphyses (PTM). METHODS: Four-month-old Sprague-Dawley female rats were sham-operated and treated with vehicle or ovariectomized and treatedwith either vehicle, total tanshinone (2.0 mgkg-1d-1, equivalent to 35 gkg-1d-1 of tanshinone II A and 16 mgkg -1d-1 ofcryptotanshinone), or 17a-ethynylestradiol (30 gkg-1d-1 as positive treatment group) starting one day post-sur-gery for 10 weeks. Double in vivo fluorochrome labeling was administered to all rats. The undecalcified longitu-dinal LV4.and PTM sections were cut and stained with Goldners Trichrome (4.m thickness) or unstained (8-mthickness) for the bone histomorphometric analysis. RESULTS: A significant decrease in trabecular bone volume(BV/TV) and trabecular number (Tb.N) and a significant increase in osteoclast surface (OCS/BS) and mineralizingsurface (MS/BS) were found in both LV and PTM of vehicle-treated OVX rats compared with sham *** completely prevented the decreases in BV/TV and Tb.N and the increase in OCS/BS in the LV4. andpartially prevented the decreases in BV/TV and Tb.N in the PTM of OVX rats. In addition, tanshinone increasedtrabecular thickness (***) whereas it did not alter MS/BS. Moreover, tanshinone had no effect on uterine weightand body weight of OVX rats. Estrogen treatment increased BV/TV and Tb.N and decreased OCS/BS, but, alsomarkedly decreased MS/BS and increased uterine weight in OVX rats. CONCLUSION: The current study demon-strated that the adequate supply of tanshinone prevented OVX-induced cancellous bone loss in rats through inhibi-tion of elevated bone resorption.

Expression and Bioinformatics Analysis of SPACA4.in Human and Mice

Objective To analyze the expression of SPACA4.in human and mice. Methods Testes cRNA samples from Balb/c mice of different postnatal days were performed with mouse affymetrix chip to screen the expression of SPACA4.in mice. Sub-quantitative RT-PCR and bioinformatic tools were used here to describe the expression profile of SPACA4.in mice and human. Results The results of gene chip analysis indicated that the expression of mSPACA4.began after d 35 of postnatal testis in mice. Sub-quantitative RT-PCR assay showed that SPACA4.gene expressed exclusively in mouse and human testis, and mouse mSPACA4.gene expressed after d 35 of postnatal testis that was consistency with the results of gene chip analysis. By bioinformatics analysis, mSPACA4.is located in cell membrane （34.8%） or plasma membrane （34.8%）, the signal peptide cleavage site between position 19 and 2. amino acids, transmembrane region between 2.2. and 101-12. amino acids, respectively, on mSPACA4.protein. Conclusion mSPACA4.and hSPACA4.were testis-specific genes, and the expression of mSPACA4.begins after d 35 of postnatal testis in mice. SPACA4.is a candidate for targeting in a sperm-based contraceptive vaccine.

Dual fluorescence in situ hybridization in detection of HER-2.oncogene amplification in primary hepatocellular carcinoma

BACKGROUND: Molecular cytogenetics of oncogene HER-2.amplification in primary hepatocellular carcinoma (HCC) is still unknown. The aim of this study was to in vestigate the frequency of HER-2.oncogene amplification in primary HCC and its relations to clinicopathological pa rameters and prognosis. METHODS: Forty-two surgical samples from patients with primary HCC were detected for their HER-2.oncogene am plification. The number of chromosome 17 and their ratio were tested by dual fluorescence in situ hybridization (FISH) technique, and then the correlations between HER-2.amplification, clinicopathological characteristics and prog nosis were analyzed statistically. RESULTS: HER-2.oncogene amplification was detected in 9 (2..4.) of the 4. primary HCCs, including 4.patient with high copy (HC) (9.5%) and 5 patients with low copy (LC) (11.9%). HER-2.amplification was associated signifi cantly with tumor size and postoperative survival time o HCC patients (P2.gene amplification was correlated with postoperative relapse (P— 0.2.7), but not related to sex, age, AFP level, HBV infec tion, histopathological grading and clinical staging of HCC patients (P>0.05). The HER-2.oncogene copy was exa mined in 31 (73.8%) of the 4. primary HCCs, consisting of 9 patients with HER-2.amplification (2..4.) and 2. pa tients with aneuploidy (52.4.). No significant relation were observed between the HER-2.oncogene copy, patien sex, tumor size, histopathological grading, clinical stag ing, postoperative relapse and survival time (P >0.05); bu the HER-2.oncogene copy was correlated significantly to age, AFP level and HBV infection (P 2.oncogene amplification and a higher frequency of chromo- some 17 aneuploidy in primary HCC. A new type electronic particle counter (EPC, MultisizerTM 3, Beckman Coulter Inc., USA) was used to determine the volumes of human red blood cells (RBCs) in NaCl solutions of different osmolalities. The thermodynamics model describing cell response during freezing process was used to simulate the volume change of RBC in 0.9% NaCl solution during equilibrium freezing process. It was assumed that the effect of temperature on cell volume can be neglected compared to that of osmolality, then by using the phase diagram for the binary system sodium chloride/water, the osmolalities of the NaCl solution under different sub-zero temperatures can be obtained (converted from mass concentration), then the calculated values of RBC volumes can be validated by the experiments.

Either cetuximab or bevacizumab can improve the survival of patients with metastastic colorectal cancer (mCRC) if administered combided with cytotoxic agents. However, the effect of two or more target agents in combination is uncertain in these patients. Here, we reported a patient with mCRC successfully treated by a combination of target agents after the failure of chemotherapy. The patient received palliative resection of primary tumor followed by 9 cycles of postoperative XELOX regimen, cytokine-induced killer cell (CIK)-based biotherapy, traditional Chinese medicine, particle implantation in the lung metastatic lesions. The tumor progressed 2. months after the standard treatments. Then, the regimen cetuximab, bevacizumab and cefitinib was applied. During the treatment with targeted agents, grade IV acne-like rash and relatively severe parionychia of the toes occurred. Both of them recovered smoothly. The PET-CT reexamination at 4. days after the target treatment showed that the metabolism of mediastinal lymph nodes basically recovered to a normal level. The combination of multiple targeted agents obtained a progression-free survival(PFS) of 11 months and the patient with a good quality of life during this period.

Tannin inhibits HIV-1 entry by targeting gp4.

AIM: To investigate the mechanism by which tannin inhibits HIV-1 entry into target cells. METHODS: The inhibitory activity of tannin on HIV-1 replication and entry was detected by p2.0}production and HIV-1-mediated cell fusion, respectively. The inhibitory activity on the gp4. six-helix bundle formation was determined by an improved sandwich ELISA. RESULTS: Tannins from different sources showed potent inhibitory activity on HIV-1 replication, HIV-1-mediated cell fusion, and the gp4. six-helix bundle formation. CONCLUSION: Tannin inhibits HIV-1 entry into target cells by interfering with the gp4. six-helix bundle formation, thus blocking HIV-1 fusion with the target cell.
