Objective To investigate the effects of Panax notoginseng saponins(PNS)on hydrogen peroxide(H2O2)-induced apoptosis in cultured rabbit bone marrow stromal cells(BMSCs).Methods BMSCs from 3-month-old New Zealand rabbits were isolated and cultured by the density gradient centrifugation combined with adherent *** cultured BMSCs were divided into three groups:normal control,H2O2 treatment(100μmol/L),and PNS pretreatment(0.1g/L).Intracellular reactive oxygen species(ROS)levels as the index of oxidative stress were measured by using 2’7’-dichlorodihydrofluorescein *** cytometry was used to observe the apoptosis of BMSCs by staining with annexinV-FITC/*** protein expression of Bax in BMSCs was analyzed by Western *** of caspase-3 enzyme was measured by *** Pretreatment with PNS significantly decreased intracellular ROS level induced by H2O2(P38.68% to 19.24%(P3 enzyme induced by H2O2(P3 activity.

Lead-free Bi_(_(0.5))Na_(_(0.5))TiO_(3)(BNT)piezoelectric ceramics have the advantages of large coercive fields and high Curie *** the improvement of piezoelectric coefficient(d 33)is usually accompanied by a huge sacrifice of depolarization temperature(T d).In this work,a well-balanced performance of d 33 and T d is achieved in MnO_(2)-doped 0.79(Bi_(_(0.5))Na_(_(0.5))TiO_(3))-0.14(Bi_(0.5)K_(0.5)TiO_(3))-0.07BaTiO_(3)ternary *** in-corporation of 0.25 mol%MnO_(2)enhances the d 33 by more than 40%,while T d remains almost unchanged(i.e.,d 33=181 pC/N,T d=184℃).X-ray diffraction(XRD)shows that an appropriate fraction of the small axis-ratio ferroelectric phase(pseudo-cubic,P c)coexists with the long-range ferroelectric phase(tetrag-onal,T)under this MnO_(2)*** force microscopy(PFM)has revealed a special domain configuration,namely large striped and layered macro domains embedded with small *** study provides a distinctive avenue to design BNT-based piezoelectric ceramics with high piezoelectric performance and temperature stability.

Aim: To investigate the effects of bone morphogenetic protein-7 (BMP7)-expressing recombinant adeno-associated virus (AAV) vector on the differentiation of C2C12 cells. Methods: AAV-BMP7 was packaged by infecting the stable cell clone BHK-21 (integrated with recombinant AAV vector plasmid pSNAV-BMP7) with recombinant herpes simplex virus type 1, which expresses AAV-2 Rep and Cap and possesses AAV packaging functions. Following infection with AAV-BMP7 at multiplicities of infection of lx 10～5 vector genomes per cell and subse-quent culture, C2C12 cells were assessed qualitatively for BMP7 production, alka-line phosphatase activity, osteocalcin production and Cbfal and MyoD expression. Results: C2C12 cells transduced with AAV-BMP7 could produce BMP7 protein until d 28. Alkaline phosphatase in the cultured C2C12 cell lysate was elevated. Secreted osteocalcin in the culture medium was detectable at d 12 and Cbfal mRNA expression level was upregulated, coinciding with downregulation of MyoD in a temporal manner. Conclusion: The present in vitro study demonstrated that AAV-BMP7 could infect and efficiently convert C2C12 cells from myoblasts into osteoblast lineage cells.

1 IntroductionRepair of tissues like bone, cartilage, muscle,etc., is a tough problem in clinical treatment. The recent research show that there are plenty of mesenchymal stem cells (MSCs) in myeloid tissue besides hemopoietic stem cells(HSCs). Just as the pluripotential hemopoietic stem cell can give bone marrow tissue excellent hemopoietic ability and maintain the metabolism of, MSCs can give potential repair ability to bone, cartilage tissue injury[1] . But compared with the HSCs, the content of MSCs in myeloid tissue are not abundant. We separated and purified the MSCs from myeloid tissue through in vitro cell culture,and studied the proliferation and growth characteristics under in vitro culture conditions, providing experimental foundations for further research on repair epiphyseal plate cartilage defect through MSCs tissue engineering.

BACKGROUND: Myasthenia gravis (MG) is considered as a peripheral neuromuscular disorder. Some investigations demonstrate that MG affects central nervous system (CNS), and there are disorders in cognitive function, affection, sleep, mental status and others; however, there are also some different standpoints. Some questions are still disputed, such as whether or not MG exists in the CNS and cognitive function is impaired. DATA SOURCES: Using the terms of 'myasthenia gravis, neuropsychological test, memory, central, brain, cognitive dysfunction and so on', we searched the Medline database for the articles about CNS involved in MG, nerve and neuropsychology as well as cognitive function, which were published in the English language between January 1985 and December 2004. Meanwhile, we also searched CNKI database by inputting 'myasthenia gravis, neuropsychology and cognitive function' in Chinese for articles about involved neuropsychology and cognitive function which were published between October 1995 and June 2006 in the Chinese language. STUDY SELECTION: Data were checked in the first trial, and articles about the influences of MG on CNS, neuropsychology, cognitive function and central cholinergic function were selected, then those were obviously unrelated with above criteria were excluded. Articles which expound the influences of MG on neuropsychology and cognitive were included, but those with repeated contents were also excluded. DATA EXTRACTION: A total of 89 related English articles and 34 Chinese articles were collected, including 43 studies on involved CNS and 32 studies on involved central cholinergic function and cholinergic receptor. Sixty-three literatures were about neuropsychology and cognitive function, which were closely related with this article. Seventy-two articles met inclusive criteria and 66 were excluded. Aim:To examine the chondrogenic potential of human adipose derived stem cells(hASC) induced by human transforming growth factor beta2 (hTGF beta2) invitro,and to investigate if predifferentiated hASC can produce neocartilage ***:hASC were isolated from subcutaneous adipose tissue and cul-tured in pellets with the addition of hTGF *** differentiationwas assayed by RT-PCR,Western blotting,toluidine blue staining,and immuno-histochemistry staining for collagen type Ⅱ.For the in vivo study,intact inducedcell pellets or the released cells embedded in alginate gel with different concentra-tions were implanted subcutaneously in nude *** were harvested atdifferent time points and carried with histological and immunohistochemistry ex-amination to evaluate the cartilage ***:RT-PCR analysis revealedthat hASC produced aggrecan and collagen type Ⅱ after 7 d of induction andcontinued throughout the culture *** was also demonstrated by theWestern blot analysis,positive staining of toluidine blue,and immunohistochem-istry for collagen type Ⅱ.After reseeding in the monolayer,the cells isolated fromthe pellets displayed a polygonal morphology compared with the primary spindleshape,hASC were released from the induced cell pellets when embedded inalginate gel (implanted cell concentration=5×10～6/mL or higher).They producedneocartilage after 12 weeks in vivo culture;however,intact induced cell pelletsimplanted subcutaneously rapidly lost their differentiated ***:Chondrogenesis of hASC in vitro can be induced by combining pellet culture andhTGF beta2 *** hASC embedded in alginate gel have theability of producing neocartilage in vivo.

In order to find new drugs to inhibit nitric oxide （NO） production, the effects of pyrrolidine dithiocarbamate （PDTC）, a nuclear factor-kappa B （NF-κB） inhibitor, on recombinant human interleukin-1β （rhIL-1β）-induced NO production in chondrocytes were investigated. Rat chondrocytes were isolated and cultured, divided into control, P0, P1, P2, P3 and P4 groups. The chondrocytes in the P0, P1, P2, P3 and P4 groups were treated with different concentrations of PDTC （0, 3, 10, 30, and 50 ***/L respectively） for 1 h and then incubated with 5 U/mL rhIL-1β for 24 h. NO assay kit and RT-PCR were used to detect the NO content and the iNOS mRNA expression in the chondrocytes The expression level of iNOS mRNA in control, P0, P1, P2, P3 and P4 groups was 0.02±0.01, 1.24±0.13, 1.21±0.14, 0.61±0.11, 0.40±0.09, 0.21±0.06, and the relative content of NO was 15.8±2.7, 100±14.8, 92.6±9.3, 68.3±14.2, 27.5±9.8, 19.8±3.6, respectively. In the P0, P1, P2, P3 and P4 groups, the expression of iNOS mRNA and NO production were significantly increased as compared with those in the control group. As compared with the P0 group, the expression of iNOS mRNA and NO content in control group were lower. In the P2, P3 and P4 groups, PDTC could significantly inhibit the expression of iNOS and NO production induced by rhIL-1β in a concentration-dependent manner. It is suggested that PDTC can inhibit NO production and iNOS mRNA expression induced by IL-1β, which may provide an alternative method for the treatment of osteoarthritis.

Quantitative in vitro assessment of drug phototoxicity by a chemiluminescence method

Objective To establish a test model for phototoxic agents using the method of chemiluminescence Methods The phototoxicity of pipemidic acid, doxycycline, griseofuvin and chlorpromazine was detected These agents and distilled water were irradiated with ultraviolet A (UVA) in the presence of nicotinamide adenine dinucleotide reduced (NADH), and the duplicated samples were incubated in the dark as dark controls Then luminol was added to the test samples, and the chemiluminescent value was counted and calculated Results Chemiluminescent values of pipemidic acid, doxycycline and griseofuvin were significantly higher than those in controls The result of linear regression analysis showed that phototoxic intensity was linear correlated with UVA dosage The regression coefficient for distilled water was 0 56, indicating that the luminscent value (LV) rose slightly after UVA irradiation For pipemidic acid, griseofuvin and doxycycline, the regression coefficients reached 76 96 , 254 33 and 92 61 respectively, significantly increased in comparison with those of negative controls (P<0 01) Conclusion Phototoxicity of pipemidic acid, doxycycline and griseofuvin can be detected with the method of chemiluminescence
