Antifertility Effects of Orally Administration of Low Dose Gossypol Acetic Acid Combined with Methyltestosterone Plus Ethinyl Estradiol on Male Rat

Objective To investigate the feasibility and optimal regimen of orally administration of low dose gossypol acetic acid （GA） combined with methyltestosterone （MT） plus ethinyl estradiol （EE） for contraception in males.Methods Wistar male rats were randomly assigned into four groups, 20 in each group. Animals in group A or B were administered daily with 1% methyl cellulose or GA （12 mg/kg） suspended in 1% methyl cellulose, respectively. Rats in group C or D took firstly GA 12 mg/kg＋MT 20 mg/kg＋EE 0.1 mg/kg or MT 20 mg/kg＋EE 0.1 mg/kg, in asuspension with 1% methyl cellulose, via gastric intubation. After the infertilities were initiated（6 weeks for group C, 8 weeks for group D）, GA was served alone while MT＋EE were withdrawn in rats of groups C and D. The treatment was ceased after 18 weeks and some males from group C were permitted to recover. Fertility testing, 10 males per group, was served for determining infertility or restoration of fertility in treated rats. Examinations of histology and biochemistry in treated rats were used to examine the morphologic influences on sperm, testis, epididymides and viscera, and biochemical changes in blood. The growth and development of F1 generation of the rats would also be tested in a series of behavioral tests.Results Ten rats from group C were infertile at week 6 after treatment, and the fulfilled infertility was maintained with low-dose GA （12 mg/kg） only daily. Six weeks after cessation of treatment, all of treated males recovered their fertility. However, 8 of 10 rats from group D were in sterility at 6th week of treatment and all at 8th week of treatment, but the infertility could not be kept with the similar dose GA alone later on. Moreover, no adverse effects were found in our present experiments.Conclusion Administration of oral low dose GA combined with MT and EE as loading dose could successfully induce infertility in short term, whereafter the efficacy could completely be maintained by similar low dose of

Although there are plenty of evidence that single-nucleotide polymorphisms(SNPs) that fall within coding sequences of genes are involved in recurrent pregnancy loss(RPL),it is still unknown whether the polymorphisms in microRNAs(miRNAs) are related with *** uiis study,we established this kind of association by confirming significant differences in genotype distribution of rs41275794(P= 0.0005) and rs12976445(P= 0.001) within the pri-miR-125a in 217 Han Chinese patients of RPL compared with 43. *** on this observation,two-locus haplotypes were constructed and the A-T haplotype was found to be associated with an increased risk of RPL(OR=2.84,95%C.I.1.98-4.07,/M).0000000057). Further analysis showed that the levels of pre- and mature- miR-125a were down-regulated in the cells transfected with the A-T haplotype,which was consistent with in vivo detection that the level of mature miR-125a was lower in 3. pregnant women with A-T haplotype than that with G-C *** in vitro RNA processing assays,we found a similar decrease in the amount of pre-miR-125a and decline in binding capacity of nuclear factors to pri-miR-125a with A-T *** importantly,the reduction in miR-125a,as a consequence of A-T haplotype,further led to less efficient inhibition of target genes,LIFR and ERBB2,which play important roles in the embryo implantation and ***,our data collectively suggest that two common polymorphisms in pre-miR-125a might contribute to the genetic predisposition to RPL by disrupting the production of miR-125a,which consequently interfered in the expression and function of target genes of miR-125a.

The anthers of Bupleurum chinense were used for the induction of androgenesis and plant *** formation was best achieved when the anthers were cultured on BMCI supplemented with 0.1 mg/1 BA at 26°C for 9-12 weeks after the umbels were cold pretreatment at 6°C for 4 *** regeneration from anther-derived embryos was induced on 1/2-strength MS differentiation medium with 3.sucrose supplemented with 0.2 mg/1 BA for about 12 weeks. Plant regeneration was observed in both pathways,which was affected by BA concentration and culture temperature. Chromosome counting of plantlets regenerated showed that they were haploid plant(n=6).By using this protocol,complete plantlets could be regenerated through indirect somatic embryogenesis or organogenesis from the anther culture in 21-23.weeks.
