A new sesquiterpenoid compound named ligustilone was isolated from the roots of Ligusticum sinense Oliv. Based on the chemical and spectral studies especially 2DNMR including H-1, H-1 COSY, H-1, C-13 COSY and COLOC techniques, its structure has been established as a bisabolane derivative and showed as 1. A stereoselective and economic synthesis of the carbapenem sidechain (2S, 4S)-4-ace-tylsulphanyl-2-[(S)1-phenylethyl-carbamoyl] pyrrolidine-1-carboxylic acid 4-nitrobenzylester was developed. Due to the effect of spatial hindrance, only the (2S,4S) diastereomer 3 wasobtained by coupling 1 and the inexpensive racemic 2 catalyzed by EEDQ.

The tricyclic compound 1, a key intermediate of the pentacyclic core of Et 743 and its analogues, was synthesized. Its 11-epimer was found as the product of racemization when BOP was used as the coupling agent, but not when BOP-C1 used in the same reaction. The stereochemistry of both isomers was confirmed on basis of the 1H NMR and NOE-difference spectra.
