The Etiology and Outcome Analysis of Neonatal Burst Suppression EEG

The neonatal burst suppression is a severe EEG pattern and always demonstrates serious damage of nerve system. But the outcome of these patients depends on the different etiology. A total of 256 cases of video EEG recordings were analyzed in order to summarize the etiology and outcome of burst suppression. The results showed that some patients in all 17 cases of burst suppression showed EEG improvement. The etiology was the dominant factor in long term outcome. It was suggested that effective video EEG monitoring is helpful for etiologic study and prognosis evaluation.

The X-ray radiograph, CT scan and MRI appearance of 5 patients with pathologically proven fibrous dysplasia in thoracic and lumbar spine vertebrae were retrospectively analyzed. Plain radiographs, CT scans and MR images showed the presentation of eccentric lesion with intact cortex bone and marginal sclerosis in vertebral bodies without involvement of vertebral appendix and ex- traosseous soft tissue. The lesion masses were round （one being oval-shaped） and radiolucent in plain radiographs and CT scans. Homogeneous long signal was observed on Tl weighted image and strongly enhanced when gadolinium was administered. On T2 weighted MRI, short signal was found in the anterior part of the mass, long signal in the posterior part, and short and slight long signal in the middle part, without partitioning and laminating change. There was a good correlation between radiological features and surgical findings. These findings may be useful to diagnose fibrous dysplasia m spree.

The expression of angiopoietin- 1 （Ang- 1） and thrombospondin- 1 （TSP- 1） in 5/6 subtotal nephrectomy （STN） rats model, and its correlation to the renal microvasculature injury were investigated. Rat 5/6 STN model was established in adult male SD rats, and the sham-operated group and 5/6 STN group were set up. The renal function and histopathological changes were examined at the 1st, 2nd, 4th, 8th and 12th week after operation. The expression orAng-1, TSP-1 and CD31 in renal tissues was detected by using immunohistochemistry. From 2nd to 8th week after operation, Ang-1 was significantly expressed in glomeruli of rats with STN. Ang-1 staining in glomeruli of STN group was increased significantly as compared with that in sham-operated group at 4th and 8th week after operation, and subsequently decreased after the 12th week. The expression of TSP-1 was increased significantly in STN group. As compared with sham-operated group, the CD31 expression was significantly down-regulated from the 2nd week. The expression of Ang-1 mRNA was detected by using RT-PCR at the same time points. The expression of Ang-1 mRNA in renal tissue of rats with STN was significantly up-regulated at the 2nd, 4th and 8th week after operation as compared with that in STN group at other time points or in sham-operated group at the same time points, while decreased evidently at the 12th week as compared with that in sham-operated group. It is concluded that there are changes in the mRNA expression of Ang-1, and the significant up-regulation of the expression of TSP-1 in renal tissue of rats with STN, which may be involved in the remnant renal microvasculature injury.

In order to provide a new method for treating proliferative vitreoretinopathy , the effects of anti proliferation and apoptosis induction of inhibitors of telomerase and heat shock protein 90 on the cultured retinal pigment epithelial cells were investigated. The rate of apoptosis cells was measured by using TUNEL on the cultured RPE cells, the co cultured RPE cells with inhibitor of telomerase or the co cultured RPE cells with inhibitor of Hsp90 . The cell proliferation status was measured in the above three groups by using MTT method. The rate of apoptosis in the RPE cells co cultured with camptothecin or geldanamycin was increased remarkably . MTT showed the rate of growth inhibition was 8.4 %, 32.3 % and 72.3 % at the concentrations of camptothecin 1 μmol/L, 5 μmol/L, 10 μmol/L, respectively, and 6.5 %, 30.9 %, 71.9 % at the concentrations of geldanamycin 1 μmol/L, 5 μmol/L, 10 μmol/L, respectively. It was concluded that telomerase and Hsp90 can promote the proliferation of the cultured RPE cells, while the inhibitor of them can induce apoptosis and inhibit the growth of the RPE cells.

By using decoy-oligodeoxynucleotides （decoy-ODNS） technique, the effects of Stathmin gene on the proliferation and differentiation of in vitro cultured precartilainous stem cells （PSCs） were investigated. The Stathmin decoy-ODNs were transfected into PSCs in rats by using gene transfection technique. Under the induction of cortisol （1 μmol/L）, electrophoretic mobility shift assay was used the inhibitory effects of decoy-ODNS on Stathmin gene. MTT and cytometry were used to test the cell proliferation. The expression of collagen Ⅱ and Ⅴ and Stathmin protein was detected by using Western blot. The results showed that Stathmin decoy-ODNs inhibited the Stathmin activity in a dose-dependent manner. When the concentration of decoy-ODNs was 10 times of standard con- centration, the proliferation of PSCs was obviously suppressed and the differentiation happened. Compared to the control group, the difference was significant （P〈0.05）. It was concluded that decoy-ODNs could inhibit the proliferation and promote the differentiation of PSCs by antagonizing Stathmin activity.

Dracorhodin Perchlorate Suppresses Proliferation and Induces Apoptosis in Human Prostate Cancer Cell Line PC-3

The growth inhibition and pro-apoptosis effects of dracorhodin perchlorate on human prostate cancer PC-3 cell line were examined. After administration of 10-80 μmol/L dracorhodin perchlorate for 12-48 h, cell viability of PC-3 cells was measured by MTT colorimetry. Cell proliferation ability was detected by colony formation assay. Cellular apoptosis was inspected by acridine orange-ethidium bromide fluorescent staining, Hoechst 33258 fluorescent staining, and flow cytometry （FCM） with annexin Ⅴ-FITC/propidium iodide dual staining. The results showed that dracorhodin perchlorate inhibited the growth of PC-3 in a dose- and time-dependent manner. IC50 of dracorhodin perchlorate on PC-3 cells at 24 h was 40.18 μmol/L. Cell clone formation rate was decreased by 86% after treatment with 20 μmol/L of dracorhodin perchlorate. Some cells presented the characteristic apoptotic changes. The cellular apoptotic rates induced by 10-40 μmol/L dracorhodin perchlorate for 24 h were 8.43% to 47.71% respectively. It was concluded that dracorhodin perchlorate significantly inhibited the growth of PC-3 cells by suppressing proliferation and inducing apoptosis of the cells.

Aim:The objectives of this study were to investigate the inhibitory action ofverapamil on wild-type（WT） and mutation HERG K+channel current (IHERG),andto determine whether mutations in the $6 region are important for the inhibition ofIHERGby ***:HERG channels （WT,Y652A,and F656A） wereexpressed in oocytes of Xenopus laevis and studied using the 2-electrode volt-age-clamp ***:WT HERG is blocked in a concentration-depen-dent manner by verapamil (half-maximal inhibition concentration [IC50]=5.1 μmol/L),and the steady state activation and inactivation parameters are shifted to morenegative ***,mutation to Ala of Y652 and F656 located on the S6domain produced 16-fold and 20-fold increases in IC50for IHERGblockade,***,the steady state activation and inactivation para-meters for Y652A are also shifted to more negative values in the presence of ***:Verapamil preferentially binds to and blocks open ***-652 and Phe-656,2 aromatic amino-acid residues in the inner （S6）helix,are critical in the verapamil-binding site.

BACKGROUND: It has been demonstrated that adenosine can induce glial cell to release cytochrome C, enhance expression of apoptotic gene bax, inhibit anti-apoptotic gene bcl-2, and activate caspase-3 to apoptosis; Whereas inosine can inhibit neuronal apoptosis which is similar to bcl-2. OBJECTIVE: To observe the effects of inosine on neuronal apoptosis and expression of cytochrome C mRNA in rats after focal cerebral ischemia/reperfusion, and analyze the pathway of its neuroprotective effect. DESIGN: A randomised controlled animal trial. SETTINGS: department of Neurology, Rongcheng Second People's Hospital; department of Neurology, Affiliated Union Hospital, tongji medical college, huazhong university of science and technology. MATERIALS: Sixty-eight rats, weighing 230-280 g and clean grade, were used. TdT-mediated dUTP-biotin nick end labeling (TUNEL) and cytochrome C mRNA in situ hybridization kits and DAB staining kit were purchased from wuhan Boster Biological Co., Ltd.; Inosine injection [200 mg (2 mL) each] from Qingdao First Pharmaceutical Factory. METHODS: The experiment was accomplished in the animal experimental center in tongji medical college of huazhong university of science and technology from December 2003 to June 2005. ① Sixty-four rats were made into focal ischemia by middle cerebral artery occlusion (MCAO) with a nylon monofilament suture. The successfully induced rats were assigned to inosine group (n =32) and model group (n =32) at random. Rats in the inosine group were intraperitoneally administrated with inosine in dose of 100 mg/kg preoperatively, twice a day, 7 days in all. The rats in the control group were injected with the same dose of saline solution by the similar way preoperatively. Each group was randomized into ischemia /reperfusion 2, 6, 12, 24 hours, 2, 3, 7 and 14 days subgroups consisted of 4 rats. Aim:To investigate anticancer effects and molecular mechanism of deguelin onhuman Burkitt’s lymphoma Daudi cells in vitro and compare the cytotoxicities ofdeguelin on Daudi cells and human peripheral blood monocular cells(PBMC).Methods:The effects of deguelin on the growth of Daudi cells were studied by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium(MTT)*** dectected through Hoechst 33258 staining and Annexin V/PI *** effect of deguelin on the cell cycle of Daudi cells were studied bya propidium iodide *** expressions of cyclin D 1 and pRb were checkedby Western ***:The proliferation of Daudi cells were decreased indeguelin-treated group with a 24-h IC50value of 51.55 nmol/*** inducedDaudi cells apoptosis was in a time-and dose-dependent manner.G0/G1phaseincreased and S phase decreased in Daudi cells treated with *** deguelin0,5,10,20,and 40 nmol/L treatment for 24 h,G0/G1phase increased from 37.34% to56.56%,whereas S phase decreased from 37.72% to 21.36%.PBMC was lesssensitive to the cytotoxic effect of deguelin than Daudi *** expression ofcyclin D 1 and pRb protein were decreased sharply in Daudi cells treated ***:Deguelin is able to inhibit the proliferation of Daudi cellsby regulating the cell cycle that arrested cells at G0/G1phase and inducing the ***,deguelin selectively induced apoptosis of Daudi cells withlow toxicity in *** antitumor effects of deguelin were related to down-regulating the expression of cyclin D1 and pRb protein.
