Antibiotic Resistance of Probiotic Strains of Lactic Acid Bacteria Isolated from Marketed Foods and Drugs

Objective To identify the antimicrobial resistance of commercial lactic acid bacteria present in microbial foods and drug additives by analyzing their isolated strains used for fermentation and probiotics. Methods Antimicrobial susceptibility of 41 screened isolates was tested with disc diffusion and E-test methods after species-level identification. Resistant strains were selected and examined for the presence of resistance genes by PCR. Results Distribution of resistance was found in different species. All isolates were susceptible to chloramphenicol, tetracycline, ampicillin, amoxicillin/clavulanic acid, cephalothin, and imipenem. In addition, isolates resistant to vancomycin, rifampicin, streptomycin, bacitracin, and erythromycin were detected, although the incidence of resistance to these antibiotics was relatively low. In contrast, most strains were resistant to ciprofloxacin, amikacin, trimethoprim/sulphamethoxazole, and gentamycin. The genes msrC, vanX, and dfrA were detected in strains of Enterococcus faecium, Lactobacillus plantarum, Streptococcus thermophilus, and Lactococcus lactis. Conclusion Antibiotic resistance is present in different species of probiotic strains, which poses a threat to food safety. Evaluation of the safety of lactic acid bacteria for human consumption should be guided by established criteria, guidelines and regulations.

Despite widespread vaccine coverage, the incidence of whooping cough (pertussis) , has again risen recently. The causative agents, Bordetella brochiseptica, Bordetella parapertussis and Bordetella pertussis, are closely related gram-negative pathogen. B. pertussis is exclusively human-adapted and is the primary etiologic agent of acute childhood respiratory whooping cough;B. parapertussis has been only associated with similar respiratory infection in human and sheep;B. brochiseptica infects a wide range of mammalian host, including rat, rabbit, dog and human, and typically causes chronic, asymptomatic infections.

蛋白质组学是在基因组学基础上发展起来的新兴学科,其基本技术包括样品制备、蛋白质分离和蛋白质鉴定分析,其中的核心技术是双向凝胶电泳技术(2-Dimensional Electrophoresis,2-DE)和质谱技术(Mass Spectrometry,MS)。近年来,蛋白质组学技术已应用于结核分枝杆菌的研究领域。应用蛋白质组学技术分离、鉴定、检测结核分枝杆菌致病株的全菌蛋白及分泌蛋白,分析其蛋白组成,可深入解析结核分枝杆菌的致病机理和耐药机制。通过对结核分枝杆菌致病株抗原的分析,为研制预防结核病的新型疫苗拓展了空间。通过对结核分枝杆菌临床分离株的蛋白组成分析还发现了一些有意义的结核病早期诊断标志物。蛋白质组学技术还应用于寻找新的药物靶标,在研制和筛选新的抗结核药物等方面展示了一些有价值的研究成果,为更好地开展结核病的预防、早期诊断及治疗打下了基础。

Background Currently available vaccines against leptospirosis are low efficacy, have an unacceptable side-effect profile, do not induce long-term protection, in addition, provide no cross-protection against the different serovars of pathogenic leptospira. There is a need to screening vaccine candidate genes encoding surface-exposed proteins in the genome of Leptospira interrogans. The reverse vaccinology, which has been successful in the identification of vaccines candidates, can be used to identifying the potential vaccine candidates against leptospirosis. Although several papers tried to identify the surface-exposed proteins of leptospire, but vaccine candidate genes encoding the surface-exposed proteins that can not only provide cross-protection against the different serovars of pathogenic leptospires but also have relatively high transcription level are still awaited. Results Bioinformatics, comparative genome analysis as well as transcriptional analysis were used for the identification of novel vaccine candidates in the genome of Leptospira interrogans strain #56601. There were 686 genes (14.5%, 686/4727) were predicted encoding surface-exposed proteins by P-CLASSIFIER, currently the most accurate predicting system for subcellular localization in Gram-negative bacteria analysis combined with signal peptides prediction, a-helix transmembrane topology prediction, integral β-barrel outer membrane protein and putative lipoproteins prediction. The DNA microarray of L. interrogans strain #56601 has been constructed for comparative genome hybridization (CGH) and transcriptome analysis at 37癈 in vitro. 307 differential genes were identified in ten pathogenic serogroups by CGH. The numbers of high transcriptional level (cy3 signal ≥342 and cy5 signal ≥363.5 respectively) are 1427 genes. Leptospira interrogans is an important mammalian pathogen. Transmission from an environmental source requires adaptations to a range of new environmental conditions in the organs and tissues of the infected host Since many pathogenic ’ bacteria utilize temperature to discern their environment and regulate the synthesis of appropriate proteins, we investigated the effects of temperature on protein synthesis in L. interrogans. We used the L. interrogans serovar lai Strain 56601 genome to construct the microarray. DNA microarray analysis was used to investigate the influence of temperature on global gene expression in strain 56601 grown to mid-exponential phase at 28癈 and 37癈. A total of 112 genes expression varied significantly in expression at 37癈 relative to 28癈. The differentially expressed genes included ten kinds of genes which included membrane proteins, hemolysins encoded genes, heat shock proteins, lipoproteins, two-component system, chemotaxis and flagellar proteins, transcriptional regulators, transport and binding protein, metabolic related protein and proteins with no known homologue. Real-time reverse transcription-PCR assays confirmed the microarray data. The data delineate the spectrum of temperature-regulated gene expression in an important human pathogen and provide many unforeseen results of pathogenesis investigation.

The complete genome sequences of Leptospira interrogans, the agent of leptospirosis, have been obtained. Many sequenced bacterial genomes harbored phage-like elements or cryptic prophages. No phage-like element which has been implicated in pathogenesis, serotype conversion and phage immunity had been studied in L interrogans. BLAST searched using known phage-associated sequences with L interrogans whole genome sequence and phage-like fragment was found around LA0183-LA0219 which included integrate-like gene, repressor-like gene and other functional phage genes. Operon analysis showed that this phage-like fragment was composed of four transcripts. The function of repressor-like gene (LA0195) was tested using EMSA based on promoter sequence of promoter LA0195 and promoter LA0194 which overlapped in intergenic sequence of LA0194 and LA0195. The results showed that LA0195 protein could bind to the promoter sequence. It is not known whether this fragment still function to its host or only .is a record of the prophage relics. There is currently a lively discussion about the relative contribution of vertical and horizontal elements of evolution in bacteria. Further study of this fragment can shed light on the evolution of both prophages of *** and its hosts.

RNA m^6A modification and its function in diseases

N^6-methyladenosine （m^6A） is the most common post-transcriptional RNA modification throughout the transcriptome, affecting fundamental aspects of RNA metabolism, m^6A modification could be installed by m^6A ＂writers＂ composed of core catalytic components （METTL3/METTL14/WTAP） and newly defined regulators and removed by m^6A ＂erasers＂（FTO and ALKBH5）. The function of m^6A is executed by m^6A ＂readers＂ that bind to m^6A directly （YTH domain-containing proteins, eIF3 and IGF2BPs） or indirectly （HNRNPA2B1）. In the past few years, advances in m^6A modulators （＂writers,＂＂erasers,＂ and ＂readers＂） have remarkably renewed our understanding of the function and regulation of m^6A in different cells under normal or disease conditions. However, the mechanism and the regulatory network of m^6A are still largely unknown. Moreover, investigations of the m^6A physiological roles in human diseases are limited. In this review, we summarize the recent advances in m^6A research and highlight the functional relevance and importance of m^6A modification in in vitro cell lines, in physiological contexts, and in cancers.
