The well preserved eclogitic rocks of the Tso Morari dome in eastern Ladakh, northwest Himalaya, provide information relevant to the exhumation of high pressure/low temperature rocks, and the early stage of the Himalayan orogeny. The Tso Morari unit outcrops south of the Indus suture zone (Fig.1). The eclogitic dome is underlined on its eastern part by the Zildat normal fault where serpentinite lenses and partially hydrated peridotites are abundant. The close association of the high pressure rocks and serpentinites suggests a possible role of serpentinites in the exhumation of ultrahigh\|pressure rocks. To evaluate this possibility, geochemical analyses were carried out on the serpentinites closely associated with the Tso Morari eclogites.

Structures using constructors are of ordinary use in functional programming to represent data structures of unbound size. Lack of associativity of constructors, however, hinders program analyses or efficient executions. This paper describes ideas of abstraction toward constructors, and similarly abstraction from constructing functions, which we call functional constructors. We demonstrate our ideas making program analyses easier and enable transformation to efficient execution.

A new method for predicting the gene acceptor site based on multi-objective optimization isintroduced in this *** models for the acceptor,branch and distance between acceptor site andbranch site were constructed according to the characteristics of the sequences from the exon-intron databaseand using common biological *** acceptor function,branch function and distance functionwere defined respectively,and the multi-objective optimization model was constructed to recognize thesplice *** test results show that the algorithm used in this study performs better than the SplicePredictor,which is one of the leading acceptor site detectors.

AIM: To establish a high pressure liquid chromatography (HPLC) method for determination of the concentration of gemcitabine (dFdC) and its metabolite(dFdU) in human plasma. METHODS: Plasma 1.0 mL spikedwith floxuridine as an internal standard was extracted with 3.0 mL of methanol-acetonitrile (v/v, 1:9). The supernatant was evapo- rated at 60 oC and the residue was reconstituted with 0.5 mL of the solution used as the mobile phase. After centrifugation, 50 μL of the supernatant was injected into the HPLC system. Separation was achieved on a C18 (4.6 mm×250 mm, 5 μm) column at 25 oC with the flow rate of the mobile phase set to 0.8 mL/min. The compounds were detected at 268 nm. The mobile phase consisted of 40.0 mmol/L acetate ammonium buffer solution (pH 5.5) and acetonitrile (v/v, 97.5:2.5). RESULTS: The linear range was 0.20-10.0 mg/L(r=0.9999) for dFdC and 0.50-50.0 mg/L(r=0.9999) for dFdU. The limit of detection (LOD) was 0.10 mg/L for dFdC and 0.25 mg/L for dFdU, while the limit of quantification (LOQ) was 0.20 mg/L (RSD3 %) for dFdU. The average recovery of dFdC and dFdU by this method were 103.3 % and 98.7 %, respectively. For intra-day and inter-day, the corresponding standard deviations of the measurements of dFdC and dFdU were both less than 5.5 %. CONCLUSION: An analytical method was established to measure the concentrations of dFdC and dFdU in human plasma and was effectively applied to the dFdC and dFdU pharmacokinetic studies of 8 Chinese patients with malignant tumors.

The adhesion of leukocytes to vascular surface is an important biomedical problem and has drawn extensive attention. In this study, we propose a compound drop model to simulate a leukocyte with a nucleus adhering to the surface of blood vessel under steady shear flow. A two-dimensional computational fluid dynamics (CFD) is conducted to determine the local distri- bution of pressure on the surface of the adherent model cell. By introducing the parameter of deformation index (DI), we investigate the deformation of the leukocyte and its nucleus under controlled conditions. Our numerical results show that: (i) the leukocyte is capable of deformation under external exposed flow field. The deformation index increases with initial contact angle and Reynolds number of external exposed flow. (ii) The nucleus deforms with the cell, and the de- formation index of the leukocyte is greater than that of the nucleus. The leukocyte is more de- formable while the nucleus is more capable of resisting external shear flow. (iii) The leukocyte and the nucleus are not able to deform infinitely with the increase of Reynolds number because the deformation index reaches a maximum. (iv) Pressure distribution confirms that there exists a region downstream of the cell, which produces high pressure to retard continuous deformation and provide a positive lift force on the cell. Meanwhile, we have measured the deformation of human leukocytes exposed to shear flow by using a flow chamber system. We found that the numerical results are well consistent with those of experiment. We conclude that the nucleus with high viscosity plays a particular role in leukocyte deformation.

INDIA/ASIA CONVERGENCE AND HIMALAYAN SHORTENING

Different attempts have been done to deduce the shortening of the Himalayan belt during the India\|Asia convergence. Dewey et al. (1989) and Le Pichon et al.(1992) calculated an India\|Asia shortening of 2300～2150km and 2800～3000km in the western and eastern syntaxes, respectively, since the late 45Ma. According to seafloor\|spreading reconstruction, a total shortening of 3000～500km was estimated after the initial contact of the two plates at 55～50Ma (Molnar and Tapponier, 1975 ; Molnar et al., 1988 ; Replumaz, 1999). Since 40Ma, the part of shortening only accommodated by the Himalayan belt was estimated around 470km in the western part (Coward and Butler, 1985) and 550 to 630km to the east (Ratsbacher et al., 1994 ; Replumaz, 1999). In contrast, global plate reconstructions suggest that the shortening in the Himalaya is of about 1250～250km (Achache et al., 1984 ; Powell et al., 1988 ; Dewey et al.,1989 ; Klootwijk et al., 1992 ; Matte et al., 1997). This discrepancy between the amount of shortening estimated by balancing the Himalayan belt and by plate reconstruction favour the existence of a greater India buried up to 1000km north of the present\|day Indus suture zone and subducted before Middle Eocene time (Klootwijk et al., 1979 ; Patriat and Achache, 1984).

Three-dimensional （3D） topological insulators represent a new state of quantum matter with a bulk gap and odd number of relativistic Dirac fermions on the surface. The unusual surface states of topological insulators rise from the nontrivial topology of their electronic structures as a result of strong spin-orbital coupling. In this review, we will briefly introduce the concept of topological insu- lators and the experimental method that can directly probe their unique electronic structure： angle resolved photoemission spectroscopy （ARPES）. A few examples are then presented to demonstrate the unique band structures of different families of topological insulators and the unusual properties of the topological surface states. Finally, we will briefly discuss the future development of topological quantum materials.

AIM: To construct PGEX-1λT-FALL-39 expression vector and its mutant vector, and study the relationship of function and structure. METHODS: A cDNA encoding mature FALL-39 was cloned from SPCA-1 cell mRNA and the prokaryotic expression vector PGEX-1λT-FALL-39 was constructed. Two kinds of polymerase chain reaction (PCR) for the site-direction mutagenesis were used to construct FALL-39 mutant expression vector, FALL-39-Lys- 32 and FALL-39-Lys-24. Minimal effective concentration, minimal inhibitory concentration, and minimal bacteri- cidal concentration were used to assay the antibacterial activities of these peptides. Effects of different solution on the antibacterial activity of FALL-39 and FALL-39-Lys-32 were observed by CFU determination. The hemolytic effects of these peptides were also examined on human red blood cells. RESULTS: Two site-specific mutants FALL-39-Lys-32 and FALL-39-Lys24 were obtained by PCR-induced mutagenesis. In comparison with two-step PCR which required two pairs of primers, one step PCR which required one pair of primers is a simple and efficient method for the PCR based site-specific mutagenesis. Using the prokaryotic expression system, the E coli-based products of recombinant FALL39 and its mutant peptides were also obtained. The antibacterial assay showed that FALL-39-Lys-32 and FALL-39-Lys24 were more potential in the antibacterial activity against E coli ML35p and Pseudomonas aeruginosa ATCC27853 than that of FALL-39, and no increase in hemolysis was observed at the antibacterial concentrations. The antibacterial activity of FALL-39-Lys-32 against E coli was more potent than that of FALL-39 in NaCl-containing LB medium, while its activity was almost the same as FALL-39 in SO4 containing 2- Medium E. CONCLUSION: PCR-based mutagensis is a useful model system for studying the structure and function relationship of antimicrobial peptides.
