A Microenvironment, Rather than Chemical, Initiates the Cardiomyogenic Differentiation of Marrow Stromal Cells

To investigate the potential of cardiomyogenic differentiation of rat bone marrow stromal cells (MSCs), they were exposed to 5-azacytidine tretments (single/repeat) at varying concentrations (3, 5, 10 μmol/L) and the fates of the cells were analyzed by immunocytochemistry, Western blot and the reporter gene of enhanced cyan fluorescent protein (ECFP) under the control of ventricular myosin light chain 2.(MLC2.) promoter. MSCs were also cocultured with cardiomyocytes for periods up to 16 days, the expression of cardiac myosin heavy chain (MHC) and troponin I (Tn I) proteins was analyzed. After the induction with 5-azacytidine, neither spontaneously beating cells nor myotubes were found; MHC and Tn I proteins were also undetectable and no ECFP-positive MSCs were detected. But when cocultured with cardiomyocytes, spontaneously contracting MSCs were observed and cardiac specific proteins could be detected. The results proved that the novel effects of 5-azacytidine on the cardiomyogenic differentiation of MSCs should be questioned and a direct intercellular communication with cardiomyocytes is necessary for MSCs to differentiate into cardiomyocytes. Quantitative analysis of intestinal gas in patients with acute pancreatitis

BACKGROUND: Disturbance of gastrointestinal function is a common complication in the early phase of acute pancreatitis (AP). Intestinal gas may reflect the function of the gut. Using plain abdominal radiographs, we investigated whether intestinal gas volume is related to AP. METHODS: Plain abdominal radiographs of 68 patients with AP within 2. hours after admission and 2. normal controls were digitized and transmitted to a computer. The region of intestinal gas was identified by an image manipulation software and the gas volume score (GVS) was calculated. The relationships between the GVS values and various clinical factors of AP were analyzed. RESULTS: The GVS in the AP group was 0.084±0.016, in the mild AP (MAP) group 0.070±0.005, and in the severe AP (SAP) group 0.094±0.013; all values were higher than that in the control group (P2. P<0.01) and the acute physiology and chronic health evaluation II (APACHE II) scores (r=0.801, P<0.01). In addition, the GVS in patients with secondary pancreatic and/or peripancreatic infection was 0.107±0.014, higher than that in patients without secondary infection (P<0.01). GVS was not related to gender, age, etiology or clinical outcome of AP. CONCLUSIONS: Intestinal gas volume is significantly elevated in patients with AP. It is closely related to Ranson’s and APACHE II score and secondary pancreatic and/or peripancreatic infection. GVS may be a new prognostic tool for assessing the severity of AP in the early course of the disease.

Objective:To study the biological effect of magnetic field in mice ***: With a piece of permanent magnet embeded in mice bodies beside the femoral artery and vein to measure the electrophoretic velocity(um/s).Result: The magnetic field in mice bodies on the experiment group that the electrophoretic velocity is faster more than control and free group. Conclusion:The magnetic field in animal’s body can raise the neg ative electric charges on the surface of erythrocyte to improve the microcirculation, this is the biological effect of magnetic field.

Clinical and endoscopic analysis of gastric Dieulafoy’s lesion

AIM:To investigate the incidence,location,clinical presentation,diagnosis and effectiveness of endoscopic treatment of gastric Dieulafoy's lesion(DL)in china. METHODS:All patients who received emergency upper gastrointestinal(GI)endoscopy due to gastric DL from February 2.00 to August 2.08 at GI endoscopy center of renmin {2. of wuhan university were included in this *** clinical presentation,medical history,location and characteristics of DL methods and effectiveness of therapy of patients with DL were retrospectively analysed by chart ***-term follow-up data were collected at outpatient clinics or telephone interviews. RESULTS:Fifteen patients were diagnosized with DL,which account for 1.04%of the source of bleed- ing in acute non-variceal upper GI *** comorbidities were found in one patient with hypertension and diabetic *** or combined therapy with hemoclip produced primary hemostasis in 92.8%(13/14) of patients. CONCLUSION:DL is uncommon but life-threatening in *** proved to be safe and effective in controlling bleeding from DL.

AIM: To investigate the effect of theophylline on the naive T cell differentiation and the probable role of interleukin- 12.(IL-12.. METHODS: Naive cord blood T cells were treated with theophylline 10 mg/L for 3 d after stimulation with PHA 100 mg/L. Differentiation of Tcells was analyzed by flow cytometry. Theophylline 10 mg/L and IL-12. mAb 0.02. mg/L were added in cord blood mononuclear cell (CBMC) cultures primed with LPS 1 mg/L to detect the levels of IL-12.and IL-12.40. The whole blood cultures were obtained from twelve health volunteers with or without administration of theophylline (2.0 mg). Cytokines were measured by enzyme linked immuno-sorbent assay. RESULTS: Theophylline promoted T helper 1 (Th1) cells differentiation from naive T cells (2..9 %±10.3 % vs 9.4 %±5.6 %, P2.deviation. But theophylline inhibited the production of IL-12.and IL-12.40 by CBMC in vitro (2.±6 ng/L vs 57±14 ng/L and 88±34 ng/L vs 2.4±82.ng/L, P2.and IL-12.40 levels in whole blood cultures from healthy subjects (19±11 ng/L vs 31±15 ng/L and 92.13 ng/L vs 196±49 ng/L, P2.seemed not to be involved in this process.

Effect of angiotensin II receptor 1 antisense oligodoexynucleotides on physiological and pathophysiological growth of cardiomyocytes

AIM: To evaluate the role of angiotensin II receptor 1 antisense oligodexynucleotides (AT1R-AS-ODNs) on physi-ological and pathophysiological growth of cardiomyocytes from normotensive rats. METHODS: Cardiomyocyteswere transfected with AT1R-AS-ODNs (2.0 nmol/L) followed by treatment with or without angiotensin II (1 mol/L).In situ hybridization and Western blot were used for AT1R mRNA and protein detection, respectively. c-Jun N-terminal protein kinase (JNK) activity was characterized by immune complex kinase assay. c-Jun protein expres-sion was examined by immunocytochemistry. DNA content was detected by flow cytometric assay. Atrial natri-uretic factor (ANF) expression was identified by radioimmunoassay. RESULTS: Treatment with AT1R-AS-ODNsfor 2. h resulted in 51.2.% decrease in AT1R mRNA and 60.7 % in protein (P2.h, c-Jun protein expression was increased. By 2. h, angiotensin II caused a markedincrease both in G0/G1 and G2.M DNA content, and increased ANF expression by 1.8-fold. All these were inhibitedby AT1R-AS-ODNs pretreatment. In contrast, sense sequence was ineffective. CONCLUSION: Decrease ofAT1R expression by AS-ODNs did not interfere with normal growth, but protected cardiomyocytes from angio-tensin II-dependent pathophysiological growth.

Comprehensive Diagnostic Value of P53, p2.WAF1 and Proliferating Cell Nuclear Antigen for Lung Cancer

The expression of P53, p2.WAF1 and proliferating cell nuclear antigen （PCNA） was detected in 114 samples of lung cancer patients （with 89 cases benign lung tissue as control） and the diagnostic value of these markers was evaluated. The results show the following：（1） The positive expression rates of P53, p2.WAF1 and PCNA in samples of lung cancer were 47.37%, 75.44% and 80.70%, respectively, which were significantly higher than that in the samples of benign lung diseases （ p 〈 0. 001 ）. The odds ratios were 39.15, 5.75, and 6.76, respectively. This indicates that the expression of P53, p2.WAF1 and PCNA was helpful for the diagnosis of lung cancer. （2. For the diagnosis of lung cancer, the positive likelihood ratio of P53 was 2..08, which were significantly higher than that of p2.WAF 1 （2.16）, PCNA （2.11） and of all the combined tests. This shows that P53 expression was the most valuable for diagnosis of lung cancer. （3） For the diagnosis of lung cancer, the negative likelihood ratio of P53/p2.WAF 1/PCNA parallel test was 0.057 1, which was lower than that of other single and combined tests. This indicates that P53/p2.WAF 1/PCNA parallel test has high diagnostic value for exclusion of lung cancer.

Detection of type Ⅳ collagenase activity in malignant ascites

AIM: Type IV collagenase participates in invasion and metastasis of cancer cells. Malignant asdtes is a manifestation of advanced malignant disease that is associated with invasion and metastasis of the peritoneal cavity. Thus, it is reasonable to hypothesize that type IV collagenase is linked to malignant ascites. The purpose of our study was to detect type IV collagenase activity in malignant ascites so as to provide the scientific basis for clinic diagnosis and treatment of malignant ascites. METHODS: Cirrhotic ascites (n=36), tuberculous ascites (n=8) and malignant ascites (n=-2.) from patients with gastric cancer (n=6), colon cancer (n=5), ovarian cancer (n=8) and other cancers (n=4), including 2.hepatocellular cancers, 1 pancreatic cancer, 1 primary peritoneal carcinoma were collected by paracentesis. The ascites were made cellfree by centrifugation and stored frozen at -70℃ before determination. Type IV collagenase activity was determined by gelatin zymography. RESULTS: The activity of matrix metalloproteinases-2.and -9 could not be detected in ascites of hepatic cirrhosis and tuberculous peritonitis but could be detected in 2. and 18 out of 2. malignant ascites respectively. The positive rate of type IV collagenase (MMP-2. 87.0% and MMP-9, 78.3%) was higher than that by routine ascites tests (P2.was higher than that of MMP-9 (P=0.02.<0.05). CONCLUSION: Type IV collagenase is positive in malignant ascites. Detection of type IV collagenase activity is useful in qualitative diagnosis of ascites. Type IV collagenase may play an important role in malignant ascites formation.
