AIM: Hepatic stellate cell (HSC) plays a pivotal role inliver fibrosis and is considered as the therapeutic targetfor the treatment of hepatic fibrosis. Tyrosine proteinkinase plays an important role in the proliferation,activation of HSC. The purpose of the study is toinvestigate the effects of the tyrosine protein kinaseinhibitor genistein onthe proliferation and activationof cultured rat HscMETHODS: Rat HSC were isolated from Wistar rats by insitu perfusion of collagenase and pronase and single-stepdensity Nycodenz gradient. Culture-activated HSC wereserum-starved and incubated with 10-3 to 10-5 mol/Lconcentration of genistein for 2., 48 or 72.h. In PDGF-induced HSC proliferation, HSC were stimulatel with L0μg@L-1PDGF-BB for 15 min, and then treaeed with genisteinfor the same time. Cell proliferation was measured byMTr assay and based on flow cytometric analysis of cellcycle. The a-smooth muscle actin (α-SMA) expression inHSC was studied with confocal laser microscopy and flowcytometry. c-fos c-jun and cyclin D1 expression in HSCwas also detected by flow ***: Genistein inhibited basal and PDGF-inducedproliferation of HSC at the concentration of 10-8 to 10-5mol/L, and treatment with 10-7 mol/L concentration ofgenistein for 48 h inhibited the HSC proliferationsignificantly (the inhibition rate was 70.3 %, P＜0.05).Immunofluorescence detected by confocal lasermicroscopy and flow cytometry showed that treatmentwith 10-7 mol/L genistein for 48 h suppressed theexpression of α-SMA significantly in HSC (the specificfluorescence intensity were 60.2.2..5 vs 35.3±11.6and 12.8± 10.4 vs9.54±6.39, respectively, both P＜0.05).The intensity of c-fos, c-jun and cydin D1 expression ofHSCs treated with 10-7 mol/L genistein for 48 h wasalso significantly decreased compared with the ***: Genistein influences proliferation of HSC,suppresses the expression of α-SMA in HSC and tinhibits the intensity of c-fos, c-jun and cyclin D1expression of HSCs. BACKGROUND: It has been demonstrated that traditional antiepileptics, such as phenytoin, carbamazepine (CBZ), phenobarbital, etc., can result in the decrease of thyroid hormone of epileptic patients. However, there is still no sufficient evidence for the studies about the effect of new-type antiepileptics, such as topiramate (TPM), on thyroid hormones. OBJECTIVE: To observe the effects of TPM and CBZ on the level of thyroid hormones in serum of adults with epilepsy. DESIGN: A comparative observation. SETTING: department of Neurology, sichuan Provincial People's Hospital. PARTICIPANTS: Totally 100 outpatients or inpatients newly diagnosed to have epilepsy were selected from the department of Neurology, sichuan Provincial People's Hospital from July 2.03 to August 2.05, including 60 males and 40 females, aged 18-70 years. All the patients were accorded with the standard for the classification of epilepsy set by International League Against Epilepsy (ILAE) in 1981; Had been Informed and agreed with the detection; Had no history of thyroid gland disease; Had not taken any drugs could affect the thyroid function. Meanwhile, 40 adult healthy examinees were selected from our hospital as the control group, including 2. males and 16 females, aged 18-65 years. METHODS: ① The 100 epileptic patients were randomly divided into TPM group (n =50) and CBZ group (n =50), and they were treated with TPM (Xian-Janssen Pharmaceutical, Ltd.; Batch number: 03AS032. Norm: 2. mg/tablet) and CBZ (Shanghai Sunve Pharmaceutical Co., Ltd.; Batch number: 0302.1, Norm: 100 mg/tablet) respectively. The initial dosage of TPM was 2. mg per day, increased by 2. mg every week, the objective dosage of 100-2.0 mg per day was maintained when the symptoms were satisfactorily controlled. The dosage of CBZ was 6-8 mg/kg per day. AIM: To explore the role of focal adhesion kinase (FAI)in the apoptosis in culture-activated rat hepatic stellatecells (HSCs) using a specific anti-FAK ***: Rat HSCs were prepared from Wistar rats byin situ perfusion of collagenase and pronase and single-step density Nycodenze gradient. Culture-activatedHSCs were serum-starved and treated with the anti-FAK antibodies for 2., 48 or 72.h. The apoptosis of HSCwas detected by DNA-fragment assay, flow cytometryand caspase-3 activity determination. The expressionof tissue inhibitor of metalloproteinase-1 (TIMP-1)mRNA was assessed by reverse transcriptionpolymerase chain reaction (RT-PCR).RESULTS: The experiment showed that anti-FAKantibodies induced apoptosis of culture-activated ratHSCs. This phenomenon displayed the classical featuresof apoptotic cell death (DNA fragmentation, cell cycleanalysis) after treated with 30 mg@L-1 FAK antibody for72.h, and accompanied by a significant increase ofcespase-3 activity(12.8±76) vs (309±2.) nmol@min-1@ g-1, t=-2.8.5, P＜0.05. Meanwhile, treatment with theFAK antibody in HSCs could markedly decrease theTIMP-1 mRNA expression (0.07±0.01 vs 0.38±0.03, t=2.72. P＜0.05).CONCLUSION: FAK plays an important role in the survivalof HSCs and the specific anti-FAK antibody could inducethe apoptosis in rat HSCs.

BACKGROUND: Ideal model of poststroke depression (PSD) may be induced in rats guided by the theoretical evidence that 'primary endogenous mechanism' and 'reactivity mechanism' theories for PSD in human being. OBJECTIVE: To investigate the feasibility of comprehensive methods to induce PSD models in rats. DESIGN: A randomized controlled animal trial. SETTING: department of Neurology, Affiliated Hospital of chengdu University of Traditional Chinese Medicine. MATERIALS: Male SD rats of SPF degree, weighing 350-500 g, were provided by the experimental animal center of chengdu University of Traditional Chinese Medicine. The rats were raised for 1 week adaptively, then screened behaviorally by open-field test and passive avoidance test. Forty-five rats with close scores were randomly divided into normal control group (n =10), simple stroke group (n =10), stress group (n =10) and PSD group (n =15). METHODS: The experiments were carried out in the laboratory of chengdu University of Traditional Chinese Medicine from July 2.02.to February 2.03. ① Rat models of focal cerebral ischemia were induced by thread embolization, then treated with separate raising and unpredictable stress to induce PSD models. ②The neurologic deficit was evaluated by Longa 5-grade standard (the higher the score, the severer the neurologic deficit) and horizontal round rod test (normal rat could stay on it for at least 3 minutes). ③ The behavioral changes of PSD rats were evaluated by the saccharin water test, open-field text and passive avoidance test, including the changes of interest, spontaneous and exploratory activities, etc. ④ The levels of monoamine neurotransmitters, including norepinephrine (NE), serotonin (5-HT) and dopamine, in brain were determined using fluorospectrophotometry. AIM: To investigate the correlation between subcellulardaunorubicin distribution and the multidrug resistancephenotype in drug-resistant cell line SMMC-772./***: The multidrug resistant cell line SMMC-772./R, a human hepatocellular carcinoma cell line,was established. Antisense oligonudeotides (AS-ODN)were used to obtain different multidrug resistancephenotypes by inhibiting the expression of mdr1 geneand/or multidrug resistance-related protein gene(mrp)using Lipofectamine as delivery agent. Expression ofmdr1 and mrp genes was evaluated by RT-PCR andWestern blotting. Intracellular daunorubicin (DNR)concentration was measured by flow *** DNR distribution was analyzed by confocallaser scanning microscopy. Adriamycin (ADM) and DMRsensitivity was examined by MTT ***: Low level expression of mdr1 and mrp mRNAsand no expression of P-Glycoprotein(P-gp) andmultidrug resistance-related protein (P190) weredetected in parental sensitive cells SMMC-772./S, butover-expression of these two genes was observed indrug-resistant cell SMMC-772./R. The expression ofmdr1 and mrp genes in SMMC-772./R cells was downregulated to the level in the SMMC-772./S cells by AS-ODN. Intracellular DNR concentration in SMMC-772./S cells was 10 times higher than that in SMMC-772./Rcells. In SMMC772./S cells intracellular DNRdistributed evenly in the nucleus and cytoplasm, whilein SMMC-772./R cells DNR distributed in a punctatepattern in the cytoplasm and was reduced in thenucleus. DNR concentration in SMMC-772./R cells co-transfected with AS-ODNs targeting to mdr1 and mrpmRNAs recovered to 2. percent of that in SMMC772./Scells. Intracellular DNR distribution pattern in drug-resistant cells treated by AS-ODN was similar to drug-sensitive cell, and the cells resistance index (RI) to DNRand ADM decreased at most from 88.0 and 116.0 to 4.0and 2.3, respectively. AIM:To evaluate the value of 18F-fluorodeoxyglucose positron emission tomography/computed tomography(18F-FDG PET/CT) in the restaging of resected rectal ***:From January 2.07 to Sep 2.08,2. patients who had undergone curative surgery resection for rectal carcinoma with suspicious relapse in conventional imaging or clinical findings were retrospectively enrolled in our *** patients underwent 2.
