Chronic myeloid leukemia(CML)is a common adult *** the acute phase of the disease and the adverse effects of anti-cancer treatments can lead to a poor *** N6-methyladenine(m^(6)A)modification plays an important regulatory role in various physiological and pathological ***1429 is a known m^(6)A regulator,but the biological role of KIAA1429 in CML is *** this study,we observed that the m^(6)A levels and KIAA1429 expression were significantly up-regulated in patients with blast phase ***,KIAA1429 regulated the total level of RNA m^(6)A modification in the CML cells and promoted the malignant biological behaviors of CML cells,including proliferation,migration,and imatinib *** KIAA1429 in CML cells reduced the stability of RAB27B mRNA through the m^(6)A/YTHDF1 axis,consequently inhibiting CML proliferation and drug efflux,ultimately increasing the sensitivity of CML cells to ***,the knockdown of RAB27B also inhibited the proliferation and drug resistance of CML cells and promoted their ***,a recently developed anti-cancer agent,suppressed the expression of KIAA1429 and CML cell proliferation and promoted cell *** also inhibited the tumorigenesis of CML cells in *** combined use of rucaparib and imatinib enhanced the sensitivity of CML cells to *** study provides evidence that elevated KIAA1429 expression in the blast phase of CML enhances the stability of RAB27B mRNA through the m^(6)A/YTHDF1 axis to up-regulate RAB27B expression,thereby promoting CML *** exerts inhibitory effects on KIAA1429 expression and thus reduces CML progression.

Hepatic ischemia-reperfusion injury(HIRI)is the cause of postoperative hepatic dysfunction and failure,and even *** an important biological effector molecule,hydrogen sulfide(H_(2)S)of mitochondria as a gasotransmitter that is usually used to protect against acute HIRI ***,the exact relationship between HIRI and mitochondrial H_(2)S remains tangled due to the lack of an effective analytical ***,we have fabricated a mitochondria-targeted H_(2)S-activatable fluorogenic probe(Mito-GW)to explore the stability of mitochondrial H_(2)S and track the changes of mitochondrial H_(2)S during the *** virtue of pyridinium electropositivity and its amphiphilicity,Mito-GW could accumulate in *** goes through an analyte-prompted immolation when reacts with H_(2)S,resulting in the releasing of the fluorophore(GW).Therefore,the extent of Mito-GW conversion to GW can be used to evaluate the changes of mitochondrial H_(2)S level in living cells and *** proof-of-principle,we have used MitoGW to demonstrate the mitochondria H_(2)S-levels increase and then decrease during HIRI in vitro and in *** research highlights the tremendous potential of Mito-GW as a mitochondrial H_(2)S fluorogenic probe in elucidating the pathogenesis of HIRI,providing a powerful tool for promoting future research on hepatology.

Integrating single Ni site and PtNi alloy on two-dimensional porous carbon nanosheet for efficient catalysis in fuel cell

The performance of catalyst depends on the intrinsic activity of active sites and the structural characteristics of the ***,we simultaneously integrate single nickel(Ni)sites and platinum-nickel(PtNi)alloy nanoparticles(NPs)on a two-dimensional(2D)porous carbon nanosheet,demonstrating remarkable catalytic performance in the oxygen reduction reaction(ORR).The single Ni sites can activate the oxygen molecules into key oxygen-containing intermediate that is further efficiently transferred to the adjacent PtNi alloy NPs and rapidly reduced to H_(2)O,which establishes a relay catalysis between active *** porous structure on the carbon nanosheet support promotes the transfer of active intermediates between these active sites,which assists the relay catalysis by improving mass ***,the obtained catalyst demonstrates a half-wave potential of up to 0.942 V,a high mass activity of 0.54 A·mgPt^(−1),and negligible decay of activity after 3.,000 cycles,which are all superior to the commercial Pt/C catalysts with comparable loading of *** theoretical calculation results reveal that the obtained catalyst with defect structure of carbon support presents enhanced relay catalytic effect of PtNi alloy NPs and single Ni sites,ultimately realizing improved catalytic *** work provides valuable inspiration for developing low platinum loading catalyst,integrating single atoms and alloy with outstanding performance in fuel cell.

BACKGROUND Colorectal anastomotic occlusion is a serious complication of colorectal cancer *** several treatment strategies have been proposed,the mana-gement of anastomotic occlusion remains *** this report,we present a case of anastomotic occlusion recanalization performed using a novel technique involving two endoscopes,one for radial incision and the other serving as a guide *** novel technique offers significant advantages in terms of operational feasibility,reduced invasiveness,rapid recovery,and shortened hospital *** SUMMARY A 3.-year-old man underwent low anterior resection and prophylactic double-lumen ileostomy for rectal cancer in June,*** months later,complete anastomotic occlusion was observed on ***,we developed a novel atresia recanalization *** endoscopes were placed,one through the colonic anastomosis and the other through the anus.A radial incision was successfully made from the colonic side,guided by the light of the endoscope from the anal *** recanal-ization was performed within 20 *** weeks after recanalization,colonoscopy revealed that the diameter of the colorectal anastomosis was approximately 16 mm and the patient therefore underwent stoma reversal in *** the follow-up period of approximately one year,the patient remained well and no stenosis or obstruction symptoms were *** Endoscopic atresia recanalization of colorectal anastomotic occlusion assisted by an opposing light source is safe and effective.

Vanadium disulfide(VS_(2)) as a typical two-dimensional transition metal chalcogenide has excellent competitiveness for sodium-ion storage due to its wide layer spacing(0.575 nm),high theoretical capacity of 93.mAh·g^(-1) originating from multi-electron electrochemical ***,continuous sodiation process accompanied by crystal structural evolution and collapse cause rapid capacity ***,novel few-layer VS_(2)nanosheets with open(001) crystal planes are in-situ constructed on reduced graphene oxide to solve these issues mentioned *** indicates that few-layer VS_(2)provides more Na^(+) storage activity due to the low Na^(+)surface migration energy barrier on exposed crystal(001)*** flexible and high electronic conductivity of carbon matrix also effectively builds multi-level buffer structure and electron transport kinetics to boost the Na^(+)insertion/conversion reactive activity on VS_(2) as well as Na^(+) pseudocapacitance storage kinetics on edges and defects of nano *** coupling effects result in high rate capability and long cycling stability as a battery/capacitor *** delivers conspicuous high energy density of 81 and 40 Wh·kg^(-1) at power density of 118 and 10,286W·kg^(-1),as well as 80% energy retention rate after 5000cycles,confirming its great application potential in sodiumbased storage devices.

Defects engineering is an effective strategy for manipulating electromagnetic parameters and enhancing electromagnetic wave(EMW)absorption ***,the relationship between them is not clear,especially in solid solution *** this work,a series of(Cr_(1-x)V_(x))_(2)AlC MAX phase solid solutions with layered structure were prepared via tuning the ratio of Cr and V to explore their EMW absorption *** experimental results indicated that the doping of V atoms at the M-site could effectively regulate its impedance matching and EMW absorption properties by introducing appropriate numbers of defects in the crystal,such as twin boundaries,dislocations and lattice *** them,if Cr:V=3.1,Cr_(1.5)V_(0.5)AlC,as radar absorption materials,could reach a strong reflection loss of-51.8 dB at the frequency of 12.8 GHz under an ultra-thin thickness of 1.3.*** reflection loss value could attain-10 dB in a wide frequency range of 2.7-18 GHz and thickness range of 1-5 *** addition,after high temperature and acid-alkali immersion treatment,this sample still had good EMW absorption capability,and the effective absorption bandwidth was enhanced from 2.3.to 2.6 GHz after concentrated acid immersion or 3.1 GHz after concentrated alkali *** work has great reference significance for the research and development of high-performance MAX-based EMW absorption materials in harsh environments.

Cymbidium(Orchidaceae:Epidendroideae),with around 60 species,is widely-distributed across Southeast Asia,providing a nice system for studying the processes that underlie patterns of biodiversity in the ***,phylogenetic relationships of Cymbidium have not been well resolved,hampering investigations of species diversification and the biogeographical history of this *** this study,we construct a plastome phylogeny of 56 Cymbidium species,with four well-resolved major clades,which provides a framework for biogeographical and diversification rate *** dating and biogeographical analyses show that Cymbidium likely originated in the region spanning northern IndoBurma to the eastern Himalayas during the early Miocene(~21.10 Ma).It then rapidly diversified into four major clades in East Asia within approximately a million years during the middle *** *** to the adjacent regions(borneo,Philippines,and Sulawesi)primarily occurred during the Pliocene-Pleistocene *** analyses indicate that the net diversification rate of Cymbidium has decreased since its origin,and is positively associated with changes in temperature and monsoon *** hydrothermal conditions brought by monsoon intensification in the early Miocene possibly contributed to the initial rapid diversification,after which the net diversification rate was reduced with the cooling climate after the middle *** transition from epiphytic to terrestrial habits may have enabled adaptation to cooler environments and colonization of northern niches,yet without a significant effect on diversification *** study provides new insights into how monsoon activity and temperature changes affected the diversification dynamics of plants in Southeast Asia.

Hyperlipidemia is a major risk factor for erectile dysfunction(ED).Oxidative stress and phenotypic modulation of corpus cavernosum smooth muscle cells(CcSMCs)are the key pathological factors of ED.N-acetylcysteine(NAC)can inhibit oxidative stress;however,whether NAc can alleviate pathological variations in the corpus cavernosum and promote erectile function recovery in hyperlipidemic rats remains unclear.A hyperlipidemia model was established using 27 eight-week-old male Sprague-Dawley(SD)rats fed a high-fat and high-cholesterol diet(hyperlipidemic rats,HR).In addition,9 male SD rats were fed a normal diet to serve as controls(NC).HR rats were divided into three groups:HR,HR+normal saline(NS),and HR+NAC(n=9 for each group;NS or NAc intraperitoneal injections were administered daily for 16 weeks).Subsequently,the lipid profiles,erectile function,oxidative stress,phenotypic modulation markers of ccsMCs,and tissue histology were *** experimental results revealed that erectile function was significantly impaired in the HR and HR+NS groups,but enhanced in the HR+NAC *** lipid levels,over-activated oxidative stress,and multi-organ lesions observed in the HR and HR+NS groups were improved in the HR+NAC ***,the HR group showed significant phenotypic modulation of CCSMCs,which was also inhibited by NAC *** report focuses on the therapeutic effect of NAc in restoring erectile function using a hyperlipidemic rat model by preventing CcSMC phenotypic modulationand attenuating oxidativestress.
