The role of sHLA-G on the abnormal pregnant outcome induced by Toxoplasma gondii infection

摘      要:Background:T.gondii can affect fetus via vertical transmission and lead to adverse pregnant outcomes.The soluble human leukocyte antigen molecules G(sHLA-G)play a key role in the success of pregnancy through interacting with decidual natural killer cell(dNK)inhibitory receptors at the maternal-fetal interface.Objective:In this study,we aimed to investigate the role of sHLA-G on the abnormal pregnancy outcomes induced byToxoplasma gondii through the vivo and vitro experiments.Materials and methods:Concentration of the sHLA-G in BeWo culture supernatant,and the Qa-2 in the placenta supernatant,amniotic fluid and serum of pregnant CBL/6 mice were all analyzed by ELISA.Levels of NKG2A in mouse and KIR2DL4 expressed on human dNK cells were analyzed by flow cytometry,respectively.Results:Results showed that the levels of Qa-2 and its ligand NKG2A of mice in infected group were significantly higher than that controls(p0.05).Similar results were showed that the sHLA-G and the KIR2DL4 in human were significantly up-regulated(p0.05)with T.gondii infection.Furthermore,blocking experiment of sHLA-G revealed that sHLA-G could up-regulate the expression of KIR2DL4.Conclusions:Theses results demonstrated that sHLA-G could cause the imbalance of immune molecules leading to excessive immunological tolerance at the maternal-fetal interface,which then contributed to T.gondii immunity escaping and affecting fetus via vertical transmission and resulted in adverse pregnant outcomes.
