Directed yeast genome evolution by controlled introduction of trans-chromosomic structural variations

Naturally occurring structural variations(SVs)are a considerable source of genomic variation that can reshape the 3D architecture of *** methods aimed at introducing the complex SVs and their related molecular mechanisms have remained *** this study,an SV-prone yeast strain was developed using Synthetic Chromosome Rearrangement and Modification by LoxP-mediated Evolution(SCRaMbLE)technology with two synthetic chromosomes,namely synV and *** biosynthesis of astaxanthin is used as a readout and a proof of concept for the application of SVs in *** findings showed that complex SVs,including a pericentric inversion and a trans-chromosome translocation between synV and synX,resulted in two neo-chromosomes and a 2.7-fold yield of ***,genetic targets were mapped,which resulted in a higher astaxanthin yield,thus demonstrating the SVs’ability to reorganize genetic information along the *** rational design of trans-chromosome translocation and pericentric inversion enabled precise induction of these ***,this study provides an effective tool to not only accelerate the directed genome evolution but also to reveal the mechanistic insight of complex SVs for altering phenotypes.

An organic-inorganic composite dip-coating method was adopted in order to obtain ideal water repellent cotton fabrics. To be specific, a dual-functional coating with both superhydrophobic and luminescent properties was prepared on cotton fabric by using a dip-coating solution comprising （3-aminopropyl） trimethoxysilane （APTMS） modified SrA1204：Eu2＋,Dy3＋particles, SU8, and fluorinated alkyl silane （FAS）. The micro/nano-scale roughness generated by SrA1204：Eu2＋,Dy3＋ particles, together with low-sur- face-energy FAS, rendered the cotton fabric superhydrophobic with a water contact angle of about 160° and a sliding angle as small as 2°. The coated fabric could withstand at least 100 cycles of standard laundry. The emission spectra of the coated fabric showed two emission peaks at 440 and 520 nm, which belonged to the blue and yellow-green color areas, respectively. The afterglow duration of the coated fabric was mainly influenced by the different depths of the trap levels in the SrA1204：Eu2＋,Dy3＋.

Synthesizing ring structure aviation fuels from biomass-derived platform molecules is challenging,especially for bridged ring structure aviation fuels which are typically achieved from fossil-derived ***,we report the synthesis of a series of ring structure biofuels in two steps by a combination of a solvent-free Michael-cyclization reaction and a hydrodeoxygenation(HDO)reaction from lignocellulosederived 5,5-dimethyl-1,*** biofuels are obtained with high overall yields up to 90%,which exhibit high densities of 0.81 g cm^(-3)-0.88 g cm^(-3)and high volumetric neat heat of combustion(VNHOC)values of 36.0 MJ L^(-1)-38.6 MJ L^(-1).More importantly,bridged-ring structure hydrocarbons can also be achieved in two steps by a combination of a Robinson annulation reaction and an HDO reaction to afford the final products at high overall yields up to 90%.The bridged-ring structure products have comparable high densities and high VNHOC values to the best artificial fuel JP-10(0.94 g cm^(-3)and 39.6 MJ L^(-1)).The results demonstrate a promising way for the synthesis of high-density aviation fuels with different fuel properties at high yields.

The ReaxFF can describe the properties of energetic materials(EMs)at equilibrium state,but does not work properly in simulating high-energy particle irradiation process because of its weak short-range *** this paper,a modification was made for such a potential by connecting ZieglerBiersack-Littmark(ZBL)potential to ReaxFF-lg through comparing to Density Functional Theory(DFT)results to accurately describe short-range *** modification,the newly fitted ReaxFF-lg/ZBL potential predicts better the equation of state for EMs In displacement cascade simulations,comparing to results from ab initio molecular dynamics(AIMD),ReaxFF-lg/ZBL presented the similar transferred energy from a primary knock-on atom to surrounding atoms,better than the original ReaxFF-lg *** large-scale displacement cascade simulations indicated ReaxFF-lg/ZBL could be applied for cascade simulations with PKA energy from less than 1 keV to high energy(e.g.35 keV)cases,which is suitable for effectively simulating high-energy displacement cascades in EMs using molecular dynamics method.

Genetic Structure Analysis of Human Remains from Khitan Noble Necropolis

Ancient DNA was extracted from 13 skeletal remains from the burial groups of Khitan nobles, which were excavated in northeast China. The hypervariable segment Ⅰ sequences （ HVS Ⅰ ） of the mitochondrial DNA control region, in the 13 individuals, were used as genetic markers to determine the genetic relationships between the individuals and the genetic affinity to other interrelated populations by using the known database of mtDNA. Based on the phylogenetic analysis of these ancient DNA sequences, the genetic structures of two Khitan noble kindreds were obtained, including the Yel Yuzhi＇s kindred and the Xiao He＇s kindred. Furthermore, the relationships between the Khitan nobles and some modern interrelated populations were analyzed. On the basis of the result of the analysis, the gene flows of the ancient Khitans and their demographic expansion in history was deduced.

In order to improve the elderly people's quality of life,supporting their walking behaviors is a promising ***,based on one ultrasonic motor,a wire-driven series elastic mechanism for walking assistive system is proposed and investigated in this *** contrast to tradition,it innovatively utilizes an ultrasonic motor and a wire-driven series elastic mechanism to achieve superior system performances in aspects of simple structure,high torque/weight ratio,quiet operation,quick response,favorable electromagnetic compatibility,strong shock resistance,better safety,and accurately stable force *** proposed device is mainly composed of an ultrasonic motor,a linear spring,a steel wire,four pulleys and one rotating *** overcome the ultrasonic motor's insufficient output torque,a steel wire and pulleys are smartly combined to directly magnify the torque instead of using a conventional gear *** the pulleys,there is one tailored pulley playing an important role to keep the reduction ratio as 4.5 ***,the prototype is manufactured and its actual performance is verified by experimental *** a one-second operating cycle,it only takes 86 ms for this mechanism to output an assistive torque of 1.6 N·*** this torque,the ultrasonic motor's speed is around 4.1 rad/***,experiments with different operation periods have been conducted for different application *** study provides a useful idea for the application of ultrasonic motor in walking assistance system.

Emerging contaminants(ECs)have aroused great concerns due to their widespread presence as well as their potential health and ecological *** monitoring data about ECs in Chinese rural areas are still *** this study,Taige Canal basin was selected as a typical rural area for a pilot *** occurrence of ECs,including pharmaceutical and personal care products(PPCPs),pesticides,plasticizers and flame retardants,in surface water and aquaculture ponds,as well as in wastewater treatment facilities,was comprehensively *** effectiveness of rural decentralized wastewater treatment facilities and centralized wastewater treatment plant(WWTP)in removing ECs was also *** 126 commonly used pesticides for target analysis,25 compounds were detected in surface water with high frequency of>80%.Besides,55 PPCPs,21 pesticides and 18 other industrial products were identified by non-target analysis using high resolution mass spectrometry(HRMS).Totally 18 compounds were detected in surface water with a frequency of>60%and 6 of them were found in all samples,including irbesartan,valsartan,propazine,buprofezin,bis(2-ethylhexyl)phthalate(DEHP)and tri-(2-chloroisopropyl)phosphate(TCPP),with mean concentrations of 3022,195,125,218,3738 and 545 ng/L,*** are 29 compounds(11 PPCPs,8 pesticides and 10 other industrial products)detected in water samples from the aquaculture ponds,of which 13 compounds were found in all three *** removal efficiencies of ECs in wastewater treatment facilities are generally limited,except valsartan(82%–86%),nobiletin(100%)and diisodecylphthalate(100%)were found to be significantly removed.

Impact of receiver inter-frequency bias residual uncertainty on dual-frequency GBAS integrity

Dual-Frequency Ground-Based Augmentation Systems(GBAS)can be affected by receiver Inter-Frequency Bias(IFB)when Ionosphere-Free(Ifree)smoothing is *** the framework of the proposed GBAS Approach Service Type F(GAST-F),the IFB in the Ifree smoothed pseudorange can be ***,IFB residual uncertainty still exists,which may threaten the integrity of the *** paper presents an improved algorithm for the airborne protection level considering the residual uncertainty of IFBs to protect the integrity of dual-frequency *** IFB residual uncertainty multiplied by a frequency factor is included in the Ifree protection level together with the uncertainty of other error *** verify the proposed protection level algorithm,we calculate the IFB residual uncertainties of ground reference receivers and user receiver based on BDS B1I and B3I dual-frequency observation data and carry out a test at the Dongying Airport GBAS *** results show that the proposed Ifree protection level with IFB residual uncertainty is 1.48 times the current protection level on *** probability of Misleading Information(MI)during the test is reduced from 3.2×10^(-4)to the required *** is proven that the proposed protection level can significantly reduce the integrity risk brought by IFB residual uncertainty and protect the integrity of dual-frequency GBAS.

A novel cationic Pt(Ⅱ)complex 2 with 2-(2,4-difluorophenyl)pyridine as the cyclometalating ligand and 1,10-phenanthroline as the auxiliary ligand has been synthesized and fully *** complex exhibits much higher aggregation-induced phosphorescent emission activity than that of a nonfluorinated complex 1 in CH_(3)CN/H_(2)*** complex 2 demonstrates efficient detection on picric acid(PA)in CH_(3)CN/H_(2)O,providing a high quenching constant(K_(SV)=2.3×10^(4) L/mol)and a low limit of detection(LOD=0.26μmol/L).In addition,complex 2 shows high selectivity for detection of PA in real water *** functional theory calculations and proton nuclear magnetic resonance spectra suggest that the detection mechanism is attributed to the photo-induced electron transfer.
