Early diagnosis and accurate boundary delineation are the key steps of tumor precision *** tumor cells(CTCs)detection of liquid biopsy can provide abundant information for early diagnosis of *** detection specificity and good enrichment features are two key factors for CTCs accurate identification in peripheral blood *** this purpose,iron oxide(IO)-based surface-enhanced Raman scattering(SERS)bioprobes with good biocompatibility,high detection sensitivity,remarkable detection specificity,and good enrichment efficiency,were developed for detecting different types of *** SERS bioprobes combined with programmed death ligand-1(PD-L1)antibody are regarded as an effective way to boost the targeting ability and detection specificity,benefiting for accurately capturing and identifying rare *** types of CTCs with different PD-L1 expression were accurately distinguished among white blood cells via high-resolution SERS mapping images and stable Raman ***,CTCs blood samples obtained from the triple negative breast cancer patients were also successfully recognized compared to that of health people,indicating IO@AR@PDA-a PD-L1 SERS bioprobe possessed great potential for CTCs detection in liquid ***,IO-based bioprobe exhibited excellent dual-modal imaging abilities of high-resolution SERS imaging mode and microimaging magnetic resonance imaging *** two highly complementary imaging modes endowed IO-based bioprobes unrivalled capacity in tumor boundary differentiation,supporting tumor accurate resection and precise *** our best knowledge,this is the first time that biocompatible IO-based SERS bioprobes without noble metal element were reported not only for CTCs accurate detection,but also for precise tumor boundary delineation,showing great advantages in tumor diagnosis and treatment.

P-tuned FeN2 binuclear sites for boosted CO2 electro-reduction

The recycling of CO2through electrochemical processes offers a promising solution for alleviating the greenhouse effect;however,the activation of CO2and desorption of*CO in electrocatalytic CO2reduction（ECR） frequently encounter high energy barriers and competitive hydrogen evolution reactions（HERs）,which are urgent problems that need to be *** this study,a catalyst(P100-Fe-N/C) with homogeneous P-tuned FeN2binuclear sites（N2PFe-FePN2） was successfully synthesised,demonstrating satisfactory performance in the ECR to CO.P100-Fe-N/C attains a peak FECOof 98.01% and a normalized TOF of664.7 h-1at-0.7 VRHE,surpassing the performance of the Fe binuclear catalyst without P and singleatoms *** the MEA cell,a FECOexceeding 90% can still be *** functional theory analysis indicates that the asymmetric coordination configuration induced by the incorporation of P facilitates a reduction in the system's *** modulation of P results in the d-band centre of the catalyst being positioned closer to the Fermi level,which facilitates the interaction of the catalyst with CO2,allowing more electrons to be injected into the CO2molecule at the Fe binuclear sites and inhibiting the *** P-tuned FeN2binuclear sites effectively lower the*CO desorption barrier.

Fighting hypoxia to improve photodynamic therapy-driven immunotherapy: Alleviating, exploiting and disregarding

Innovative anti-cancer therapies that activate the immune system show promise in combating cancers resistant to conventional treatments. Photodynamic therapy (PDT) is one such treatment, which not only directly eliminates tumor cells but also functions as an in situ tumor vaccine by enhancing tumor immunogenicity and triggering anti-tumor immune responses through immunogenic cell death (ICD). However, the effectiveness of PDT in enhancing immune responses is influenced by factors, such as photosensitizers and the tumor microenvironment, particularly hypoxia. Current clinically used PDT heavily relies on oxygen (O2) availability and can be limited by tumor hypoxia. Additionally, the tumor immunosuppressive microenvironment induced by hypoxia affects the anti-tumor immunity of tumor-infiltrating effector T cells. Meanwhile, the immunosuppressive myeloid-lineage cells are recruited to the hypoxic tumor tissue and exhibit higher immunosuppressive capabilities under hypoxia conditions. Consequently,numerous strategies have been developed to modulate tumor hypoxia or to create hypoxia-compatible PDT, aiming to reduce the effects of tumor hypoxia on PDT-driven immunotherapy. This review investigates these strategies, including approaches to alleviate, exploit, and disregard tumor hypoxia within the context of PDT/immunotherapy. It also emphasizes the role of advanced nanomedicine and its benefits in these strategies, while outlining current challenges and future prospects in the field.

The applications of single-cell and spatial technologies in recent times have revolutionized the present understanding of cellular states and the cellular heterogeneity inherent in complex biological systems. These advancements offer unprecedented resolution in the examination of the functional genomics of individual cells and their spatial context within tissues. In this review, we have comprehensively discussed the historical development and recent progress in the field of single-cell and spatial genomics. We have reviewed the breakthroughs in single-cell multi-omics technologies, spatial genomics methods, and the computational strategies employed toward the analyses of singlecell atlas data. Furthermore, we have highlighted the advances made in constructing cellular atlases and their clinical applications,particularly in the context of disease. Finally, we have discussed the emerging trends, challenges, and opportunities in this rapidly evolving field.

Synthesis, potential anticonvulsant and antidepressant effects of 2-(5-methyl-2,3-dioxoindolin-1-yl)acetamide derivatives

A new series of 2-(5-methyL2,3-dioxoindolin-1 y)acetamide derivatives were synthesized and evaluated for their anticonvulsive activity in a pentylenetetrazole (PTZ)-evoked convulsion model and antidepressant activity in the forced swimming test (FST) model. Eleven synthesized compounds were found to he protective against PTZ-induced seizure and showed the anticonvulsant activity. In addition, four of the synthesized compounds (41, 4m, 4p and 4q) showed potent antidepressant-like activity. Among these compounds, compound 41 was found to have the most potent antidepressant-like activity, and significantly reduced the duration of immobility time at 100 mg/kg dose level when compared to the vehicle control, which is similar to the reference drug lluoxetine. Gelatins and antioxidant peptides from Skipjack tuna(Katsuwonus pelamis)skins:Purification,characterization,and cytoprotection on ultraviolet-A injured human skin fibroblasts

For full use of Skipjack tuna(Katsuwonus pelamis)canning by-products,gelatins(STG)were extracted from tuna skins using acid(STG-A),enzyme(STG-E)and hot water(STG-H)methods with yields of 65.30±1.56%,39.83±1.61%,and 50.97±1.44%,*** to the high imino acid contents(228,216,and 213 residues/1000 residues for STG-A,STG-E,and STG-H,respectively),STG-A showed the highest transparency and gel strength *** acid analysis,sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE),ultraviolet(UV)and Fourier transform infrared spectroscopy(FTIR)analysis confirmed that STG-A,STG-E,and STG-H kept the main structure of type I collagen,but enzyme and hot water extraction methods showed much stronger hydrolysis ability onαandβ***,neutrase hydrolysate(STG-AH)of STG-A showed the highest 1,1-diphenyl-2-picrylhydrazyl radical(DPPH•)scavenging activity and the fraction STG-AH-І(products for preventing UVA injury.

High Fischer ratio oligopeptides from hard-shelled mussel:Preparation and hepatoprotective effect against acetaminophen-induced liver injury in mice

High Fischer ratio oligopeptides(HFOPs)have a variety of physiological activities,but there are few literatures reporting the hepatoprotective effects of HFOPs against acetaminophen(APAP)-induced liver *** ob-jectives of this study were to establish the preparation process of HFOPs from hard-shelled mussel(Mytilus coruscus)(HFOPs-HM)and investigate the hepatoprotective effects and potential mechanisms of HFOPs-HM against APAP-induced liver injury in ***,trypsin and papain were selected as the endo and exog-enous proteases for preparing HFOPs-HM according to the degree of hydrolysis(DH)of hard-shelled mussel hydrolysates,and activated carbon of XHJ-200 displayed the best effect to remove aromatic amino acids ac-cording to the Fischer ratio(OD220/OD280)and its parameters were optimized as adsorption time of 3.0 h and solid-liquid ratio of 1:*** HFOPs-HM’s Fischer ratio and average molecular weight were 26.03(>20)and 878.54 Da,***,a mice model of liver injury induced by APAP was ***-HM could significantly improve liver index and histopathological features,decrease the serum alanine aminotrans-ferase(ALT)and aspartate transaminase(AST)activities,increase the glutathione content and superoxide dis-mutase(SOD)and catalase(CAT)*** research demonstrated that HFOPs-HM could alleviate liver oxidative stress in liver tissue via activating the Nrf2 pathway to initiate the intracellular antioxidant enzyme *** addition,HFOPs-HM also suppressed liver inflammation by downregulating the secretion of TNF-α,IL-1β,and ***,HFOPs-HM could serve as an auxiliary functional dietary molecule applied in function products to alleviate drug-induced liver injury.

Identification and Expression Profile of a Neuropeptide LFRFamide-Like Gene During Different Stages of Gonadal Development in the Cephalopod Sepia pharaonis

Neuropeptides are widely distributed in vertebrates and invertebrates,regulating a variety of physiological activities in the organisms,such as metabolism,feeding and *** this study,to explore the function of neuropeptide LFRFamide in Sepia pharaonis,the full-length cDNA of LFRFamide-like gene(named SpLFRFL,MG869822.1)was identified with rapid amplification of cDNA ends(RACE)*** sequence of SpLFRFL was 860 bp in length and encoded 188 amino acids containing 4 different mature peptides:1 copy of PHTPFRFamide,NSLFRFamide,TIFRFamide,and 3 copies of *** alignment and phylogenetic analysis results showed that SpLFRFL shared high identity with LFRFamides of Sepia officinalis and Sepiella japonica and had the closest relationship with *** quantitative Real-time PCR(qRT-PCR),it was found that the SpLFRFL gene was highly expressed in the optic lobe and brain at three different stages during gonad development in both ***,the four mature peptides at a concentration of 0.01μmol L^(−1) could inhibit the protein synthesis in the Chinese hamster ovary cell strain-K1(CHOK1)induced by *** data suggest that SpLFRFL might be involved in the development and reproduction of *** results can contribute to future studies on neuropeptide evolution and function and benefit the cuttlefish farming.

Characterization of acid-and pepsin-soluble collagens from spines and skulls of skipjack tuna(Katsuwonus pelamis)

Acid-soluble collagen(ASC) and pepsin-soluble collagen(PSC) from the spine(ASC-SP and PSC-SP) and skull(ASC-SK and PSC-SK) of the skipjack tuna, Katsuwonus pelamis, were successfully isolated and characterized. The yields of ASC-SP, PSC-SP, ASC-SK and PSC-SK were(2.47 ± 0.39)%,(5.62 ± 0.82)%,(3.57 ± 0.40)%, and(6.71 ± 0.81)%, respectively, on the basis of dry weight. The four collagens contained Gly(330.2-339.1 residues/1 000 residues) as the major amino acid, and their imino acid contents were between 168.8 and 178.2 residues/1 000 residues. Amino acid composition, SDS-PAGE, and FTIR investigations confirmed that ASC-SP and ASC-SK were mainly composed of type I collagen, and had higher contents of high-molecular weight cross-links than those of PSC-SK and PSC-SP. The FTIR investigation also certified all the collagens had triple helical structure. The denaturation temperatures of ASC-SK, PSC-SK, ASC-SP, and PSC-SP were 17.8, 16.6, 17.6, and 16.5 °C, respectively. All isolated collagens were soluble at acidic pH(1-5) and lost their solubilities when the NaCl concentration was above 2%(W/V). The isolated collagens from the spines and skulls of skipjack tuna could serve as an alternative source of collagens for further application in food, cosmetic, biomedical, and pharmaceutical industries.

Pyropia haitanensis polysaccharide(LP)have been found for having many excellent functions such as *** Caenorhabditis elegans models,we evaluated the anti-aging activity of LP by observing the lifespan,reproduction,pharyngeal pumping,stress response,quantitative fluorescence of polyglutamic acid,and nuclear localization of DAF-16 of *** results reveal that LP could extend the adult lifespan of wild-type and polyQ nematodes,indicating a connection of its anti-aging benefit with the toxicity-suppressing *** number of polyglutamic acid aggregates in high concentration groups decreased by 24.39%(P<0.05)to the *** high-dose group strongly induced DAF-16 nuclear translocation over intermediate and cytosolic localizations compared with the control(P<0.001).Therefore,we believe that LP could extend the lifespan and reduce the protein aggregation in *** through nuclear DAF-16∷GFP expression.
