Transcriptome analysis of salt-responsive genes and SSR marker exploration in Carex rigescens using RNA-seq

Carex rigescens （Franch.） V. Krecz is a wild turfgrass perennial species in the Carex genus that is widely distributed in salinised areas of northern China. To investigate genome-wide salt-response gene networks in C. rigescens, transcriptome analysis using high-throughput RNA sequencing on C. rigescens exposed to a 0.4% salt treatment （Cr_Salt） was compared to a non-salt control （Cr_Ctrl）. In total, 57 742 546 and 47 063 488 clean reads were obtained from the Cr Ctrl and Cr Salt treatments, respectively. Additionally, 21 954 unigenes were found and annotated using multiple databases. Among these unigenes, 34 were found to respond to salt stress at a statistically significant level with 6 genes up-regulated and 28 downregulated. Specifically, genes encoding an EF-hand domain, ZFP and AP2 were responsive to salt stress, highlighting their roles in future research regarding salt tolerance in C. rigescens and other plants. According to our quantitative RT-PCR results, the expression pattern of all detected differentially expressed genes were consistent with the RNA-seq results. Furthermore, we identified 11 643 simple sequence repeats （SSRs） from the unigenes. A total of 144 amplified successfully in the C. rigescens cultivar LOping 1, and 69 of them reflected polymorphisms between the two genotypes tested. This is the first genome-wide transcriptome study of C. rigescens in both salt-responsive gene investigation and SSR marker exploration. Our results provide further insights into genome annotation, novel gene discovery, molecular breeding and comparative genomics in C. rigescens and related grass species.

Arbuscular Mycorrhizal Fungal Diversity Associated with Olea europaea L. Growing in yunnan Province, Southwestern China

Effect of nitrogen fertilization on yield, N content, and nitrogen fixation of alfalfa and smooth bromegrass grown alone or in mixture in greenhouse pots

Planting grass and legume mixtures on improved grasslands has the potential advantage of realizing both higher yields and lower environmental pollution by optimizing the balance between applied N fertilizer and the natural process of legume biological nitrogen fixation. However, the optimal level of N fertilization for grass-legume mixtures, to obtain the highest yield, quality, and contribution of N2 fixation, varies with species. A greenhouse pot experiment was conducted to study the temporal dynamics of N2 fixation of alfalfa （Medicago sativa L.） grown alone and in mixture with smooth bromegrass （Bromus inermis Leyss.） in response to the addition of fertilizer N. Three levels of N （0, 75, and 150 kg ha-1） were examined using 15N-labeled urea to evaluate N2 fixation via the 15N isotope dilution method. Treatments were designated NO （0.001 g per pot）, N75 （1.07 g per pot） and N150 （2.14 g per pot）. Alfalfa grown alone did not benefit from the addition of fertilizer N; dry matter was not significantly increased. In contrast, dry weight and N content of smooth bromegrass grown alone was increased significantly by N application. When grown as a mixture, smooth bromegrass biomass was increased significantly by N application, resulted in a decrease in alfalfa biomass. In addition, individual alfalfa plant dry weight （shoots＋roots） was significantly lower in the mixture than when grown alone at all N levels. Smooth bromegrass shoot and root dry weight were significantly higher when grown with alfalfa than when grown alone, regardless of N application level. When grown alone, alfalfa＇s N2 fixation was reduced with N fertilization （R2=0.9376,P=0.0057）. When grown in a mixture with smooth bromegrass, with 75 kg ha-1 of N fertilizer, the percentage of atmospheric N2 fixation contribution to total N in alfalfa （%Ndfa） had a maximum of 84.07 and 83.05% in the 2nd and 3rd harvests, respectively. Nitric oxide, g-aminobutyric acid, and mannose pretreatment influence metabolic profiles in white clover under water stress

Nitric oxide(NO),γ-aminobutyric acid(GABA),and mannose(MAS)could be important regulators of plant growth and adaptation to water *** application of sodium nitroprusside(SNP,a NO donor),GABA,and MAS improved plant growth under watersufficient conditions and effectively mitigated water stress damage to white *** metabonomic analysis showed that both SNP and GABA application resulted in a significant increase in myo-inositol content;the accumulation of mannose was commonly regulated by SNP and MAS;GABA and MAS induced the accumulation of aspartic acid,quinic acid,trehalose,and glycerol under water *** addition,citric acid was uniquely up-regulated by SNP associated with tricarboxylic acid(TCA)cycle under water *** induced the accumulation of GABA,glycine,methionine,and aconitic acid related to GABA shunt,amino acids metabolism,and TCA cycle in response to water *** uniquely enhanced the accumulation of asparagine,galactose,and D-pinitol in association with amino acids and sugars metabolism under water ***-,GABA-,and MAS-induced changes of metabolic profiles and associated metabolic pathways could contribute to enhanced stress tolerance via involvement in the TCA cycle for energy supply,osmotic adjustment,antioxidant defense,and signal transduction for stress defense in white clover.

Klebsiella as an α-tocopherol source facilitating Lactobacillus plantarum fermentation in rice straw silage

Effects of neutral salt and alkali on ion distributions in the roots,shoots,and leaves of two alfalfa cultivars with differing degrees of salt tolerance

The effects of neutral salt and alkali on the ion distribution were investigated in two alfalfa （Medicago sativa L.） cultivars, including Zhongmu 1, a high salt-tolerant cultivar, and Algonquin, a low salt-tolerant cultivar. The alkali stress expressed more serious growth inhibition than the neutral salt stress at the same Na＋ concentration. Compared with Algonquin, Zhongmu 1 did not exhibit a higher alkali tolerance under the Na2CO3-NaHCO3 treatment with the low Na＋ concentration （50 mmol L-l）. The alkali increased the accumulation of Na＋, Ca2＋, and Mg2＋ in the root and changed the Ca2＋ and Mg2＋ balance in the entire alfalfa plant. The salt and alkali stresses decreased the K＋ and Fe3＋ contents of the roots and leaves, the root Mn2＋ content, and the shoot Zn2＋ content, but they increased the Fe3＋ accumulation of the shoots, the shoot and leaf Cu2＋ contents, and the leaf Zn2＋ content in both alfalfa cultivars. Based on the results obtained under the conditions of this experiment, we found that the salt and alkali stresses reduced the plant growth in both alfalfa cultivars, while the alkali caused a stronger stress than the neutral salt in alfalfa. Thus, we conclude that under hydroponic conditions, the deleterious effects of the alkali on plants are due to the distribution change of some trophic ion balance in the roots, shoots, and leaves of the plants by causing of Na＋, CO3^2-, and/or HCO3- stresses.

To efficiently use lignocellulosic materials in ruminants,it is crucial to explore effective enzymes,especially bifunctional *** this study,a novel stable bifunctional cellulase-xylanase protein from buffalo rumen metagenome was expressed and characterized,*** enzyme displayed optimal activity at pH 6.0 and 45℃.The residual endoglucanase and xylanase activities were 90.6%and 86.4%after a 60-min pre-incubation at 55℃.Hydrolysis of rice straw,wheat straw,sheepgrass and sugar beet pulp by CelXyn2 showed its ability to degrade both cellulose and hemicellulose *** with CelXyn2 improved the hydrolysis of agricultural residues with an evident increase in production of total gas,lactate and volatile fatty *** results of 16S rRNA and real-time PCR showed that the effect on in vitro ruminal microbial community depended on fermenta-tion *** study demonstrated that CelXyn2 could strengthen lignocellulose hydrolysis and in vitro ruminal *** characteristics of CelXyn2 distinguish it as a promising candidate for agricultural application.

Two new species of Polyalthiopsis(Annonaceae),*** *** & Bin Yang from Guangxi and yunnan Provinces and *** *** & Bin Yang from yunnan Province,are described and *** is morphologically similar to *** in having narrowly elliptic-oblong,lemon to yellowish green petals,but differs by having obovoid monocarps,a higher number of leaf secondary veins,leaf blades usually widest above the middle,and a lower ratio of leaf blade length to *** is morphologically similar to *** in having axillary inflorescences,1-3(-4) flowers,elliptic leaves,and ellipticovate petals,but differs in the numbers of carpels per flower and ovules per *** molecular phylogenetic analysis using five plastid markers confirm that the two new species belong to the genus Polyalthiopsis and show clear interspecific divergences between *** and *** and between them and other species in the *** descriptions,colored photographs,and habitat and distribution data for the two new species are *** addition,the fruit morphology of *** is described for the first time,based on living *** distributions and a diagnostic key for all Polyalthiopsis species are also presented.

Historical tillage promotes grass-legume mixtures establishment and accelerates soil microbial activity and organic carbon decomposition

Perennial grass-legume mixtures have been extensively used to restore degraded grasslands,increasing grassland productivity and forage *** is crucial for seedbed preparation and sustainable weed management for the establishment of grass-legume ***,a common concern is that intensive tillage may alter soil characteristics,leading to losses in soil organic carbon(SOC).We investigated the plant community composition,SOC,soil microbial biomass carbon(MBC),soil enzyme activities,and soil properties in long-term perennial grass-legume mixtures under two different tillage intensities(once and twice)as well as in a fenced grassland(FG).The establishment of grass-legume mixtures increased plant species diversity and plant community coverage,compared with *** with once tilled grassland(OTG),twice tilled grassland(TTG)enhanced the coverage of high-quality leguminous forage species by 380.3%.Grass-legume mixtures with historical tillage decreased SOC and dissolved organic carbon(DOC)concentrations,whereas soil MBC concentrations in OTG and TTG increased by 16.0%and 16.4%,respectively,compared with *** significantly decreased the activity of N-acetyl-β-D-glucosaminidase(NAG)by 72.3%,whereas soil enzymeβ-glucosidase(βG)in OTG and TTG increased by 55.9%and 27.3%,respectively,compared with *** analysis indicated a close association of the increase in MBC andβG activities with the rapid decline in *** result suggested that MBC was a key driving factor in soil carbon storage dynamics,potentially accelerating soil carbon cycling and facilitating biogeochemical *** establishment of grass-legume mixtures effectively improves forage quality and boosts plant diversity,thereby facilitating the restoration of degraded *** tillage assists in establishing legume-grass mixtures by controlling weeds,it accelerates microbial activity and organic carbon *** findings provide a foundation for understanding the
