Cerebral ischemia is a major health risk that requires preventive approaches in addition to drug *** exercise enhances neurogenesis and sy.aptogenesis,and has been widely.used for functional rehabilitation after *** this study.we determined whether exercise training before disease onset can alleviate the severity.of cerebral *** also examined the role of exercise-induced circulating factors in these *** mice were subjected to 14 day. of treadmill exercise training before surgery.for middle cerebral artery.*** found that this exercise pre-conditioning strategy.effectively.attenuated brain infarct area,inhibited gliogenesis,protected sy.aptic proteins,and improved novel object and spatial memory.*** analysis showed that circulating adiponectin play. a critical role in these preventive effects of *** activation of adiponectin receptors by.Adipo Ron mimicked the effects of exercise,while inhibiting receptor activation abolished the exercise *** summary.our results suggest a crucial role of circulating adiponectin in the effects of exercise pre-conditioning in protecting against cerebral ischemia and supporting the health benefits of exercise.

Allostery.is a fundamental element during channel gating in response to an appropriate stimulus by.which events occurring at one site are transmitted to distal sites to regulate activity. To address how binding of the first Caion at one of the eight chemically.identical subunits facilitates the other Ca-binding events in Mth K, a Ca-gated K+channel containing a conserved ligand-binding RCK domain, we analy.ed a large collection of Mth K structures and performed the corresponding thermody.amic and electrophy.iological measurements. These structural and functional studies led us to conclude that the conformations of the Ca-binding sites alternate between two quaternary.states and exhibit significant differences in Caaffinity. We further propose an allosteric model of the Mth K-gating mechanism by.which a cascade of structural events connect the initial Ca-binding to the final changes of the ring structure that open the ion-conduction pore. This mechanical model reveals the exquisite design that achieves the allosteric gating and could be of general relevance for the action of other ligand-gated ion channels containing the RCK domain.

An auxin‐responsive endogenous peptide regulates root development in Arabidopsis

Auxin play. critical roles in root formation and development. The components involved in this process, however, are not well understood. Here, we newly.identified a peptide encoding gene, auxin-responsive endogenous poly.eptide 1 （AREP1）, which is induced by.auxin, and mediates root development in Arabidopsis. Expression of AREP1 was specific to the coty.edon and to root and shoot meristem tissues. Amounts of AREP1 transcripts and AREP1-green fluorescent protein fusion proteins were elevated in response to indoleacetic acid treatment. Suppression of AREP1 through RNAi silencing resulted in reduction of primary.root length, increase of lateral root number, and expansion of adventitious roots, compared to the observations in wild-ty.e plants in the presence of auxin. By.contrast, transgenic plants overexpressing AREP1 showed enhanced growth of the primary.root under auxin treatment. Additionally. rootmorphology. including lateral root number and adventitious roots, differed greatly.between transgenic and wildty.e plants. Further analysis indicated that the expression of auxin-responsive genes, such as IAA3, IAA7, IAA17, GH3.2, GH3.3, and SAUR-AC1, was significantly.higher in AREP1 RNAi plants, and was slightly.lower in AREP1 overexpressing plants than in wildty.e plants. These results suggest that the novel endogenous peptide AREP1 play. an important role in the process of auxinmediated root development.

Gastric cancer(GC)is one of the most common cancers and ranks the third in cancer mortality.all over the *** goal of this study.was to identify.potential hub-genes,highlighting their functions,signaling pathway.,and candidate drugs for the treatment of GC *** used publicly.available next generation sequencing(NGS)data to identify.differentially.expressed(DE)*** top DE genes were mapped to STRING database to construct the protein-protein interaction(PPI)network and top hub genes were selected for further *** found a total of 1555 DE genes with 870 upregulated and 685 downregulated genes in *** selected the top 400(200 upregulated and 200 downregulated)genes to construct a PPI network and extracted the top 15 hub *** gene ontology.GO)term and ky.to ency.lopedia of genes and genomes(KEGG)pathway.enrichment analy.es of the 15 hub genes exposed some important functions and signaling pathway. that were significantly.associated with GC *** survival analysis of the hub genes disclosed that the lower expressions of the three hub genes CDH2,COL4A1,and COL5A2 were associated with better survival of GC *** three genes might be the candidate biomarkers for the diagnosis and treatment of ***,we considered 3 key.proteins(genomic biomarkers)(COL4A1,CDH2,and CO5A2)as the drug target proteins(receptors),performed their docking analysis with the 102 meta-drug agents,and found Everolimus,Docetaxel,Lanreotide,Venetoclax,Temsirolimus,and Nilotinib as the top ranked 6 candidate drugs with respect to our proposed target proteins for the treatment against GC ***,the proposed drugs might play.vital role for the treatment against GC patients.

Inflammasomes are essential complexes of innate immune sy.tem, which form the first line of host defense against pathogens. Mounting evidence accumulates that inflammasome signaling is highly.correlated with coronavirus disease 2019 (COVID-19). However, there remains a significant gap in our understanding of the regulatory.mechanism of inflammasome signaling. Combining mathematical modeling with experimental analysis of NLRP1b inflammasome signaling, we found that only.the expression levels of caspase-1 and GSDMD have the potential to individually.switch cell death modes. Reduction of caspase-1 or GSDMD switches cell death from py.optosis to apoptosis. Caspase-1 and GSDMD present different thresholds and exert distinct pathway.choices in switching death modes. Py.optosis switches to apoptosis with an extremely.low threshold level of caspase-1, but with a high threshold of GSDMD. Caspase-1-impaired cells employ.ASC-caspase-8-dependent pathway.for apoptosis, while GSDMD-impaired cells primarily.utilize caspase-1-dependent pathway. Additionally. caspase-1 and GSDMD can severally.ignite the cooccurrence of py.optosis and apoptosis. Landscape topography.unravels that the cooccurrence is dramatically.different in caspase-1- and GSDMD-impaired cells. Besides py.optosis state and apoptosis state, a potential new “coexisting” state in single cells is proposed when GSDMD acts as the driving force of the landscape. The “seesaw model” is therefore proposed, which can well describe the death states that are controlled by.caspase-1 or GSDMD in single cells. Our study.sheds new light on NLRP1b inflammasome signaling and uncovers the switching mechanisms among various death modes, providing potential clues to guide the development of more rational control strategies for diseases.

Mollusks are well known for their detailed recording of paleoenvironmental and paleoclimatic changes in their carbonate shells. In this study. we constructed 18-y.ar blue color intensity.and oxy.en isotope profiles of a14C dated （AD 990±40） fossil giant clam,Tridacna gigas, from Shidao Island, South China Sea. Theδ18O profile of theT. gigas specimen display.d regular annual cy.les and was probably.controlled by.seasonal variations of the climatic parameters. The blue color intensity.profile showed good agreement with theδ18O series, and both had 18 clear annual cy.les in accordance with the 18 visually.identified annual growth bands. The annual shell growth rate determined from the blue color intensity.and oxy.en isotope profiles indicated that the annual shell increment of theTridacna specimen was stable after the onset of sexual ma-turity. Spectral analysis of theδ18O and blue color intensity.time series suggested that the El Ni？o-Southern Oscillation （ENSO） period observed in the instrumental temperature and precipitation records of the South China Sea during the past 50 y.ars also existed in medieval times. Our results showed that fossil giant clams could provide a good archive of historical intra-seasonal to decadal climate variations.

The floods caused by.the extreme precipitation in the y.ngtze River basin(y.B)and Murray.Darling River basin(MDRB),the largest basins in China and Australia,have significant impacts on the society.and regional *** on the spatial–temporal analysis of the daily.precipitation extremes(DPEs)during 1982–2016,we found that for both basins,the whole-basin-ty.e DPEs have the highest proportion and a sy.chronous DPE interannual variation characteristic exists in the two basins,with the 3-y. running correlation coefficient of the annual DPE day.(DPEDs)reaching almost 0.7(significant at the 0.01 level).The El Ni?o–Southern Oscillation(ENSO),which is one of the most significant climate disturbance factors in the world,play. an important role in modulating the variability.of the DPEs in the two *** value decomposition(SVD)analysis revealed that both the y.B and the MDRB’s whole-basin-ty.e DPEs are closely.coupled with the procedure that the preceding winter eastern Pacific(EP)-ty.e El Ni?o faded to a central Pacific(CP)-ty.e La *** means that the DPEs in the y.B and MDRB may.sy.chronously.occur more frequently.when the above process *** to the atmosphere–ocean interaction from the east–west dipole sea surface temperature(SST)anomaly.pattern,the atmospheric circulation disturbance exhibits a pattern in which the equatorial eastern Pacific region is a mass source anomaly.with a higher pressure,drier air,and weaker convection,while the equatorial western Pacific region is a mass sink anomaly.with a lower pressure,wetter air,and stronger ***,two wave trains that originated from the tropical western Pacific were found to extend to the y.B and *** interaction between the wave train’s interphase dy.amics and water vapor transport disturbance results in the ascent conditions and enhanced water vapor transport,which leads to the sy.chronous occurrence of DPEs in the y.B and MDRB on an interannual scale.

The effect of reactive element y.trium on purification and carbide precipitation of superalloy.K4169 has been studied through vacuum induction *** results show that y.trium has a strong deoxidizing and desulfurizing ability.and the impurity.elements of sulfur and oxy.en in melting K4169 are greatly.reduced to 2-5 ppm and 4-5 ppm,*** metallurgical reaction product is mainly.y.(2)O_(3)and y.trium containing sulfides have not been *** the increase of the content of y.trium from0.005 wt.%to 0.033 wt.%,the morphology.and size of MC carbides in casting are significantly.influenced due to that the y.trium changes the diffusion of the carbide forming elements and solid fraction during *** the alloy. without y.trium,only.isolated blocky.or strip carbides exist,while skeletonlike carbides are observed in the alloy. with *** the average size and the number density.of skeleton carbides decrease monotonously.with y.trium content *** y.trium increasing to 0.033 wt.%,a phase of hexagonal Ni17 y. is observed to precipitate in the form of sy.biosis with MC carbide and *** addition of y.trium is not beneficial for the properties of K4169 alloy. at the room temperature,which is quite related to the MC carbides with a large skeleton-like morphology.through the analysis of tensile fracture.
