Objective To research the anti-influenza effect of active ingredients in Compound Yizhihao Granule （CYG）. Methods The cytotoxicity and cytopathic effects （CPE） were observed under the phase-contrast microscope, besides 50% toxicity concentration （TC50） and 50% inhibitory concentration （IC50） were also calculated using Reed-Muench method, then the antiviral activity in vitro according to Selection Index （SI = TC50/IC50） was evaluated. In PR8 virus-infected mice, survival time, death rate, and lung index were observed in order to evaluate the protective effect. Besides, the effective ingredients were determined using HPLC method, and their contents were calculated by external standard method. Results CYG could inhibit the influenza virus-induced CPE, with ICso of 4.6 mg/mL （equal to herbal extracts 262.2 μg/mL）, and no direct cytotoxic effect at this concentration. PR8-infected mice were ig given CYG, the lung index and mortality were significantly reduced, and survival time was obviously prolonged. HPLC analysis indicated CYG contained many kinds of antivirus active components, including rupestonic acid, epigoitrin, and adenosine. Conclusion CYG is an effective natural anti-influenza medicine, its antiviral effect should be the synergic effect of a variety of antiviral active ingredients.

OBJECTIVE:To evaluate the effects of Dajizhi(Euphorbium)on selenite-induced cataracts.METHODS:Wistar rat pups were divided into 9 groups.Rats in group 1 were subcutaneously injected with saline,and rats in the other groups were injected with sodium-selenite.Every right eye was treated with 5μL eye drops 3 times per day,and the left eye received no treatment.The eyes of rats in group 3 were treated with pirenoxine;rats in groups 4,5,6,7,8 and 9 were respectively treated with Dajizhi(Euphorbium)(25 mg/m L),Dajizhi(Euphorbium)(5 mg/m L),Dajizhi(Euphorbium)methanol extract(25 mg/m L),Dajizhi(Euphorbium)methanol extract(5 mg/m L),euphol(25 mg/m L),euphol(5 mg/m L).Cataracts were observed by a slit lamp before and after treatment.Electroretinograms were acquired at set intervals.The morphological changes of the rat eyes were observed in vitro,and the levels of glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)in the lenses and aqueous humour were estimated at set intervals.RESULTS:Slit lamp examination showed decreased degrees of cataracts after administration of the different treatments.Morphological comparison showed that Dajizhi(Euphorbium)can reduce the turbidity of the lenses,meaning that Dajizhi(Euphorbium)has the anti-cataract effects.Low-concentration of Dajizhi(Euphorbium),its methanol extract and euphol treatment prevented the b-wave amplitudes of the electroretinograms from falling.Euphorbium treatment significantly restored GSH-Px and SOD levels in the lenses and aqueous humour,especially after 10 and 25 d of administration.Euphorbium may help lenses fight oxidative stress caused by selenite.CONCLUSION:The administration of Dajizhi(Euphorbium)can inhibit selenite-induced cataracts.

Objective:To investigate the immunomodulatory activity of polysaccharides from the roots of Brassica rapa.Methods:The crude polysaccharide from roots of B.rapa(BRP)was extracted and purified to further investigate the active fraction of BRT for inducing macrophage phagocytosis.Results:Effects on RAW264.7 cells demonstrated that BRP behaved better phagocytic capacity and had potent immunomodulatory activity,including increasing production of nitric oxide(NO),tumor necrosis factor a(TNFa)and upregulating mRNA levels of inducible NO synthase(iNOS)and TNFa.Furthermore,modulation of macrophage by BRP was indicated to be mediated via the activation of Akt and nuclear factor-kappa B(NF-κB).Conclusion:The beneficial effects of BRP could be used as an immunotherapeutic adjuvant in treatment of inflammatory diseases.

Objective To investigate the hepatoprotective activities of the extracts from Citrullus colocynthis(ECC),a native plant used as traditional Uigur Medicine on acute liver injury in mice.Methods The activities of ECC of petroleum ether(ECCPE),chloroform(ECCC),ethyl acetate(ECCEA),n-butyl alcohol(ECCBA),and water(ECCW) were evaluated in vivo using two experimental models,carbon tetrachloride(CCl4)- and bacillus calmette-guerin(BCG) plus lipopolysaccharide(LPS)-induced acute hepatotoxicity in mice.The contents of aspartate aminotransferase(AST) and alanine aminotransferase(ALT) in serum were determined and the liver histological examination was carried out,respectively.Results The pretreatment with ECC for 7 d obviously reduced the impact of CCl4toxicity on the serum markers of liver damage,ECCEA and ECCC with a significant difference of AST(P < 0.01,0.05,respectively) and ALT(P < 0.05,0.01,respectively).The protective activity was reconfirmed against BCG + LPS-induced injury and the serum enzymatic levels were obviously elevated,for ECCEA and ECCC with a significant difference of AST(P < 0.05,0.01,respectively) and ALT(P < 0.01,0.05,respectively).Conclusion That ECCEA and ECCC are the potent hepatoprotective extracts that could protect liver against the acute injury,and this ability might be attributed to their hepatoprotective potentials.

Objective To provide experimental evidence for the antihypertensive activity of the flavonoids in flower buds of Coreopsis tinctoria（CT-F）and to investigate the underlying mechanism.Methods The spontaneously hypertensive rats（SHRs）were divided into model,captopril（positive control）,and CT-F groups,and the Wistar-Kyoto rats were set as control group,eight in each group.The blood pressure of SHRs,the activity of angiotensin II（Ang-II）in plasma,nitric oxide（NO）,malondialdehyde（MDA）and thoracic aorta media thickness in SHRs were measured by tail-cuff method,radioimmunity method,nitrate reductase method,thibabituric acid（TBA）method,and the hematoxylin-eosin staining method.Q-PCR analysis was performed to determine the relative quantity of angiotensin-converting enzyme（ACE）,ACEII,angiotensin type 1receptor（AT1R）,and TGF-β1 mRNA in left ventricle.Results CT-F could lower the systolic blood pressure of SHRs dramatically（P〈0.01）.The levels of MDA in serum and Ang II in plasma of SHRs treated with CT-F decreased markedly（P〈0.05,0.01）,the level of NO in serum increased significantly（P〈0.01）.In addition,thoracic aorta media thickness in SHRs treated with CT-F was thinner than that of the model group（P〈0.05）.The mRNA expression of ACE,AT1R,and TGF-β1 in left ventricle was markedly decreased（P〈0.05）,while that of ACE II was increased（P〈0.05）.Conclusion CT-F is effective to lower the blood pressure of SHRs,and its antihypertensive effect is probably associated with lowering the oxidative stress by reducing MDA,ameliorating aorta remodeling,dilating vessel by increasing NO and decreasing Ang-II,and regulating the expression of rennin-angiotensin System-related various genes.

OBJECTIVE:To investigate the efficacy of Yiqigubiao pill(YQGB)on chronic obstructive pulmonary disease(COPD)in rats with the COPD induced by lipopolysaccharide(LPS)and cigarettesmoke fumigation.METHODS:In this study,six groups of rats were set up,including control group,model group,positive control group(aminophylline)and YQGB(high,medium and low doses)groups.Tracheal injection of lipopolysaccharide(LPS)and cigarette-smoke fumigation induced COPD in rats.The general condition,incubation period and coughing times,lung function,level of inflammatory factors,leukocyte condition and pathological changes of bronchus and lung tissue were observed in rats of each group.RESULTS:In the COPD rats,the latent period of coughing was shortened and the cough frequency was increased significantly;the pulmonary function was significantly decreased,which was manifested by the increased lung tissue resistance and respiratory system resistance,and the decreasing percentage of forced expiratory volume and forced expiratory volume in the 0.3 s(FEV0.3/FVC);the contents of tumor necrosis factor-alpha(TNF-α)and interleukin-4 in serum were obviously increased,and the NEUT%in bronchoalveolar lavage fluid was significantly increase.YQGB could obviously prolong the latent period of cough,and reduce the cough frequency and the content of TNF-αin serum.YQGB can also significantly reduce respiratory resistance and increase FEV0.3/FVC value.The results of histopathology showed that YQGB significantly reduced the pathological changes of tracheal mucosa and lung caused by COPD.YQGB obviously increased level of AQP1,which was down-regulated in the COPD rats.CONCLUSION:YQGB could significantly improve the pulmonary function,reduce inflammation and alleviate lung and bronchial diseases in the COPD rats.

An Examination of the Function of Male Flowers in an Andromonoecious Shrub Capparis spinosa

The pollen donor and pollinator attractor hypotheses are explanations for the functions of the male flowers of andromonoecious plants. We tested these two hypotheses in the andromonoecious shrub Capparis spinosa L. （Capparaceae） and confirmed that pollen production and cumulative volume and sugar concentration of nectar do not differ between male and perfect flowers. However, male flowers produced larger anthers, larger pollen grains and smaller ovaries than perfect flowers. Observations on pollinators indicated that two major pollinators （Xylocopa valga Gerst and Proxylocopa sinensis Wu） did not discriminate between flower morphs and that they transferred pollen grains a similar distance. However, there were more seeds per fruit following hand pollination with pollen from male flowers than from perfect flowers. Individuals of C. spinosa with a larger floral display （i.e. bearing more flowers） received more pollen grains on the stigma of perfect flowers. Female reproductive success probably is not limited by pollen. These results indicate that male flowers of C. spinosa save resources for female function and that they primarily serve to attract pollinators as pollen donors.

The microbial transformation of glycyrrhetinic acid（1） by Cunninghamella blakesleana CGMCC 3.970 led to the production of five new metabolites（2-6）.The structures of the metabolites were determined by extensive spectroscopic（HR-ESIMS,1D and 2D NMR） data analyses.The involved reactions exhibited specific hydroxylations at C-24,C-7,and C-15,and oxidation at C-3.Moreover,compounds 2,5,and 6showed significant neural anti-inflammatory activity by inhibiting lipopolysaccharide-induced NO production in mouse microglia BV2 cells with IC（50） values of 0.76,0.94,and 0.16μmol/L,respectively.

Objective: A sensitive HPLC-DAD detection method was established for the comparative analysis of the four active compounds(including baicalin, baicalein, wogonoside and wogonin) of Baikal Skullcap and its classical TCM prescriptions according to different clinical curative effects. And analyze the relationship between compatibility of medicines, content and clinical curative effect.Methods: Water extracts were analyzed by high-performance liquid chromatography-diode array detection(HPLC-DAD), using an Agilent extend-C_(18)(4.6 mm×250 mm,5μm) column by using solvent A(0.2% phosphoric acid water) and solvent B(methanol) in the ratio of 52:48(v/v) at a flow rate of 1 ml/min. The column temperature was maintained at 25°C and the detection wavelength was set at 280 nm.Results: In quantitative analysis, the four selected markers showed good regression(R^2>0.999) within test ranges. The average recoveries were between 99.75~100.60% and their RSD values were between 0.97%~1.71%. The proportions of the four chemical compounds for different efficacy were 33.51~35.76: 6.78~6.87: 3.45~3.64: 1; 18.6~23.64: 3.63~4.27: 1.36~1.53: 1; 29.94~31.78: 5.14~5.23: 3.28~3.56: 1; 33.99~41.91:5.83~7.42: 2.83~3.39: 1.Conclusion: Clinical practice of thousands of years have proved that the curative effect of Traditional Chinese Medicine depends on the compatibility of different kinds of medicine material crude slices. According to this study we found that different compatibility of medicines leads to different proportions of the chemical compounds. We assume that there is a certain correlation between therapeutical effect of TCM and compatibility of medicines.
