Background-Occupational medicamentosa-like dermatitis induced by trichloroethylene(OMLDT) is an autoimmune disease and it has become a serious occupational health *** aim of this study was to develop potential serum biomarkers for OMLDT using a fast and efficient method of high-performance liquid chromatography(HPLC) coupled to MALDI-TOF/TOF-MS. Methods-We collected fasting blood samples from patients with OMLDT(n = 18) and healthy volunteers(n = 33) to explore serum peptidome *** proteins were purified with magnetic bead-based weak cation exchange chromatography and analyzed with *** and supervised neural network(SNN) algorithms were used on the training and testing data sets to establish classification *** protein/peptide peaks were identified by high mass accuracy measurements combined with top-down tandem mass spectrometry. Results-Thirty protein/peptide peaks were significantly different between the two groups and were selected as potential biomarkers of OMLDT *** SNN algorithm utilized three peaks(m/z 2106.3,2134.5,and 3263.67 Da) to separate the OMLDT patients from the healthy controls,with a cross validation of 94.44%and a recognition capability of 100%.The test sensitivity was 95.5%,and the test specificity was 73.8%.HPLC-TOF/TOF-MS identified two proteins that may play a role in the pathology of OMLDT:ATP-binding cassette transporter,and a fragment of albumin. Conclusion-Specific proteomic fingerprints in the sera of OMLDT patients could be used to identify OMLDT cases with high specificity and *** HPLC-TOF/TOF-MS approach will be of great value for exploring potential biomarkers and clinical studies of OMLDT.

观察新风胶囊对类风湿关节炎(RA)B淋巴细胞、炎性介质、FAK/CAPN/PI3K通路的影响,探讨新风胶囊改善RA临床症状的机制。观察RA患者关节及全身症状,检测实验室指标[血红蛋白(HGB)、血小板计数(PLT)、血沉(ESR)、免疫球蛋白(Ig)G、IgA、IgM、类风湿因子(RF)、抗环瓜氨酸抗体(CCP-AB)、C反应蛋白(CRP)]。采用酶联免疫吸附法检测血清白细胞介素(IL)-1β、IL-10、IL-33、趋化因子5(CCL5)、血管内皮生长因子(VEGF),流式细胞术测定外周血CD3^(-)CD19^(+)B细胞,免疫印迹法检测FAK、p-FAK、CAPN、PI3K蛋白。结果显示,新风胶囊能明显改善RA关节及全身症状,提高临床疗效。且新风胶囊能升高HGB,降低PLT、CCP-AB、CRP、ESR指标,上调IL-10表达,下调IL-1β、IL-33、CCL5、VEGF、CD3^(-)CD19^(+)B细胞、FAK、p-FAK、CAPN、PI3K表达(P<0.01)。结果表明,新风胶囊可能通过下调CD3^(-)CD19^(+)B细胞表达,调控炎性细胞因子和趋化因子平衡,抑制FAK/CAPN/PI3K通路异常激活,改善RA临床症状。

Countermeasures for China's Ecological Environment Construction in Land Reclamation

Basic situation of the current ecological environment construction in land consolidation and rehabilitation of China is introduced. The related advanced experience abroad is summarized from mainly four aspects, namely policy, system as well as subject development, planning guidance, design method of project planning and engineering study, and practical countermeasures to China's ecological environment construction in land reclamation are proposed. In terms of policy, system and subject, relevant regulations, policies and system construction should be strengthened and the framework of policy, system and subject development in ecological landscape construction of China's land reclamation should be proposed, integrating ecological environment construction into land reclamation. At the level of strategic planning, the spatial layout and construction key points of green infrastructure in land reclamation of "urban multi-functional developing region, eastern economic developed region, central modern agricultural region and northwestern ecological fragile region" should be achieved. At the level of project planning design, ecological principles, sustainable landscape design principles and vernacular landscape design methods are to be integrated into the planning procedure to form operable technique regulations or introductions. At the level of engineering design, engineering technique system, standards and regulations of ditches, roads, forests and channels with local distinctions should be formulated according to the characters and strategic demands of urbanization, industrialization, agriculture modernization and ecological environment preservation in different regions.

Overall Estimation of Anti-Oxidant Activity by Mammal Macrophage

Super oxide anion had been known as a critical role in a phagocytic cell when microorganisms invade inside. However, this radical seldom need for a clean environment such in a developed country. Moreover, this agent sometimes triggered as serious factor in the site of inflammation. There are many papers which concerned newly found anti-oxidative substances. However, many reports only focused on the molecular basis in vitro, how oxidative stress damaged the human tissue. The key factor for oxidative stress needs to be discussed first for immunological standpoint, such as armed reduction treaty. In other words, minimum armed force is necessary for an invasion by outer or inner irregular cells. On the basis of phagocytic function of the cell in vivo, we tried to access anti-oxidative activity by employing peritoneal murine macrophage and human monocyte. So, the purpose of this study is to establish a conventional but tailor made system for which is useful for evaluating the anti-oxidative activity to different constition. Especially in human peripheral monocyte, the sample was tested each constitution such in granulocyte rich type and lymphocyte rich typ. In order to establish the direct estimation by phagocytic cell, a murine peritoneal exudates cell suspension was obtained after one week of oral administration with anti-oxidative sample. The samples we selected were traditional Chinese medicine, Shi-Quan-Da-Bu-Tang (SDT), Bu-Zong-Ye-Qi-Tang (BYT) and Shao-Chin-Rong-Tang sCRT), Agaricus, Chlorella, and Propolis. The amount of generated super oxide anion was calculated through measuring the optical density change of this supernatant using cytochrome-C which was reduced by generated super oxide anion in the phagocytic cell. The generated super oxide anion after administration of Agaricus, Chlorella, SDT, BYT, sCRT and Propolis were 2.64, 1.95, 1.24, 1.25, 2.88, 2.55 × 10﹣5 mmol/ml, respectively, whereas 2.85 × 10﹣5 mmol/ml in control group. The microenvironment of hypoxia and immune-cold limits the therapeutic outcomes of immune checkpoint blockade(ICB)therapy in solid *** is important and imperative to search new strategies to relieve tumor hypoxia and reverse immunosuppression of cold *** this study,the oxygen(O_(2))self-replenishing nano-enabled coordination platform can be used to induce potent antitumor immune response in cold *** nanoplatform can produce O_(2)by catalyzing hydrogen peroxide(H_(2)O_(2))in tumor site effectively,showing excellent photodynamic therapy(PDT)***,it can further trigger immunogenic cell death(ICD),enhance T cell infiltration,reverse immunosuppression,and reprogram the immune-cold tumor *** vitro and in vivo results demonstrate that the nanoplatform has potential for eradicating tumors and long-term immunological memory *** nanoplatform opens up a strategy for reprograming the immunosuppressive microenvironment in cold tumors.

Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2) can induce acute inflammatory response like acute lung inflammation(ALI) or acute respiratory distress syndrome,leading to severe progression and *** for treatment of SARS-CoV-2-triggered respiratory inflammation are urgent to be *** previous study shows that Salvianolic acid C potently inhibits SARS-CoV-2 *** this study,we investigated the antiviral effects of a Salvia miltiorrhiza compound,Danshensu,in vitro and in vivo,including the mechanism of S protein-mediated virus attachment and entry into target *** authentic and pseudo-typed virus assays in vitro,Danshensu displayed a potent antiviral activity against SARS-CoV-2 with EC50 of 0.97 μM,and potently inhibited the entry of SARS-CoV-2 S protein-pseudo-typed virus(SARS-Co V-2 S) into ACE2-overexpressed HEK-293 T cells(IC50 = 0.31 μM) and Vero-E6 cell(IC50 = 4.97 μM).Mice received SARS-CoV-2 S via trachea to induce ALI,while the VSV-G treated mice served as *** mice were administered Danshensu(25,50,100 mg/kg,i.v.,once) or Danshensu(25,50,100 mg·kg-1·d-1,oral administration,for 7 days) before SARS-CoV-2 S *** showed that SARS-CoV-2 S infection induced severe inflammatory cell infiltration,severely damaged lung tissue structure,highly expressed levels of inflammatory cytokines,and activated TLR4 and hyperphosphorylation of the NF-κB p65;the high expression of angiotensinogen(AGT) and low expression of ACE2 at the mRNA level in the lung tissue were also *** oral and intravenous pretreatment with Danshensu dose-dependently alleviated the pathological alterations in mice infected with SARS-CoV-2 *** study not only establishes a mouse model of pseudo-typed SARS-CoV-2(SARS-CoV-2 S) induced ALI,but also demonstrates that Danshensu is a potential treatment for COVID-19 patients to inhibit the lung inflammatory response.

证据分级水平是进行临床决策的重要考虑因素。中医药领域完全照搬国际常用证据评价体系存在一定局限性,故提出中医临床证据分级思路,即在遵循国际证据分级标准基础上,充分考虑中医药临床实践特点,以GRADE(Grades of Recommendation,Assessment,Development,and Evaluation)证据评价系统为基础,结合中医古籍证据及现代名医经验证据分级标准,进而整合形成中医干预类临床证据分级系统,其中古籍证据从医案来源、诊疗内容的全面性和传承性三方面进行评价,现代名医经验证据根据名医的资质、经验内容完整性、经验传承性进行评价。针对中医古籍、现代名医经验及现代研究多来源证据的整合,以定性综合为主,多来源证据的分级水平与现代研究GRADE证据评价保持一致,而中医古籍及现代名医经验作为辅助证据呈现,以期更加客观、全面地评价中医临床证据,进而指导临床实践。
