Background:Bendamustine was approved in china on May 26th,2019 by the National Medical Product Administration for the treatment of indolent B-cell non-Hodgkin lymphoma(NHL).The current study was the registration trial and the first reported evaluation of the efficacy,safety,and pharmacokinetics of bendamustine in chinese adult patients with indolent B-cell NHL following relapse after chemotherapy and rituximab ***:This was a prospective,multicenter,open-label,single-arm,phase 3 study(NcT01596621;c18083/3076)with a 2-year follow-up *** patients received bendamustine hydrochloride 120 mg/m2 infused intravenously on days 1 and 2 of each 21-day treatment cycle for at least six planned cycles(and up to eight cycles).The primary endpoint was the overall response rate(ORR);and secondary endpoints were duration of response(DoR),progression-free survival(PFS),safety,and *** were classified according to their best overall response after initiation of *** of patients in each response category(complete response[cR],partial response[PR],stable disease,or progressive disease)were summarized along with a twosided binomial exact 95%confidence intervals(cIs)for the ***:A total of 102 patients were enrolled from 20 centers between August 6th,2012,and June 18th,*** the time of the primary analysis,the ORR was 73%(95%cI:63%–81%)per Independent Review committee(IRc)including 19%cR and 54%*** the follow-up period,the median DoR was 16.2 months by IRc and 13.4 months by investigator assessment;the median PFS was 18.6 months and 15.3 months,*** most common non-hematologic adverse events(AEs)were gastrointestinal toxicity,pyrexia,and *** 3/4 neutropenia was reported in 76%of *** AEs were reported in 29 patients and five patients died during the *** analysis indicated that the characteristics of bendamustine and its metabolites M3 and M4 were generally consistent

Synthesis of aminoalkylsilanes with oligo(ethylene oxide) unit as multifunctional electrolyte additives for lithium-ion batteries

Aminoalkylsilanes with oligo(ethylene oxide) units were designed and synthesized as multifunctional electrolyte additives for lithium-ion batteries. The chemical structures were fully characterized by nuclear magnetic resonance (NMR) spectroscopy and their thermal properties, viscosities, electrochemical windows, and ionic conductivities were systematically measured. With adding one of these compounds (1 vol. %, DSc3N1) in the baseline electrolyte 1.0 M LiPF 6 in Ec: DEc (1:1, in volume), Li/LicoO 2 half cell tests showed an improved cyclability after 100 cycles and improved rate capability at 5c rate condition. Electrochemical impedance spectroscopy (EIS), X-ray photoelectron spectroscopy (XPS), and energy dispersive spectroscopic (EDS) analysis confirmed the acid scavenging function and film forming capability of DSc3N1. These results demonstrated that the multifunctional organosilicon compounds have considerable potential as additives for use in lithium-ion batteries.

cytotoxicity and autophagy induction by graphene quantum dots with different functional groups

Graphene quantum dots(GQDs) possess great potential in various applications due to their superior physicochemical properties and wide array of available surface ***, the toxicity of GQDs has not been systematically assessed, thus hindered their further development; especially, the risk of surface modifications of GQDs is largely unknown. In this study, we employed a lung carcinoma A549 cells as the model to investigate the cytotoxicity and autophagy induction of three types GQDs, including cGQDs(cOOH-GQDs), hGQDs(OH-GQDs), and aGQDs(NH_2-GQDs). The results showed hGQDs was the most toxic, as significant cell death was induced at the concentration of 100 μg/mL,determining by WST-1 assay as well as Annexin-V-FITc/PI apoptosis analysis, whereas cGQDs and aGQDs were non-cytotoxic within the measured concentration. Autophagy detection was performed by TEM examination, Lc3 fluorescence tracking, and westernblot. Both aGQDs and hGQDs induced cellular autophagy to various degrees except for cGQDs. Further analysis on autophagy pathways indicated all GQDs significantly activated p-p38 MAPK; p-ERK1/2 was inhibited by aGQDs and hGQDs but activated by c GQDs. p-JNK was inhibited by aGQDs and c GQDs, while activated by hGQDs. Simultaneously, Akt was activated by hGQDs but inhibited by aGQDs. Inhibition of autophagy by 3-MA significantly increased the cytotoxicity of GQDs, suggesting that autophagy played a protective role against the toxicity of GQDs. In conclusion, c GQDs showed excellent biocompatibility and may be considered for biological applications. Autophagy induction may be included in the health risk assessment of GQDs as it reflects the stress status which may eventually lead to diseases.

Highly Sensitive and Specific Monoclonal Antibody-Based Serological Methods for Rice Ragged Stunt Virus Detection in Rice Plants and Rice Brown Planthopper Vectors

Rice ragged stunt virus（RRSV） is a serious rice disease in Asia, causing serious yield losses on rice. The capsid protein（cP） gene of the major outer capsid protein of RRSV was expressed in Escherichia coli BL21（DE3） using the pMAL-c2 X expression vector. The recombinant protein was used as the immunogen to immunize BALB/c mice. A hybridoma cell line 8A12 secreting monoclonal antibody（MAb） against RRSV was obtained by fusing mouse myeloma cells（Sp 2/0） with spleen cells from the immunized BALB/c mice. western blot analysis showed that the MAb 8A12 can specifically react with RRSV cP. Using the MAb, an antigen-coated-plate enzyme-linked immunosorbent assay（AcP-ELISA）, a dot enzyme-linked immunosorbent assay（dot-ELISA）, and immunocapture-RT-PcR（Ic-RT-PcR） assay were developed to detect RRSV. The established AcP-ELISA, dot-blot ELISA and Ic-RT-PcR methods could detect RRSV in infected rice tissue crude extracts with dilutions of 1：40 960, 1：1 280 and 1：655 360（w/v, g mL-1）, respectively. The AcP-ELISA and dot-blot ELISA methods could detect RRSV in infected insect vector crude extracts with dilutions of 1：12 800 and 1：1 600（an individual planthopper μL-1）, respectively. The field survey revealed that Rice ragged stunt disease occurs on rice in Hainan, Yunnan, Guangxi, Sichuan, Guizhou, Fujian, Hunan, Jiangxi and Zhejiang in china.

Solution of the effects of twinning in femtosecond X-ray protein nanocrystallography

With the development of the XFEL （X-ray free electron laser）, high quality diffraction patterns from nanocrystals have been achieved. The nanocrystals with different sizes and random orientations are injected to the XFEL beams and the diffraction patterns can be obtained by the so-called ＂diffraction-and-destruction＂ mode. The recovery of orientations is one of the most critical steps in reconstructing the 3D structure of nanocrystals. There is already an approach to solve the orientation problem by using the automated indexing software in crystallography. However, this method cannot distinguish the twin orientations in the cases of the symmetries of Bravais lattices higher than the point groups. Here we propose a new method to solve this problem. The shape transforms of nanocrystals can be determined from all of the intensities around the diffraction spots, and then Fourier transformation of a single crystal cell is obtained. The actual orientations of the patterns can be solved by comparing the values of the Fourier transformations of the crystal cell on the intersections of all patterns. This so-called ＂multiple-common-line＂ method can distinguish the twin orientations in the XFEL diffraction patterns successfully.

Relationship of cell compositions in AIIografts with Outcomes after Haploidentical Transplantation for Acquired Severe Aplastic Anemia： Effects of cD34＋ and cD14＋ cell Doses

Background： The dose of certain cell types in allografts affects engraftment kinetics and clinical outcomes after allogeneic stem cell transplantation （ScT）. Hence, the present study investigated the association of cell compositions in allografts with outcomes after unmanipulated haploidentical ScT （haplo-ScT） for patients with acquired severe aplastic anemia （SAA）. Methods： A total of 131 patients with SAA who underwent haplo-ScT were retrospectively enrolled. cell subsets in allografts were determined using flow cytometry. To analyze the association of cellular compositions and outcomes, Mann-Whitney U nonparametric tests were conducted for patient age, sex, weight, human leukocyte antigen mismatched loci, ABO-matched status, patient ABO blood type, donor-recipient sex match, donor-recipient relationship, and each graft component. Multivariate analysis was performed using logistic regression to determine independent influence factors involving dichotomous variables selected from the univariate analysis. Results： A total of 126 patients （97.7%） achieved neutrophil engraftment, and 121 patients （95.7%） achieved platelet engraftment. At 100 days after transplantation, the cumulative incidence of II-IV acute graft-versus-host disease （GVHD） was 32.6%. After a median follow-up of 842 （range： 124-4110） days for surviving patients, the cumulative incidence of total chronic GVHD at 3 years after transplantation was 33.7%. The probability of overall survival at 3 years was 83.0%. Multivariate analysis showed that higher total doses of cD14＋（P = 0.018） and cD34＋ cells （P 〈 0.001） were associated with a successful platelet engraftment. A successthl platelet was associated with superior survival （P 〈 0.001）. Increased cD133^+ cell infiltration in the rat brain following fluid percussion injury

The prominin-1/cD133 epitope is expressed in craniocerebral trauma in animal models of fluid undifferentiated cells. Studies have reported that percussion injury induces production of a specific stem cell subgroup. It has been hypothesized that fluid percussion injury induces cD133＋ cell infiltration in the brain tissue. The present study established a traumatic brain injury model through fluid percussion injury. Immunohistochemical staining showed significantly increased cD133 antigen expression in the rat brain following injury. cD133＋ cells were mainly distributed in hippocampal cA1 3 regions, as well as the dentate gyrus and hilus, of the lesioned hemisphere. Occasional cells were also detected in the cortex. In addition, reverse transcription-PcR revealed that no change in cD133 mRNA expression in injured brain tissue. These results suggested that fluid percussion injury induced cD133 antigen expression in the brain tissues as a result of conformational epitope changes, but not transcriptional expression.
