Toll-like receptors(TLRs)play important roles in immune responses against pathogens and ***,TLR8 has gained attention because of its association with multiple inflammatory diseases,infections and antitumor ***8 senses the degradation products of single-stranded RNA from microbes and self-released RNA to induce type I interferons,inflammatory gene expression and nucleotide-binding and oligomerization domain–,leucine-rich repeat–and pyrin domain–containing protein 3(NLRP3)inflammasome *** far,the understanding of TLR8 function in vivo is still limited,partially because of lacking a reliable rodent animal *** Tlr8 cannot sense the ligands of human *** mammals,TLR8 distinguishes live bacteria from dead bacteria to regulate the magnitude of immune ***,TLR8 has been reported to recognize severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)RNA to induce inflammatory responses,suggesting that TLR8 participates in coronavirus disease 2019(COVID-19).In this review,we discuss the mechanism of ligand recognition by TLR8,TLR8-mediated signaling pathways and signaling crosstalk between TLR8 and other molecules,and untangle the contribution of TLR8 to inflammatory diseases,infectious diseases,antitumor immunity and vaccination.

By modulating hepcidin production, an organism controls intestinal iron absorption, iron uptake and mobilization from stores to meet body iron need. In recent years there has been important advancement in our knowledge of hepcidin regulation that also has implications for understanding the physiopathology of some human disorders. Since the discovery of hepcidin and the demonstration of its pivotal role in iron homeostasis, there has been a substantial interest in developing a reliable assay of the hormone in biological fluids. Measurement of hepcidin in biological fluids can improve our understanding of iron diseases and be a useful tool for diagnosis and clinical management of these disorders. We reviewed the literature and our own research on hepcidin to give an updated status of the situation in this rapidly evolving field.

Objective To establish a comprehensive analytical method based on SPE‐UPLC‐MS for the simultaneous determination of bisphenol A （BPA）, nonylphenol （NP）, and octylphenol （OP） in urine samples. Methods Sixty urine samples collected from healthy subjects were analyzed for BPA, NP, and OP concentrations. The samples were de‐conjugated by adding β‐glucuronidase and sulfatase. After the enzymatic treatment, the samples were subjected to the OASIS HLB column solid phase extraction cartridges so as to be cleaned and concentrated. The UPLC separation was performed on a Acquity UPLCTM BEH C18 column （2.1×100 mm, 1.7 μm） with a gradient elution system of methanol‐water as the mobile phase. Triple‐quadrupole mass spectrometry analyzer was used for the qualitative and quantitative analysis of UPLC‐MS/MS system. Results The limit of detection of BPA, NP, and OP was 0.10, 0.10, and 0.15 ng/mL, respectively. The recoveries of BPA, NP and OP were 80.1%‐108%, 81.3%‐109%, and 81.5%‐98.7%, respectively. Among the 60 urine samples, BPA was detected in 8 samples at the level of 0.297‐32.7ng/mL, NP was detected in 29 samples at the level of 1.69‐27.8 ng/mL, and OP was detected in 17 samples at the level of 0.407‐11.1 ng/mL. Conclusion The method is simple with high sensitivity and selectivity, and is suitable for the determination of BPA, NP, and OP in urine. As shown by our analysis , BPA, NP, and OP appear to be prevalent in human urine. This is particularly true for NP. The results from our study is therefore valuable for future studies to assess the exposure to BPA, NP, and OP in the general population.

Sperm banking can preserve male fertility effectively, but the current conditions of sperm cryopreservation in China have not been investigated. This retrospective investigation was based on data collected at multiple centres in China from January 2003 to December 2008. The collected data included urogenital history, indication for cryopreservation, semen parameters, use rate, type of assisted reproductive technique （ART） treatment and pregnancy outcome. The study population included 1 548 males who had banked their semen during the study period at one of the clinics indicated above. Approximately 1.9% （30/1 548） of the cryopreserved semen samples were collected from cancer patients; about 88.8% （1 374/1 548） of the patients had banked their semen for ART and 8.6% （134/1 548） had a male infertility disease （such as anejaculation, severe oligozoospermia and obstructive azoospermia）. The total use rate of cryopreserved semen was 22.7% （352/1 548）, with 119 live births. The cancer group use rate was 6.7% （2/30）, with one live birth by intracytoplasmic single sperm injection （ICSI）. The ART group use rate was 23.2% （319/1 374）, with 106 live births. The reproductive disease group use rate was 23.1% （31/134）, with 12 live births. The semen parameters in each category varied; the cancer patient and infertility disease groups had poor semen quality. In vitro fertilization （IVF） and ICSI were the most common ART treatments for cryopreserved sperm. Semen cryopreservation as a salvage method is effective, but in many conditions it is underutilized, especially in cancer patients. Lack of awareness, urgency of cancer treatment and financial constraints are the main causes of the low access rate. The concept of fertility preservation should be popularized to make better use of this medical service in China.

Background： Successful antiretroviral therapy （ART） has been demonstrated to be effective in reducing the infectivity of human immunodeficiency virus （HIV）. We conducted a study to predict the potential effect of ART on the spread of HIV in Chaoyang District, Beijing, China, using the Asian Epidemic Model （AEM）. Methods： The AEM baseline workbook was used to determine the current infection status and to project the future spread of HIV under current conditions. We changed the input on the ART coverage from 2014 to 2025 and also modified the treatment eligibility in the AEM intervention workbook, in order to allow for analysis of the projected downstream impact of ART. Results： By gradually increasing the ART coverage rate from 29.7% （rate of 2013） to 40.0%, 50.0%, 60.0%, 70.0%, 80.0%, and 90.0% （at CD4＋ ≤350 cells/μl）, and by changing the dates of coverage from 2014 to 2020, the number of new infections showed a cumulative decline of 0.60%, 1.59%, 2.94%, 5.33%, 9.32%, and 14.98%, respectively. After 2020, the projected rates of infection rebounded slightly, so with the exception of the years with very high coverage （90.0%）, new infections continued to decrease. When we changed the initial threshold of therapy to CD4＋ cell counts ≤500 cells/μl, new infections decreased 6.00%, 11.64%, 15.92%, 21.11%, 26.92%, 33.05%, and 38.75%, respectively, under varying ART coverages. Conclusion： Our study demonstrates that the early initiation of ART for people living with HIV/acquired immune deficiency syndrome （AIDS） has a positive effect in slowing the spread of HIV.

Investigation outer environment pollution for avian influenza virus from live poultry markets in Guangdong Province during 2012-2015 to discover potential human avian influenza incidence of risk factors and to provide scientific reference for prevention and treatment of human avian influenza virus. Use information management system data about Infectious disease monitoring platform and descriptive epidemiology,and analysis epidemiological features of various subtypes of virus from live poultry markets and investigate outer environment pollution for avian influenza virus from live poultry markets in Guangdong Province during 2012-2015. A total of 33079 environmental samples were collected and tested from 2012 to 2015 in twenty-one cities,and the overall positive rate of avian influenza A virus was 24.23%. the positive rate of highly pathogenic H5、H7 and H9 viruses was 3.70%、3.89% and 13.53% respectively. Apart from the positive rate with the seasonal increase in 2012,the peak of Flu A positive rate of emergence was in the spring and winter. Collecting different parts or site specimens,slaughter poultry chopping board surface was the highest positive rate(Flu A 39.49%,H5 8.41%,H7 7.41%,H9 23.84%),while feces specimens was the lowest positive rate(Flu A 14.99%,H5 1.73%,H7 2.38%,and H9 7.23%).Collecting specimens of related species,chicken(64.08%),duck(55.84%) and bird(51.92%),the positive rate of avian influenza virus is all above 50% and H5,H7 and H9 all can be checked out from related species. Also found that more regions distribution of H7 subtypes in the environment and the number of cases of corresponding regions H7 N9 human infection have significant correlation(r = 0.689,P < 0.689). Selecting 2322 samples to detect nucleic acid of H6 subtype,the total positive rate was 2.58%,at the same time,selecting 153 specimens about H5,H6 and H9 subtype to detect the N subtype,H5N1,H5N2,H5N6,H6N2 and H9N2 subtype scan be checked out. Dementia is a comprehensive category of brain diseases that is great enough to affect a person＇s daily functioning. The most common type of dementia is Alzheimer＇s disease, which makes most of cases. New researches indicate that gastrointestinal tract microbiota are directly linked to dementia pathogenesis through triggering metabolic diseases and low-grade inflammation progress. A novel strategy is proposed for the management of these disorders and as an adjuvant for psychiatric treatment of dementia and other related diseases through modulation of the microbiota （e.g. with the use of probiotics）.
