Microglia are resident macrophage cells in the central nervous system that search for pathogens or abnormal neural activities and migrate to resolve the *** effective search and targeted motion of macrophages mean dearly to maintaining a healthy brain,yet little is known about their migration *** this work,we study microglial motion with and without the presence of external *** discover that the cells are promptly attracted by the applied forces(i.e.,mechanotaxis),which is a tactic behavior as yet unconfirmed in ***,in both the explorative and the targeted migration,microglia display dynamics that is strikingly analogous to bacterial run-and-tumble motion.A closer examination reveals that microglial run-and-tumble is more sophisticated,e.g.,they display a short-term memory when tumbling and rely on active steering during runs to achieve mechanotaxis,probably via the responses of mechanosensitive ion *** differences reflect the sharp contrast between microglia and bacteria cells(eukaryotes ***)and their environments(compact tissue ***).Further analyses suggest that the reported migration dynamics has an optimal search efficiency and is shared among a subset of immune cells(human monocyte and macrophage).This work reveals a fruitful analogy between the locomotion of 2 remote systems and provides a framework for studying immune cells exploring complex environments.

Wound healing and tissue repair are recognized as basic human health problems *** to accelerate the reparative process are focused on developing functional wound ***,we present novel Janus textiles with anisotropic wettability from hierarchical microfluidic spinning for wound *** hydrophilic hydrogel microfibers from microfluidics are woven into textiles for freeze-drying treatment,followed by the deposition of electrostatic spinning nanofibers composed of hydrophobic polylactic acid(PLA)and silver *** electrospun nanofiber layer can be well coupled with the hydrogel microfiber layer to generate Janus textiles with anisotropic wettability due to the roughness of the hydrogel textile surface and the incomplete evaporation of PLA solution when reaching the *** wound treatment with the hydrophobic PLA side contacting the wound surface,the wound exudate can be pumped from the hydrophobic to the hydrophilic side based on the wettability differential derived drainage *** this process,the hydrophobic side of the Janus textile can prevent excess fluid from infiltrating the wound again,preventing excessive moisture and preserving the breathability of the *** addition,the silver nanoparticles contained in the hydrophobic nanofibers could impart the textiles with good antibacterial effect,which further promote the wound healing *** features indicate that the described Janus fiber textile has great application potential in the field of wound treatment.

Immune-mediated liver injury (ILI) is a condition where an aberrant immune response due to various triggers causes the destruction of hepatocytes. Fibroblast growth factor 4 (FGF4) was recently identified as a hepatoprotective cytokine;however, its role in ILI remains unclear. In patients with autoimmune hepatitis (type of ILI) and mouse models of concanavalin A (ConA)- or S-100-induced ILI, we observed a biphasic pattern in hepatic FGF4 expression, characterized by an initial increase followed by a return to basal levels. Hepatic FGF4 deficiency activated the mitochondria-associated intrinsic apoptotic pathway, aggravating hepatocellular apoptosis. This led to intrahepatic immune hyper-reactivity, inflammation accentuation, and subsequent liver injury in both ILI models. Conversely, administration of recombinant FGF4 reduced hepatocellular apoptosis and rectified immune imbalance, thereby mitigating liver damage. The beneficial effects of FGF4 were mediated by hepatocellular FGF receptor 4, which activated the Ca^(2+)/calmodulin-dependent protein kinasekinase 2 (CaMKKβ) and its downstream phosphatase and tensin homologue-induced putative kinase 1 (PINK1)-dependent B-cell lymphoma 2-like protein 1-isoform L (Bcl-XL) signalling axis in the mitochondria. Hence, FGF4 serves as an early response factor and plays a protective role against ILI, suggesting a therapeutic potential of FGF4 and its analogue for treating clinical immune disorder-related liver injuries.

Diagnosing individuals with autism spectrum disorder(ASD)accurately faces great chal-lenges in clinical practice,primarily due to the data's high heterogeneity and limited sample *** tackle this issue,the authors constructed a deep graph convolutional network(GCN)based on variable multi‐graph and multimodal data(VMM‐DGCN)for ASD ***,the functional connectivity matrix was constructed to extract primary ***,the authors constructed a variable multi‐graph construction strategy to capture the multi‐scale feature representations of each subject by utilising convolutional filters with varying kernel ***,the authors brought the non‐imaging in-formation into the feature representation at each scale and constructed multiple population graphs based on multimodal data by fully considering the correlation between *** extracting the deeper features of population graphs using the deep GCN(DeepGCN),the authors fused the node features of multiple subgraphs to perform node classification tasks for typical control and ASD *** proposed algorithm was evaluated on the Autism Brain Imaging Data Exchange I(ABIDE I)dataset,achieving an accuracy of 91.62%and an area under the curve value of 95.74%.These results demon-strated its outstanding performance compared to other ASD diagnostic algorithms.
