[Objective] To study the prokaryotic expression and antigenicity identification of S1 gene from avian infectious bronchitis virus （IBV）. [Method] The S1 gene was cloned into a pMD18-T vector to yield a recombinant plasmids pMD18-T-IBV-S1. Then S1 gene was inserted into the multiple cloning site of a prokaryotic expression vector pET-32a （＋）. The recombinant plasmid was transformed into E. coil BL21. The recombinant protein was induced by IPTG and measured by SDS-PAGE and western-blotting. [Result] The S1 gene was successfully expressed in E. coil BL21, the fusion proteins were about 66.0 kDa in a form of inclusion body. Western-blotting test showed that the recombinant proteins could be identified by IBV polyclonal antibody. [ Conclusion] The recombinant proteins of S1 gene have the antigenicity, which lays a good foundation for further research on new generation vaccine of IBV.

Infectious Bronchitis (IB) is highly contagious disease of commercial poultry causing substantial economic loses by producing poor quality meat in broilers and effecting production in breeder birds. The causative agent has been reported as most hazardous pathogen among other infectious agent even after being immunized with multi-variant strain vaccine. Currently, different strain such as H-120, 4/91 and D274 have been used extensively for immunoprophylaxis against velogenic strain across Pakistan with minimal protection reported. In current study PCR analysis was used to investigate the molecular nature of IB isolates from Punjab and Sind province of Pakistan in 2016 epidemics. Total of 100 tracheal samples were considered for virus inoculation in 10 days old chicken embryonated eggs. The IBV infected amniotic fluid was neutralized with monoclonal antisera of H-120, 4/91 and D274 strains. The IBV screened samples were subjected for RNA extraction and subsequent to PCR using type specific primer of each strain. The amplified product of 840 bp was sequenced through Sanger sequencing. On the basis of PCR results, four similar amplified products from both regions were obtained showing similarities in agarose gel electrophoresis, but they differ from each other on the basis of nucleotides sequence. Phylogenetic analysis revealed that nucleotide sequences of isolates from Karachi were similar to the IBV H-120, Mass-41 and Connecticut 46 reference strains. Whereas, isolates from the Punjab province are analogous to the Mans-2, Mans-3, 9/41(UK) but did not show significant similarity with other reference strain. Therefore, it is recommended that use of M-41 and H-120 in vaccine production could be effective measure against velogenic infectious agent in Sindh particularly in Karachi, whereas, it would be better to incorporate either of the variant GQ281656.1, AY279533.1 in vaccine because of their highest level of resemblance with genetically sequenced isolates from Lahore a

This study was conducted to establish a high performance liquid chromatography（ HPLC） method for the determination of artemether in the artemether injection using Hypersil ODS C18 chromatographic column（ 5 μm,4. 6 mm × 150 mm）. Mobile phase,column temperature,flow rate and detection wavelength were optimized. Acetonitrile-water-tetrahydrofuran（ 62∶ 37∶ 1,V/V） was selected as the mobile phase,and the HPLC was performed with column temperature at30 ℃ and the flow rate at 1. 0 ml/min; and the detection wavelength was set at 216 nm. The HPLC detection system of artemether had good suitability. The linearity was good in 100-800 μg/ml concentration range,and the regression equation was y = 302. 36 x-682. 02,R2= 0. 999 8. The overall average recovery was97. 58%,and the RSD was 1. 58%. Three batches of artemether injection samples were determined by the method,showing RSD of 1. 42%. The method could be used for the detection of artemether content in artemether injection.

The lipid-secreting exocrine Harderian gland contains a large amount ofporphyrins (mainly protoporphyrin IX, PPIX)in the glandular cells, the physiologicalsignificance of which is rather poorly understood. In the present study, the possibilityof using Fura-2 to measure intracellular calcium ([Ca]) changes in these cells wasassessed. It was found tha when Fura-2 loaded oolls were excited by ligh at 340/380nm, [Ca] increased spontaneously, indicating a photodynamic action powered byligh at 340/380 nm. In contrast, with the visible. spectrum calcium probe Fluo-3 (λ475 nm), carbachol at 10 μM induced [Ca]. increase;[Ca] did not change withoutcarbachol stimulation. Objective:To determine the temporal patterns of cumulative incidence of brucellosis using autoregressive integrated moving average ***:This cross-sectional study employed yearly and monthly data of 1117 laboratory-confirmed human brucellosis cases from January 2013 to December 2018 using the Yazd brucellosis national surveillance *** monthly incidences constructed a timeseries *** trend of cumulative incidence was perceived by tracing a line plot,which displayed a seasonal trend with ***,the ARIMA models were ***,Akaike information criteria(AIC)and Bayesian information criterion(BIC)values among different models indicated a preferable model from models which were expanded by diverse lags[(3,0,3),(2,0,3),(3,0,2),(4,0,3)and(3,0,4)].Then,the achieved ARIMA model was applied to the forecasting cumulative incidence of monthly brucellosis *** analyses were performed using Stata,version ***:For the ARIMA(3,0,4)model,MAPE value was 56.20%with standard error 0.009–0.016,and white noise diagnostic check(Q=19.79,P=0.975)for the residuals of the selected model showed that the data were completely *** monthly incidences that were fitted by the ARIMA(3,0,4)model,with AIC(25.7)and BIC(43.35)with a similar pattern of actual cases from 2013 to 2018 and forecasting incidences from January 2019 to December 2019 were,respectively,0.50,0.44,0.45,0.49,0.55,0.58,0.56,0.51,0.46,0.44,0.45 and 0.49 per 100000 ***:In summary,the study showed that the ARIMA(3,0,4)model can be applied to forecast human brucellosis patterns in Yazd province,supplementing present surveillance systems,and may be better for health policy-makers and planners.

Antihelminthic resistance is a serious concern worldwide,whose mechanisms have been attributed to evolutionary changes in parasite genomes such as alteration of drug target sites,up-regulation of degrading enzymes and enhancement of drug ***,we describe a previously unknown mechanism of antihelminthic resistance:gut symbionts of the giant panda roundworm parasite *** would enhance resistance to a wide-spectrum antihelminthic drug *** documented that the gut symbiont bacteria Burkholderia sp.(called BS-B)and Citrobacter sp.(called BS-C) play key roles in the degradation of *** genomics revealed that compared to other congeneric species,BS-B had higher expressions of genes encoding glutathione S-transferases(GSTs) and multi-functional oxidases(MFOs),while BS-C had higher expressions of genes encoding carboxylesterase(CarEs) and cytochromes P450(P450 s).The transcriptions of these genes were also significantly up-regulated when ivermectin presented in cultures of BS-B and BS-C in ***,the ivermectin-sensitive *** inoculated with isolated BS-B and BS-C conferred ivermectin resistance,confirming the key roles of BS-B and BS-C in ivennectin resistance of *** together,our findings suggest possibility that the gut symbiont mediates antihelminthic resistance and also imply a more widely relevant drug resistance mechanism than previously recognized.

This paper reports results of the ecohealth project implemented in Yunnan Province, China. The results through literature review show that, as a notifiable disease, prevalence of Brucellosis has been significantly increased in both animal and human since the period from 2003 to 2005, and ranked the 1 st of importance in zoonosis since 2007;and as a non-notifiable disease, prevalence of Toxoplasmosis has been increased in both animal and human, at both national and provincial *** retrospective investigation, in hospital for human health, total case files of 22,689 outpatients and inpatients were investigated, of which 511 abortion cases of kinds and 9 cases of premature delivery were identified, and 2 cases were confirmed as serologically positive for Toxoplasmosis. Total case files of 56,883 outpatients and inpatients, of which 1,103 cases with fever, 341 with fever for further detection, and 236 with fever over 5 days;and 118 cases with joint pain, headache and lymph node enlargement were identified, however, all cases were confirmed to be serologically negative for Brucellosis. A serological survey for prevalence of Brucellosis and Toxoplasmosis in outpatient suspect case during the period from 1 st Jul to 31 st Dec（2011） was carried out in Yiliang County and Mangshi City. The results showed that prevalence was zero for Brucellosis and 9.68%（3/31） for Toxoplasmosis in Yiliang County, and prevalence was zero for Brucellosis and 16.67%（5/30） for Toxoplasmosis in Mangshi City. A field survey including Questionnaire, IDI and FGD was carried out in the two project sites. Total 192 respondents participated in the questionnaire survey;47 respondents involved in IDI;and 17 meetings for FGD were held. Findings and conclusions concerning knowledge, attitude and behavior for zoonosis and public health were achieved through this field survey. Generally, these two indicator diseases are not listed in the most common animal diseases or zoonosis;knowledge of these two

The present study was undertaken to investigate the effect of the ethanolic extract of Syzygium aromaticum to albino rats. Forty eight Albino rats were employed to test the safety and the anti-inflammatory effect of the extract. Safety of the extract was examined on experimental animal’s model at three dose levels of the extract orally in daily doses for three weeks. Effects of S. aromaticum on rats revealed no significant effect on biochemical or haematological parameters. The anti-inflammatory effect of the extract was tested in four equal groups;groups 1 and 2 were treated with 250 and 500 mg/kg of the extract, respectively, group 3 was treated with indomethacine and group 4 was the untreated control. Carrageenan was used as an acute form inducer of inflammation. Indomethacine, the non-steroidal anti-inflammatory drugs (NSAIDs), was used as a reference compound. Oedema size was monitored at the 1st, 2nd, 4th, 6th and after 24 hours. The ethanolic extract of S. aromaticum showed significant (P 0.001) decreased in the oedema size at efficacy rates of 79.41%, 82.39% and 63.92% for the dose, 500 mg/kg body weight at the 2nd, 4th and 6th hour respectively higher than that produced by indomethacine.
