PURPOSE: Toward improved glioblastoma muhiforme treatment, we determined whether celecoxib, a selective cyclooxygenase (COX)-2 inhibitor, could enhance glioblastoma radiosensitivity by inducing tumor necrosis and inhibiting tumor angiogenesis。METHODS AND MATERIALS: U-87MG ceils treated with celecoxib, irradiation, or both were assayed for clonogenic survival and anglogenie factor protein analysis (angiopoietin-1, angiopoietin-2, and vascular endothelial growth factor [VEGF])。In vivo, survival of mice intracranially implanted with U-87MG ceils and treated with celeeoxib and/or irradiation was monitored。Isolated tumors were assessed for tumor necrosis and tumor microvascular density by yon WiUiebrand' s factor (vWF) immunohistochemical staining。RE- SULTS: Celeeoxib (4 and 30 microM; 24, 48, and 72 h) enhanced U-87MG ceil radiosensitivity by significantly reducing clonogenic survival of irradiated cells。Angiopoietin-1 and VEGF proteins were decreased, whereas angiopoietin-2 expression increased after 72 h of celeeoxib alone and when combined with irradiation。

The mainstay of type 2 diabetes treatment used to be blood glucose control in order to prevent or delay the development of various diabetic complications. On September 1, 2018, one of our patients was diagnosed with several serious illnesses: type 2 diabetes, 3 degrees of obesity, arterial hypertension, peripheral vascular atherosclerosis of the lower extremities with swelling of the shins and feet, fatty hepatosis and metabolic syndrome since June 2019. The patient was restricted in movement due to excessive body weight, shortness of breath and joint pain. Conventional medical methods did not work well for him, so the prognosis is very poor. In this case, we used a new frequency-code therapy which could transmit the signal into the patient's brain by the method of frequency code influence to cure diabetes and associated diseases, restore the patient's body and improve his living quality. According to our forecasts in September 2018 the treatment of acquired diseases and the restoration of the patient's body to a viable level, with the help of our technology, may take more a year. Carrying out step-by-step noninvasive treatment by the method of frequency-code influence on the patient's brain we received positive results at each stage of treatment which together gave a complex positive result in ridding the patient of acquired diseases and restoring his body to the level of active life-activity without restrictions. Now the patient does not follow a diet and enjoys physical activity. No side effects were observed. The human body is unique, even in adulthood, the protective functions of the body are able to defeat the most complex diseases without damage to the body. To do this need to activate the "sleeping protective mechanism" of the body or translate the "healthy memory of the brain" in the period of active resistance to premature age-related diseases.

Cannabinoids, the active components of Cannabis sativa Linnaeus, have received renewed interest in recent years due to their diverse pharmacologic activities such as cell growth inhibition, anti-inflammatory effects and tumor regression, but their use in chemotherapy is limited by their psychotropic activity. To date, cannabinoids have been successfully used in the treatment of nausea and vomiting, two common side effects that accompany chemotherapy in cancer patients. Most non-THC plant cannabinoids e.g. cannabidiol and cannabigerol, seem to be devoid of psychotropic properties. However, the precise pathways through which these molecules produce an antitumor effect have not yet been fully characterized. We therefore investigated the antitumor and anti-inflammatory activities of cannabidiol (CBD) in human prostate cancer cell lines LNCaP, DU145, PC3, and assessed whether there is any advantage in using cannabis extracts enriched in cannabidiol and low in THC. Results obtained in a panel of prostate cancer cell lines clearly indicate that cannabidiol is a potent inhibitor of cancer cell growth, with significantly lower potency in non-cancer cells. The mRNA expression level of cannabinoid receptors CB1 and CB2, vascular endothelial growth factor (VEGF), PSA (prostate specific antigen) are significantly higher in human prostate cell lines. Treatment with Cannabis extract containing high CBD down regulates CB1, CB2, VEGF, PSA, pro-inflammatory cytokines/chemokine IL-6/IL-8. Our overall findings support the concept that cannabidiol, which lacks psychotropic activity, may possess anti-inflammatory property and down regulates both cannabinoid receptors, PSA, VEGF, IL-6 and IL-8. High CBD cannabis extracts are cytotoxic to androgen responsive LNCaP cells and may effectively inhibit spheroid formation in cancer stem cells. miRNAs play important roles in the post-transcriptional regulation of most transcripts and control several biological processes. We have shown that miR-200b regulates VEGF and miR-146a regulates ECM （extracellular matrix） protein, FN （fibronectin） production. We examined the effects of these two miRNAs in wound healing in vitro and in animals with or without diabetes. Microvascular ECs （endothelial cells） with or without miR inhibitor （antagomir） transfection were tested for VEGF and FN production. Scratch assays and angiogenesis assays were performed. Wounds in the back of mice with or without streptozotocin-induced diabetes were treated with antagomirs alone or in combinations. The wounds were measured and tissues were examined for mRNAs, proteins and miRNAs and examined histologically. In the ECs, miR-200b or miR-146a inhibitors increased VEGF and FN production, cell migration and tube formation. Wound healing in the animals, along with increased production of specific proteins were accelerated by miR-200b or miR-146a inhibitor treatment and was pronounced when these two were combined In diabetes, although delayed, similar patterns were seen. These results indicated that combination of miR-146a and miR-200b inhibitor treatment is useful in wound healing both in normal and in diabetic conditions, miRNA mediated wound healing may potentially constitute a novel therapeutic approach.

Distally based perforator sural flaps from the posterolateral or posteromedial lower leg aspect are initially a neurofasciocutaneous flap that can be transferred reversely to the foot and ankle region with no need to harvest and sacrifice the deep major artery. These flaps are supplied by a perforating artery issued from the deep peroneal artery or the posterior tibial artery, and the chainlinked adipofascial neurovascular axis around the sural/saphenous nerve. It is a versatile and reliable technique for soft-tissue reconstruction of the heel and ankle region with 180-degrees rotation. In this paper, we present its developing history, vascular basis, surgical techniques including flap design and elevation, flap variations in pedicle and component, surgical indications, and illustrative case reports with different perforating vessels as pivot points for foot and ankle coverage.

Small-diameter artificial blood vessels are prone to cause intimal hyperplasia(IH)after transplantation,which leads to restenosis and low long-term patency *** main biomechanical factor for IH formation is the compliance mismatch between the artificial and host blood vessels which can cause abnormal *** there have been many studies on vascular compliance mismatches,however,little attention has been paid to the effect of the degree of compliance mismatch between graft and the host vessel on *** present,the research on compliance mismatch between the artificial and host blood vessels is still very limited,especially with regard to the specific impact of the compliance mismatch degree on ***,three end-to-end anastomosis models(the compliance of the artificial blood vessel is lower than,similar to,and higher than that of the host blood vessel,called model 1,model 2,model 3,respectively)were constructed and simulated in this *** results showed that the radial displacement difference between the artificial and host blood vessels were 0.281 mm,0.183 mm and 0.485 mm in model 1,model 2 and model 3,respectively.A low-velocity recirculation zone near the distal anastomosis was formed in model 1 which resulted in excessively low TAWSS(9.261 E-5Pa)and high OSI(0.497).Similarly,a low-velocity recirculation zone near the proximal anastomosis was formed in model 3 and lead to low TAWSS(6.007 E-4Pa)and high OSI(0.480).However,there was no low-velocity recirculation zone near the anastomosis stoma in model *** results are instructive for the design and preparation of artificial blood vessels.
