AICAR-based exercise mimetic alleviates aging-induced memory impairment through activating AMPK and improving mitochondrial quality control

Growing evidence suggests that exercise can provide neuroprotection by improving mitochondrial quality control (MQC) on the aged brain. Adenosine 5'-monophosphate (AMP)-activated protein kinase (AMPK) signaling responsiveness declines with aging. However， whether AMPK plays a role in the exercise-mediated improvement of memory and MQC in the aged hippocampus remains to be established. AICAR (5-Aminoimidazole 4-carboxamide ribonucleoside)， a pharmacological agonist of AMPK， has been proposed to be an exercise mimetic recently. However， it has not been clarified whether AICAR could mimic the effects of exercise on the aged hippocampus through improvement of MQC. In this study， AICAR (AMPK agonist) and Compound C (AMPK inhibitor) were used to investigate if AMPK plays a key role in exercise-induced improvement of MQC and if AICAR could act as an exercise mimetic through improvement of MQC in aged hippocampus. Both exercise and AICAR improved the memory of aged mice and increased AMPK phosphorylation in the aged hippocampus. Exercise， but not AICAR， improved mitochondrial respiratory function in the aged hippocampus and increased the Microtubule associated protein 1 light chain 3 (LC3) II/LC3 I ratio and the protein expression of LC3 II and Autophagy Related Protein 7 (ATG7) in the lysate of whole hippocampal tissue. Both exercise and AICAR increased the ratio of LC3 II/LC3 I and the protein expression of LC3 II in the mitochondrial fractions of the hippocampus. Regarding mitochondrial dynamics， neither exercise training nor AICAR changed the protein level of Mitofusin2 (Mfn2). Exercise， but not AICAR， increased the protein level of Dynamin-related protein 1 (Drp1). Furthermore， both exercise training and AICAR increased the protein level of peroxisome proliferator-activated receptor γ coactivator 1α (PGC-1α)， a modulator of mitochondrial biogenesis. Acupotomy ameliorates knee osteoarthritis-related collagen deposition and fibrosis in rabbit skeletal muscle through the TGF-β/Smad pathway

Objective To investigate the effects of acupotomy on skeletal muscle fibrosis and collagen deposition in a rabbit knee osteoarthritis(KOA)*** Rabbits(n=18)were randomly divided into control,KOA,and KOA+acupotomy(Apo)groups(n=6).The rabbits in the KOA and Apo groups were modeled using the modified Videman's method for 6 *** modeling,the Apo group was subjected to acupotomy once a week for 3 weeks on the vastus medialis,vastus lateralis,rectus femoris,biceps femoris,and anserine bursa tendons around the *** behavior of all animals was recorded,rectus femoris tissue was obtained,and histomorphological changes were observed using Masson staining and transmission electron *** expression of transforming growth factor-β1(TGF-β1),Smad 3,Smad 7,fibrillar collagen types I(Col-I)and III(Col-III)was detected using Western blot and real-time polymerase chain reaction(RT-PCR).Results Histological analysis revealed that acupotomy improved the microstructure and reduced the collagen volume fraction of rectus femoris,compared with the KOA group(P=.034).Acupotomy inhibited abnormal collagen deposition by modulating the expression of fibrosis-related proteins and mRNA,thus preventing skeletal muscle *** blot and RT-PCR analysis revealed that in the Apo group,Col-I,and Col-III protein levels were significantly lower than those in the KOA group(both P<.01),same as Col-I and Col-III mRNA levels(P=.0031;P=.0046).Compared with the KOA group,the protein levels of TGF-β1 and Smad 3 were significantly reduced(both P<.01),as were the mRNA levels of TGF-β1 and Smad 3(P=.0007;P=.0011).Conversely,the levels of protein and mRNA of Smad 7 were significantly higher than that in the KOA group(P<.01;P=.0271).Conclusion Acupotomy could alleviate skeletal muscle fibrosis and delay KOA progress by inhibiting collagen deposition through the TGF-β/Smad pathway in the skeletal muscle of KOA rabbits.

Purpose: Low back pain (LBP), Gonarthrosis, knee, and Carpal Tunnel Syndrome have been regarded as affecting more than three-quarters’ of individuals in their lifetime. The aim of this study was conducted to determine and compare the effects of topical patients with sand compared with control related to Gonarthrosis, LBP, knee, and Carpal Tunnel Syndrome pain. Methods: This is a prospective case and control designed study based on 101 pain case with Sand and 101 control subjects. The interventional groups in addition either topical sand or without sand 2 months after the intervention were assessed. We assessed tolerability of an established pain perception scale by the Numeric Rating Scale (NRS). Categorical variables were compared using the chi-square (χ2) test. The paired t-test was used to compare the two groups before and after the intervention. One-way analysis of variance (ANOVA) was employed for comparison of several group means. Results: The study was based on 202 patients, 41 males (20.3%) and 161 females (79.7%). There were statistically significant differences between subjects with and without Sand in term of Gonarthrosis, Rheumatoid Arthritis LBP, knee, and Carpal Tunnel Syndrome pain (p = 0.033). Majority of patients were over age 55 years old (55%) and females 78 (77.2%). The mean score of total pain experience before and after the intervention was 7.41 ± 1.1 for black sand, and 4.24 ± 2.38 for without sand as control group. Relationship between Nonexercise Activity Thermogenesis and Calf Circumference in Older Women

Purpose: Frailty is a state of declined vitality of the body and mind with age in which life functions are impaired. In addition, there is a difference in the susceptibility of older women to frailty compared with that of older men. Therefore, assessing and encouraging physical activity in older adults before they become frail is essential. We aimed to clarify the relationship between calf circumference and body composition and assess the potential association between calf circumference and physical activity indices in older women. Methods: This cross-sectional study included 18 healthy older adults (age 69.0 ± 5.7 years). The physical characteristics, calf circumference, body composition, calf muscle thickness, and physical activity questionnaire that included items on exercise-related activity thermogenesis (EAT) and nonexercise activity thermogenesis (NEAT) were assessed. The association between calf circumference and these additional measures was examined. Results: Positive and significant correlations were found between the calf circumference and body weight, body mass index (BMI), skeletal muscle mass, skeletal muscle mass index, calf muscle thickness, total questionnaire score, and NEAT score (r = 0.66 - 0.87). However, no significant correlations were observed between the calf circumference and EAT scores. Conclusion: Calf circumference in older women may reflect NEAT activity. Improving NEAT activity is an important health-promoting factor in older women.
