PoreScript:Semi-automated pore size algorithm for scaffold characterization

The scaffold pore size influences many critical physical aspects of tissue engineering,including tissue infiltration,biodegradation rate,and mechanical properties.Manual measurements of pore sizes from scanning electron micrographs using ImageJ/FIJI are commonly used to characterize scaffolds,but these methods are both time-consuming and subject to user bias.Current semi-automated analysis tools are limited by a lack of accessibility or limited sample size in their verification process.The work here describes the development of a new MATLAB algorithm,PoreScript,to address these limitations.The algorithm was verified using three common scaffold fabrication methods(e.g.,salt leaching,gas foaming,emulsion templating)with varying pore sizes and shapes to demonstrate the versatility of this new tool.Our results demonstrate that the pore size characterization using PoreScript is comparable to manual pore size measurements.The PoreScript algorithm was further evaluated to determine the effect of user-input and image parameters(relative image magnification,pixel intensity threshold,and pore structure).Overall,this work validates the accuracy of the PoreScript algorithm across several fabrication methods and provides user-guidance for semi-automated image analysis and increased throughput of scaffold characterization.

A longitudinal network analysis of social dynamics in rooks corvus frugilegus： repeated group modifications do not affect social network in captive rooks

Numerous studies have investigated the remarkable variation of social features and the resulting structures across species. Indeed, relationships are dynamic and vary in time according to various factors such as environmental conditions or individuals attributes. However, few studies have investigated the processes that stabilize the structures within a given species, and the behavioral mechanisms that ensure their coherence and continuity across time. Here, we used a dynamic actor-based model, RSiena, to investigate the consistency of the temporal dynamic of relationships of a group of captive rooks facing recurrent modifications in group composition (i.e., the loss and introduction of individuals). We found that changes in relationships (i.e., formation and removal) followed consistent patterns regardless of group composition and sex-ratio. Rooks preferentially interacted with paired congeners (i.e., unpopular attachment) and were more likely to form relationships with individuals bonded to a current social partner (i.e., “friends of friends”, or triadic closure). The sex of individuals had no effect on the dynamic of relationships. This robust behavioral mechanisms formed the basis of inter-connected networks, composed of sub-structures of individuals emerging from the enmeshment of dyadic and triadic motifs. Overall, the present study reveals crucial aspects of the behavioral mechanisms shaping rooks social structure, suggesting that rooks live in a well-integrated society, going far beyond the unique monogamous pair-bond.

On the Chow Groups of Certain Cubic Fourfolds

This note is about the Chow groups of a certain family of smooth cubic fourfolds. This family is characterized by the property that each cubic fourfold X in the family has an involution such that the induced involution on the Fano variety F of lines in X is symplectic and has a K3 surface S in the fixed locus. The main result establishes a relation between X and S on the level of Chow motives. As a consequence, we can prove finite-dimensionality of the motive of certain members of the family. Morphology, flight performance, and water crossing tendencies of Afro-Palearctic raptors during migration

Raptors primarily use soaring-gliding flight which exploits thermals and ridge lifts over land to reduce energetic costs. However during migration, these birds often have to cross water surfaces where thermal currents are weak; during these times, birds mainly use flapping (powered) flight which increases energy consumption and mortality risk. As a result, some species have evolved strategies to reduce the amount of time spent over water by taking extensive detours over land. In this paper, we conducted a meta-analysis of water-crossing tendencies in Afro-Palearctic migrating raptors in relation to their morphology, their flight performance, and their phylogenetic relationships. In particular, we considered the aspect ratio (calculated as the wing span squared divided by wing area), the energetic cost of powered flight, and the maximum water crossing length regularly performed by adult birds. Our results suggest that energy consumption during powered flight predominately affects the ability of raptors to fly over water surfaces.

Process conditions for preparing well-defined nano- and microparticles as delivery systems of alkyl gallates

dextran-covered poly(D,L-lactide) (PLA) nano-and microparticles were prepared using an emulsion/solvent evaporation (or diffusion) process for the encapsulation of alkyl gallates (AGs).In the first step,a solution of PLA and AG in ethyl acetate was emulsified to give an aqueous phase containing a hydrophobically modified dextran,which acted as a stabilizer.The second step involved solvent evaporation or diffusion.The emulsification conditions were varied,which allowed for the preparation of nanoand microparticle suspensions covering a wide range of surface-average particle diameters from 0.1 μm (sonication) to 500 μm (stirring with a magnetic bar),with narrow and reproducible size distributions.Continuous microfluidic emulsification in a flow-focusing system led to well-defined microparticles,in the 10-50 μm range.Particles loaded with octyl gallate (OG) and nonyl gallate (NG) were obtained using the three processes,and we showed that the encapsulation efficiency of OG and NG varied significantly depending on the emulsification process.The effect of particle size on the mechanism of in vitro release of encapsulated AGs was investigated.The kinetics of release were controlled either by Fickian diffusion within the solid core or swelling and hydrolytic degradation of the PLA matrix,depending on the pH of the external medium.

Hepatitis C virus(HCV)is a main cause and important risk factor for hepatocellular carcinoma(HCC)in Western countries(1).Direct antiviral agents(DAAs)for HCV are highly effective and well-tolerated antiviral drugs which allow HCV cure even in patients with advanced liver disease.However,whilst HCV cure improves liver function and decreases overall HCC risk,a significant HCC risk persists in particular in patients with advanced fibrosis including cirrhosis who have the highest risk of developing HCC(2-4).

Mediator contributes to IgH locus VDJ rearrangements by promoting usage of most distal V segments

INTRODUCTION The immunoglobulin heavy(IgH)chain locus includes a large cluster of V,D and J segments spanning thousands of kilobases in mammals and its expression implies long-range 3D-regulation of various gene modifications.1 V(D)J recombination occurs during early B-cell ontogeny.2 It is regulated by multiple transcription factors and cis-regulatory elements,and most notably by the intronic 5′Eμelement located in the JH to Cμintron.3 The intergenic control region 1(IGCR1)located between the VH and D gene clusters inhibits rearrangement of the most DH-proximal VH gene segments,while promoting usage of distal VH gene segments.4 Other downstream elements of the locus,flanking its 3′end might have more subtle roles:while the large 3′regulatory region(3′RR)shows strong internal synergies2 and behaves as a superenhancer in mature B cells.Its deletion showed no influence on the VDJ repertoire and rather increased transcription of unrearranged V segments.3–5 Insulators binding CTCF farther downstream of the 3′RR were shown,however,to promote usage of distal VH segments.6 Altogether,there are strong indications that elements involved in long-distance regulation of transcription might also influence VDJ recombination.In-frame splicing of VDJ exons is also needed for full Ig expression.7 Given the functional importance of gene architecture and long-range interactions for the IgH locus physiology,we explored the dependence of the VDJ repertoire on Med1,a known actor of long-range enhancer interactions.

Chromatin-remodeling factor OsINO80 is involved in regulation of gibberellin biosynthesis and is crucial for rice plant growth and development

The phytohormone gibberellin（GA） plays essential roles in plant growth and development. Here,we report that OsINO80, a conserved ATP-dependent chromatin-remodeling factor in rice（Oryza sativa）, functions in both GA biosynthesis and diverse biological processes. OsINO80-knockdown mutants, derived from either T-DNA insertion or RNA interference, display typical GA-deficient phenotypes, including dwarfism, reduced cell length, late flowering, retarded seed germination and impaired reproductive development. Consistently, transcriptome analyses reveal that OsINO80 knockdown results in downregulation by more than two-fold of over 1,000 genes, including the GA biosynthesis genes CPS_1 and GA_3ox_2, and the dwarf phenotype of OsINO80-knockdown mutants can be rescued by the application of exogenous GA3. Chromatin immunoprecipitation（Ch IP） experiments show that OsINO80 directly binds to the chromatin of CPS1 and GA_3ox_2 loci. Biochemical assays establish that OsINO80 specially interacts with histone variant H_2A.Z and the H_2A.Z enrichments at CPS_1 and GA_3ox_2 are decreased in OsINO80-knockdown mutants. Thus, our study identified a rice chromatin-remodeling factor,OsINO80, and demonstrated that OsINO80 is involved in regulation of the GA biosynthesis pathway and plays critical functions for many aspects of rice plant growth and development.

Conductance hysteresis and inelastic excitations at hydrogen decorated cerium atoms and clusters in a tunnel junction

Voltage-controlled conductance and switching induced by single molecules or atoms are ideally studied in scanning tunneling microscope （STM） tunnel junctions. While the objects under consideration are mostly used in their original form, little is known of the possibilities of in situ adjustments of their properties. Here, we evidence properties of a tunnel junction made of a Ce atom/cluster built by atomic manipulation on Au（111） at a temperature of 4.6 K in the presence of H2. The conductance through the object is characterized by a switching voltage corresponding to an opening or closing of an inelastic electron tunneling conductance channel at 50 mV for a Ce atom and 140 mV for a Ce cluster and by charging. We demonstrate that the electronic properties of an STM junction can be engineered in a simple way by in situ guiding of the H2 pinning at an atomic cluster.

盘状液晶作为有机光电功能材料具有学术研究价值和应用开发潜力.通过Suzuki偶联反应合成了星型1,3,5-三嗪-苯并菲盘状液晶TPPTn(n=6,8,12).化合物在溶液、薄膜与固体态都有良好的荧光发射,具有聚集诱导效应,有效克服了盘状分子聚集诱导荧光猝灭的弊端,烷氧链长度的改变对化合物溶液、薄膜或固体的荧光性质影响相对较小.重点研究的TPPT6呈现出由蓝到橙显著的溶致变色,绝对量子产率最高可达43%;TPPT6在有机溶剂中呈现出较传统分子的升温猝灭不同的随温度升高荧光增强的温度刺激响应;TPPT6还具有明显的酸碱刺激响应,核磁滴定证明质子化过程是导致酸致变色的关键;TPPTn系列化合物自组装呈矩形柱状中间相(Colrec),柱状相温度最宽达278℃,三氟乙酸通过对三嗪离子化可调控TPPT的介晶行为.此外, TPPT6可用于制备安全墨水以及绿光有机发光二极管.该系列化合物在液晶半导体、荧光传感、信息加密和有机发光二极管等领域具有潜在的应用价值.
