A Non-canonical Transferred DNA Insertion at the BRI1 Locus in Arabidopsis thaliana

Agrobacterium-mediated transformation is widely used in transgenic plant engineering and has been proven to be a powerful tool for insertional mutagenesis of the plant gsnome. The transferred DNA （T-DNA） from Agrobacterium is integrated into the plant genome through illegitimate recombination between the T-DNA and the plant DNA. Contrasting to the canonical insertion, here we report on a locus showing a complex mutation associated with T-DNA insertion at the BRI1 gene in Arabidopsis thaliana. We obtained a mutant line, named salads for its phenotype of dwarf stature and proliferating rosette. Molecular characterization of this mutant revealed that in addition to T-DNA a non-T-DNA-Iocalized transposon from bacteria was inserted in the Arabidopsis genome and that a region of more than 11.5 kb of the Arabidopsis genome was deleted at the insertion site. The deleted region contains the braesinosteroid receptor gene BRI1 and the transcription factor gene WRKY13. Our finding reveals non-canonical T-DNA insertion, implicating horizontal gene transfer and cautioning the use of T-DNA as mutagen in transgenic research.

Real case of primitive embryonal duodenal carcinoma in a young man

We report here the case of a young man suffering from a rare germ cell tumour. The patient was a 25-yearold man who was referred to our centre for asthenia, stinging epigastric pain, and an iron deficiency anaemia. Gastroscopy revealed a circumferential vegetating lesion on the second portion of the duodenum. The lesion was indurated at the third portion of the duodenum, responsible for a tight stenosis. A computerized tomography-scan of the chest, abdomen and pelvis, and a pancreatic MRI showed a circumferential lesion with a bi-ductal dilatation(i.e., of the common bile duct and Wirsung's duct) without metastatic localisation. The patient underwent a pancreaticoduodenectomy with lymph node dissection including all cellular adipose tissues of the hepatic pedicle from the hepatic common artery and of the retroportal lamina. Histological findings were suggestive of a duodenal embryonal carcinoma with pancreatic infiltration. This is the second published case highlighting the duodenal primitive localisation of an embryonal carcinoma with pancreatic infiltration.

Plant phenotypic plasticity is controlled by diverse hormone pathways, which integrate and convey infor- mation from multiple developmental and environmental signals. Moreover, in plants many processes such as growth, development, and defense are regulated in similar ways by multiple hormones. Among them, gibberellins （GAs） are phytohormones with pleiotropic actions, regulating various growth processes throughout the plant life cycle. Previous work has revealed extensive interplay between GAs and other hor- mones, but the molecular mechanism became apparent only recently. Molecular and physiological studies have demonstrated that deLLA proteins, considered as master negative regulators of GA signaling, inte- grate multiple hormone signaling pathways through physical interactions with transcription factors or reg- ulatory proteins from different families. In this review, we summarize the latest progress in GA signaling and its direct crosstalk with the main phytohormone signaling, emphasizing the multifaceted role of deLLA proteins with key components of major hormone signaling pathways.

Unique expansions in non-integer bases have been investigated in many papers during the last thirty years.They are often conveniently generated by labeled directed graphs.We give a precise description of the set of sequences generated by these graphs.This provides a geometric explanation of many former abstract results in this domain.Our results are illustrated by many examples.

Most metal-organic frameworks(MOFs) hardly maintain their physical and chemical properties after exposure to acidic,neutral,or alkaline aqueous solutions,resulting in insufficient stability,therefore limiting their applications.Thus,the design and synthesis of stable size/morphology-controlled MOF nanocrystals is critical but challenging.In this study,dual-ligand and hard-soft-acid-base strategies were used to fabricate a variety of 3D pillared-layer [Ni(thiophene-2,5-dicarboxylate)(4,4’-bipyridine)]nMOF nanocrystals(1D nanofibers,2D nanosheets and 3D aggregates) with controllable morphology by varying the concentration of 4,4’-bipyridine and thus controlling the crystal growth direction.Owing to the shorter ion diffusion length,enhanced ele ctron/ion transfer and strong interactions between thiophene-2,5-dicarboxylate and4,4’-bipyridine,the 2D nanosheets showed much larger specific capacitance than 1D nanofibers and 3D aggregates.A single device with an output voltage as high as 3.0 V and exceptional cycling performance(95% of retention after 5000 cycles at 3 mA cm-2) was realized by configuring two aqueous asymmetric super capacitive devices in series.The excellent cycling property and charge-discharge mechanism are consistent with the hard-soft-acid-base theory.

In a context of a virus that is transmissive by sputtering,wearing masks appear necessary to protect the wearer and to limit the propagation of the disease.Currently,we are facing the 2019-2020 coronavirus pandemic.Coronavirus disease 2019(COVID-19)is an infectious disease with first symptoms similar to the flu.The symptom of COVID-19 was reported first in China and very quickly spreads to the rest of the world.The COVID-19 contagiousness is known to be high by comparison with the flu.In this paper,we propose a design of a mobile application for permitting everyone having a smartphone and being able to take a picture to verify that his/her protection mask is correctly positioned on his/her face.Such application can be particularly useful for people using face protection mask for the first time and notably for children and old people.The designed method exploits Haar-like feature descriptors to detect key features of the face and a decision-making algorithm is applied.Experimental results show the potential of this method in the validation of the correct mask wearing.To the best of our knowledge,our work is the only one that currently proposes a mobile application design“CheckYourMask”for validating the correct wearing of protection mask.

Whereas prone positioning of intubated patients suffering from acute respiratory distress syndrome represents the standard of care,proning non-intubated patients,so-called“awake prone positioning(APP),”has only recently gained popularity and undergone scientific evaluation.In this review,we summarize current evidence on physiological and clinical effects of APP on patients’centered outcomes,such as intubation and mortality,the safety of the technique,factors and predictors of success,practical issues for optimal implementation,and future areas of research.Current evidence supports using APP among patients suffering from acute hypoxemic respiratory failure due to COVID-19 and undergoing advanced respiratory support,such as high-flow nasal cannula,in an intensive care unit setting.Healthcare teams should aim to prone patients at least 8 h daily.Future research should focus on optimizing the tolerance of the technique and comprehensively evaluating benefits in other patient populations.
