Development of NK cell-based cancer immunotherapies through receptor engineering

Natural killer(NK)cell-based immunotherapies are attracting increasing interest in the field of cancer treatment.Early clinical trials have shown promising outcomes,alongside satisfactory product efficacy and safety.Recent developments have greatly increased the therapeutic potential of NK cells by endowing them with enhanced recognition and cytotoxic capacities.This review focuses on surface receptor engineering in NK cell therapy and discusses its impact,challenges,and future directions.Most approaches are based on engineering with chimeric antigen receptors to allow NK cells to target specific tumor antigens independent of human leukocyte antigen restriction.This approach has increased the precision and potency of NK-mediated recognition and elimination of cancer cells.In addition,engineering NK cells with T-cell receptors also mediates the recognition of intracellular epitopes,which broadens the range of target peptides.Indirect tumor peptide recognition by NK cells has also been improved by optimizing immunoglobulin constant fragment receptor expression and signaling.Indeed,engineered NK cells have an improved ability to recognize and destroy target cells coated with specific antibodies,thereby increasing their antibodydependent cellular cytotoxicity.The ability of NK cell receptor engineering to promote the expansion,persistence,and infiltration of transferred cells in the tumor microenvironment has also been explored.Receptor-based strategies for sustained NK cell functionality within the tumor environment have also been discussed,and these strategies providing perspectives to counteract tumor-induced immunosuppression.Overall,receptor engineering has led to significant advances in NK cell-based cancer immunotherapies.As technical challenges are addressed,these innovative treatments will likely reshape cancer immunotherapy.

Engineered nanoparticles have emerged as new types of materials for a wide range of applications from therapeutics to energy.Still,fabricating nanomaterials presenting complex inner morphologies and shapes in a simple manner remains a great challenge.Herein,we report the template-free one-pot continuous gradient nanoprecipitation of different types of non-compatible polymers to spontaneously form nanostructured particles.The continuous addition of antisolvent induces precipitation and(re)organization of polymer chains at the forming particle interface,ultimately and naturally developing complex inner morphologies and shapes while particle grows.This low-energy-cost bottom-up assembly approach applies to various functional polymers,possibly embedded with metal nanoparticles,for continuous growth into well-organized nanoparticles.UV crosslinking of the particles and core removal allows both confirming the building process and leading to hollow or multivoid nanomaterials.

Decoding the nexus:branched-chain amino acids and their connection with sleep,circadian rhythms,and cardiometabolic health

The sleep-wake cycle stands as an integrative process essential for sustaining optimal brain function and,either directly or indirectly,overall body health,encompassing metabolic and cardiovascular well-being.Given the heightened metabolic activity of the brain,there exists a considerable demand for nutrients in comparison to other organs.Among these,the branched-chain amino acids,comprising leucine,isoleucine,and valine,display distinctive significance,from their contribution to protein structure to their involvement in overall metabolism,especially in cerebral processes.Among the first amino acids that are released into circulation post-food intake,branched-chain amino acids assume a pivotal role in the regulation of protein synthesis,modulating insulin secretion and the amino acid sensing pathway of target of rapamycin.Branched-chain amino acids are key players in influencing the brain's uptake of monoamine precursors,competing for a shared transporter.Beyond their involvement in protein synthesis,these amino acids contribute to the metabolic cycles ofγ-aminobutyric acid and glutamate,as well as energy metabolism.Notably,they impact GABAergic neurons and the excitation/inhibition balance.The rhythmicity of branchedchain amino acids in plasma concentrations,observed over a 24-hour cycle and conserved in rodent models,is under circadian clock control.The mechanisms underlying those rhythms and the physiological consequences of their disruption are not fully understood.Disturbed sleep,obesity,diabetes,and cardiovascular diseases can elevate branched-chain amino acid concentrations or modify their oscillatory dynamics.The mechanisms driving these effects are currently the focal point of ongoing research efforts,since normalizing branched-chain amino acid levels has the ability to alleviate the severity of these pathologies.In this context,the Drosophila model,though underutilized,holds promise in shedding new light on these mechanisms.Initial findings indicate its potential to introduce novel concepts,particularly

The Ain El Bey abandoned mine, in North-West Tunisia, fits into the geodynamic context of the European and African plate boundary. Ore deposit corresponds to veins and breccia of multiphase Cu–Fe-rich mineralization related to various hydrothermal fluid circulations. Petromineralogical studies indicate a rich mineral paragenesis with a minimum of seven mineralization phases and, at least, six pyrite generations. As is also the case for galena and native silver, native gold is observed for the first time as inclusion in quartz which opens up, thus, new perspectives for prospecting and evaluating the potential for noble metals associated with the mineralization. Scanning Electron Microscope--Energy Dispersive Spectroscopy and Transmission electron microscopy analyses show, in addition, a large incorporation of trace elements, including Ag and Au, in mineral structures such as fahlores(tetrahedrite-tennantite) and chalcopyrite ones. The mineral/mineral associations, used as geothermometers, gave estimated temperatures for the mineralizing fluids varying from 254 to 330 ℃ for phase Ⅲ, from 254 to 350 ℃ for phase Ⅳ, and from 200 to 300 ℃ for phases Ⅴ and Ⅵ. The seventh and last identified mineralization phase, marked by a deposit of native gold, reflects a drop in the mineralizing fluid’s temperature(< 200 ℃) compatible with boiling conditions. Such results open up perspectives for the development of precious metal research and the revaluation of the Cu–Fe ore deposit at the Ain El Bey abandoned mine, as well as at the surrounding areas fitting in the geodynamic framework of the Africa-Europe plate boundary.

Artificial light at night decreases the pupillary light response of dark-adapted toads to bright light

Artificial light at night(ALAN)is expanding worldwide.Many physiological effects have been reported in animals,but we still know little about the consequences for the visual system.The pupil contributes to control incoming light onto the retina.Sudden increases in light intensity evokes the pupil light reflex(PLR).Intrinsically photosensitive retinal ganglion cells(ipRGC)affect PLR and melatonin expression,which largely regulate circadian rhythms and PLR itself.IpRCG receive inputs from various photoreptors with different peak sensitivities implying that PLR could be altered by a broad range of light sources.We predicted ALAN to enhance PLR.Contrary to our prediction,dark-adapted cane toads Rhinella marina,exposed to ALAN(5 lx)for 12 days,exhibited a lower PLR than controls and individuals exposed to 0.04 lx,even after 1 h in bright light.We cannot conclude whether ALAN induced a larger pupil size in dark-adapted toads or a slower initial contraction.Nevertheless,the response was triggered by a light source with an emission peak(590 nm)well above the sensitivity peak of melanopsin,the main photoreceptor involved in PLR.Therefore,ALAN alters the capacity of toads to regulate the incoming light in the eye at night,which may reduce the performance of visually guided behaviors,and increase mortality by predators or road kills at night.This first study emphasizes the need to focus on the effect of ALAN on the vision of nocturnal organisms to better understand how this sensory system is altered and anticipate the consequences for organisms.

Discrimination of conspecifics from heterospecifics in a hybrid zone: Behavioral and chemical cues in ants

Species and nestmate recognition in social insects occurs mostly through cuticular hydrocarbons acting as chemical cues. These compounds generate a colony-specific odor profile depending on genetic and environmental factors. Species and nestmate recognition results in specific behavioral responses, regulating the level of aggression toward other individuals during an interaction. Although species discrimination and recognition cues have been poorly studied in the context of interspecific hybridization, such systems offer an opportunity to further investigate the influence of heritable and environmental factors on recognition. We explored the strength of discrimination in a hybrid zone between two ant species—Tetramorium immigrans and T. caespitum—by comparing cuticular hydrocarbon profiles and measuring intra- and interspecific worker aggression in both areas of sympatry and areas of allopatry among species. Species cuticular hydrocarbon profiles were well-differentiated and interspecific aggression was high, revealing highly discriminating species recognition cues. Hybrids’ cuticular hydrocarbon profiles consisted of a mixture of the parental bouquets, but also exhibited hybrid-specific patterns. Behavioral assays showed that T. immigrans is as aggressive toward hybrids as toward heterospecifics. Finally, aggression between heterospecific workers was lower when interacting individuals came from areas of sympatry among species than from areas of allopatry. Taken as a whole, these findings paint a particularly complex picture of the recognition system in T. immigrans, T. caespitum, and their hybrids, and highlight that hybrid zones afford a still underexplored opportunity for investigating recognition mechanisms and discrimination between species.

To develop durable bone healing strategies through improved control of bone repair,it is of critical importance to understand the mechanisms of bone mechanical integrity when in contact with biomaterials and implants.Bone mechanical integrity is defined here as the adaptation of structural properties of remodeled bone in regard to an applied mechanical loading.Accordingly,the authors present why future investigations in bone repair and regeneration should emphasize on the matrix surrounding the osteocytes.Osteocytes are mechanosensitive cells considered as the orchestrators of bone remodeling,which is the biological process involved in bone homeostasis.These bone cells are trapped in an interconnected porous network,the lacunocanalicular network,which is embedded in a bone mineralized extracellular matrix.As a consequence of an applied mechanical loading,the bone deformation results in the deformation of this lacunocanalicular network inducing a shift in interstitial fluid pressure and velocity,thus resulting in osteocyte stimulation.The material environment surrounding each osteocyte,the so called perilacunar and pericellular matrices properties,define its mechanosensitivity.While this mechanical stimulation pathway is well known,the laws used to predict bone remodeling are based on strains developing at a tissue scale,suggesting that these strains are related to the shift in fluid pressure and velocity at the lacunocanalicular scale.While this relationship has been validated through observation in healthy bone,the fluid behavior at the bone-implant interface is more complex.The presence of the implant modifies fluid behavior,so that for the same strain at a tissue scale,the shift in fluid pressure and velocity will be different than in a healthy bone tissue.In that context,new markers for bone mechanical integrity,considering fluid behavior,have to be defined.The viewpoint exposed by the authors indicates that the properties of the pericellular and the perilacunar matrices have to be systematically investigated

BACKGROUND Secondary forms and hypertension-mediated organ damage(HMOD) may differ between younger and older hypertensive patients.The aim of the present study was to explore the specificity of HMOD and secondary forms in patients≥ 65 years in comparison to younger ones in a contemporary cohort.METHODS We analysed 938 patients recruited between 2004 and 2014(Cardiology department,Croix-Rousse Hospital,lyon)who had at baseline HMOD and secondary forms screening among them 190 were≥ 65 years.RESULTS The mean(2.1±0.8 vs.1.2±0.9,P 1) and frequency of HMOD(96.3% vs.72.9%,P 1) was higher in patients≥ 65 years than younger ones.Carotid femoral pulse wave velocity> 10 m/s was the most frequent HMOD in patients≥ 65 years(90.1%),while echocardiographic left ventricular hypertrophy was the most common in the younger patients(45.0%).Among ECG left ventricular indexes,only R wave in aVL lead was significantly more frequently observed in patients≥ 65 years(32.6%) than in younger ones(19.0%,P 1).The frequency of secondary hypertension was not significantly different between younger and older patients(respectively;30.5% vs.27.8%,P=0.487).The most frequent aetiology was primary aldosteronism regardless of age,followed by renovascular hypertension(6.3% vs.3.3%,P=0.038).Among older patients,3.2% were treated with adrenalectomy and 6.3% with percutaneous transluminal renal angioplasty.CONCLUSION Extensive screening of HMOD in older patients may be questionable as nearly all patients had one;aetiology must however be explored as a third of older patients had a secondary form.

Electrochemical advanced oxidation processes (EAOPs) are effective and environmentally friendly for the treatment of refractory organic pollutants.Among EAOPs,heterogeneous electro-Fenton (EF) process with in-situ formation of hydrogen peroxide (H_(2)O_(2)) is an eco-friendly,cost-effective and easy-operable technology to generate hydroxyl radicals (;OH) with high redox potential.The generation of;OH is determined by the synergistic H_(2)O_(2)formation and activation.The surface catalytic mechanisms for H_(2)O_(2)activation in the heterogeneous EF process were discussed.Some required features such as heteroatom doping and oxygen groups for H_(2)O_(2)formation via selective two-electron oxygen reduction reaction (ORR) with carbonaceous electrode are summarized.The solid Fenton catalysts and integrated functional cathodes that widely used in heterogeneous EF for wastewater treatment are grouped into few classes.And the brief discussion on catalytic activity and stability of materials over different experimental conditions are given.In addition,the application of heterogeneous EF process on the remediation of emerging contaminants is provided.The challenges and future prospects of the heterogeneous EF processes about catalytic fall-off and multi-step/complex techniques for water purification are emphasized.
