Candida albicans isolated from urine:Phenotypic and molecular identification,virulence factors and antifungal susceptibility

Objective: To isolate Candida albicans(C. albicans) from the urine of hospitalized patients and assess the virulence factors and antifungal susceptibility profiles of the ***: Yeasts were identified using the chromogenic medium CHROMagar?, the VITEK~?2 system, hypertonic Sabouraud broth, tobacco agar, polymerase chain reaction,and DNA sequencing. The evaluated virulence factors were proteinase production,phospholipase production, and biofilm production on polystyrene. The broth microdilution technique was used to determine the minimum inhibitory ***: All yeasts isolated from urine were identified as C. albicans using both classical and molecular methods. Although 91.3% of the isolates showed no phospholipase activity, 56.5% showed strong proteinase activity and 91.7% produced biofilm. All microorganisms were sensitive to fluconazole, voriconazole and amphotericin B, but 56.5% of the yeasts showed resistance to ***: C. albicans isolates from urine have a high capacity for virulence and can be associated with infectious processes. Furthermore, the high percentage of isolates resistant to itraconazole is important because this antifungal agent is commonly used to treat fungal infections in the hospital environment.

This article intends to address the Solidary Economy Network of dourados--MS, Brazil. Treated as public policy since 2001, this mode of production and consumption has achieved increasing prominence on the world stage, its capacity for income generation and social inclusion. The program is developed from the parameters of the Municipal Social Assistance and Fellowship Economy and has the support of organizations such as the NGO （Non-Governmental Organization） Women in Motion, the Community Bank and SEBRAE. The research was based on survey data in the Registry, personal interview with the Superintendent of Solidary Economy. They still relied on data from the annual survey of the Department relating to the Network ECOSOL of 2007. This data allowed us to analyze the context of the movement, the target audience and the actions taken by the Department and effectively understand if the actions are having the desired results.

Common beans (Phaseolus vulgaris L.) in the rations for cattle in feedlot

To evaluate the inclusion of common beans in the diet of cattle in feedlot on characteristics of productive performance, nutrient digestibility and ruminal fermentation parameters, 15 bulls crossed were used in a completely randomized design. The animals were confined for 64 days, and 21 days of adaptation. We evaluated the inclusion common beans at levels of 0%, 13% and 26%. All diets were isocaloric and isonitrogenous with a 45:55 forage: concentrate ratio. The addition of beans in the diet did not affect animal performance, dry matter intake and feed efficiency. Inclusion bean changed the nutrient digestibility of DM, CP, EE, TC and OM, where the best digestibility was observed with the inclusion 13% and the lowest concentration of ruminal NH3-N. The general behavior of ruminal pH in relation to levels of inclusion of beans was 6.2. The inclusion of up to 13% of beans in the diet of feedlot cattle did not alter the apparent digestibility, ruminal and animal performance.

Storage of Campomanesia adamantium(Cambess.)*** Seeds:Influence of Water Content and Environmental Temperature

The present work evaluated the conservation of Campomanesia adamantium seeds under different conditions. The fruits used in the study were collected from matrices located in the city of Ponta Porã-MS, Brazil. After processing, the seeds were slowly dried to different water content levels and subsequently exposed to various environmental conditions: 25°C ± 2°C, 35% relative humidity (laboratory);16°C ± 1°C, 40% relative humidity (cold and dry);8°C ± 1°C, 35% relative humidity (refrigeration);and -18°C ± 1°C, 42% relative humidity (freezing). Each treatment was conducted for 0 (recently processed seeds, with superficial drying of 40 minutes), 30, 60, 90, 120, 150, and 180 days. Seed potential was physiologically evaluated based on radicle protrusion, percentage of normal seedlings, seedling length (primary root, shoot and total), and dry mass of the seedlings. The experimental design was a completely randomized factorial scheme with split-split plots (4 temperatures/environments × 5 water content × 7 storage periods). C. adamantium seeds tolerated a reduction in the water content to 15.3% but did not tolerate the storage period, confirming the recalcitrant behavior of the seeds.

Quality of Jatropha curcasL. Seeds Harvested at Different Stages of Fruit Maturation

The aim of this study was to evaluate the physiological quality of Jatropha curcas L. seeds harvested at different stages of fruit maturation and two threshing methods. To analyze the mechanical damage rate an experiment was carried out with five stages of fruit maturation (green, yellow, ripe, dried on the ground and dried on the plant) threshed mechanically and manually. To evaluate the physiological quality, three tests were made;germination, vigor and electrical conductivity, all of them were applied in each storage (0, 120 and 240 days after harvest) and five stages of fruit maturation. It was feasible to infer that the maturation stage of yellow, ripe and dried on the plant fruits showed the best results of the physiological quality of the Jatropha curcas L. seeds. The manual threshing is also recommended in order to preserve the physiological quality of Jatropha curcas L. seeds. When the fruits were harvested at the appropriate stage of maturation (yellow, ripe and dry fruit on the plant) and threshing without mechanical damage, the Jatropha curcas L. seeds are possible to store for 240 days without loss in their germination. Regardless of the threshing system and the maturation stage, the seeds of Jatropha curcas L. lost their vigor from 120 days after harvested, when these seeds were stored in the shadow and in an environment without control on the relative humidity and air temperature.

Drying,Storage and Osmotic Conditioning of Psidium guineense Swartz Seeds

This study aimed to evaluate the tolerance of Psidium guineense Swartz seeds to air-dry storage and priming. Desiccation tolerance was analyzed for seed moisture content of 15%, 10% and 5%. The longevity of seeds during storage was analyzed in seeds with 5% and 10% moisture content maintained in cold and dry chamber (16°C ± 1°C/40% RH), laboratory environment (25°C ± 2°C/60% RH), refrigerator (5°C ± 1°C) and freezer (-18°C ± 1°C) during 90 days, and the priming effect in seeds that were imbibed for 5 or 10 days in polyethylene glycol 6000 at osmotic potentials of -0.3 MPa, -0.5 MPa, -0.7 MPa and -1.3 MPa. The quality of seeds was evaluated after desiccation, storage, and priming by the seeds germination, fresh weight of seedlings, seedlings growth, and germination medium time. The experimental design was completely randomized, with four replications of 25 seeds each one. Although a reduction in seed germination and seedling growth was observed in seeds with 10% and 5% moisture content, drying did not cause complete loss of seed germination and seedling development. Sour guava seeds exhibited an orthodox response to desiccation tolerance and storage. Seed germination and seedling growth were reduced with increased osmotic potential of osmoconditioning. However, conditioning of 10 days increased the seeds germination and optimized the average germination time to 20 days. The seeds of Psidium guineense are able to tolerate desiccation and storage for up to 90 days at a temperature of 5°C ± 1°C and priming for 10 days is a promising technique for propagation of P. guineense.

Evaluation of DNA Extraction Methods for Detection of Leishmaniaby Polymerase Chain Reaction

There are several methods used to obtain DNA from cells;however, the quantity, integrity, and purity of DNA vary among the methods, which may interfere with the polymerase chain reaction (PCR) results. The objective was to determine the most efficient and cost-effective method that provides the best DNA yield and PCR results. Three methods of DNA isolation were compared: 20% sodium dodecyl sulfate (SDS), guanidine isothiocyanate-phenol-chloroform (GTPC), and DNA extraction using a commercial kit (GE Healthcare GenomicPrep Blood DNA Isolation KitTM). Human peripheral blood samples were inoculated with 104 promastigotes of Leishmania infantum. DNA was quantified and PCR was performed with 13A/13B primers. The results showed that a higher DNA yield was obtained using the GTPC technique (214.51 ng/μL), followed by SDS (26.16 ng/μL) and the commercial kit (10.99 ng/μL). We concluded that while all of the techniques were effective for obtaining DNA, the GTPC method provided the best yield and the brightest bands.

Diagnosis and Recommendation Integrated System(DRIS)to Assess the Nutritional State of Cotton Crop in Brazil

The diagnosis of nutritional state is an important tool to understand the nutritional requirement of plants and the influence of the nutritional balance on the yield. The proposed in this study was to apply the Diagnosis and Recommendation Integrated System (DRIS) to assess the nutritional state of cotton crop through the order of nutrient requirement and nutritional balance. Yield data from plots (average acreage 100 ha) and foliar concentrations of macro and micronutrients of cotton (Gossypium hirsutum r. latifolium) plants were stored in a database. The criterion to define the reference population consisted of plots with above-average yields + 2/3 standard deviation (over 4785 kg·ha-1 seed cotton yield). The DRIS method was efficient to evaluate the nutrition state of cotton. It was feasible to identify the order of plant requirement and nutritional balance. It was feasible to observe that when minor the Nutritional Balance Index (NBI) it’s higher the yield. It means that to reach high yield it is necessary to show low NBI. The Sulphur, Boron and Zinc showed the higher frequency as the most required nutrients by plant. The Magnesium and Manganese were the less required by the cotton plant, this means that both of them were limiting the yield by excess in the tissue.

One Pr（Ⅲ） lanthanide ion complex was initially synthesized and characterized by TGA-DSC in air atmosphere, as well as characterized by CHN elemental analysis, defining the stoichiometric ratio as Pr（DMBz）3. The gaseous products evolved during the thermal decomposition were also monitored in N2 atmosphere employing TGA/FT-IR system. A crystal structure is obtained by state-of-the-art powder X-rays diffraction methods measured in conventional laboratory equipment and refined by the Rietveld method, which defined it as a monoclinic system of the space group P21/C with a polymeric crystal structure, [Pr（DMBZ）3]n. FT-IR theoretical spectrum and time-dependent density functional theory（TD-DFT） were calculated from TGA-DSC and crystalline system data. The experimental and theoretical FT-IR spectra present a high correlation degree when the main stretching bands are compared, while the energy transfer（HOMO - LUMO） in their neighborhoods suggests the main contributions of the light-emitting states.

Study of the Process of Extraction and Refining of Pequi (Caryocar brasilienseCamb.) Oil

Pequi fruit is a characteristic of Central Brazil with high oil content. This study evaluated the extraction of oil from the fruit portions by chemical and physical methods and determined an optimal condition for neutralization and degumming of oil extracted by solvent pequi pulp. The solvent extraction of oil was better than the mechanical pressing method. The oil extracted by solvent of the pulp pequi had a higher yield compared to other portions of the fruit. The peels and impurities material of fruit has a negligible amount of oil. The oil extracted from the pulp has better commercial characteristics in relation to the oil obtained from almonds. The operating parameters (temperature and time) studied at the stage of degumming the crude oil were significant at the 5% significance. The Increase in temperature decreases the values of acidity and iodine of the degummed oil.
