Long-Term Cropping System, Tillage, and Poultry Litter Application Affect the Chemical Properties of an Alabama Ultisol

Sustainable agricultural practices have been steadily increasing in the last couple of decades. These management practices frequently involve cover crops, less or no-tillage, and organic fertilization. In this study, we evaluated the effects of cropping systems,tillage and no-tillage, and the application of poultry litter(PL) on selected soil physicochemical properties and soil test nutrients. Soil samples were collected from the topmost surface(0–5 cm) and subsurface(5–10 cm) layers. The general effect trend was PL application > no-tillage > cover crop > cropping type. There were more statistically significant(P ≤ 0.05) correlations between the 18 soil attributes at the topmost surface than at the subsurface. This could be due to the accumulation of external C inputs and nutrients by crop residues and PL application as well as the retaining effects of no-tillage on less mobile nutrient components. Because of their high mobility and volatile nature, total nitrogen(N), ammonia-N(NH_4^+-N), and nitrate-N(NO_3^--N) levels varied greatly(high standard deviations), showing no consistent patterns among the treatments. Compared to the soybean cropping system, corn, especially with the wheat cover crop, contributed more to the total carbon(C) and sulfur(S) in the topmost surface soils(0–5 cm). Poultry litter application greatly increased pH, cation exchange capacity(CEC), base saturation, magnesium(Mg), phosphorus(P), calcium(Ca),sodium(Na), potassium(K), manganese(Mn), copper(Cu), and zinc(Zn) in both soil layers. Contrast comparisons revealed that PL application had more of an effect on these soil chemical properties than no-tillage and cropping systems. These results will shed light on developing better nutrient management practices while reducing their runoff potentials.

Application of Advection-Diffusion Routing Model to Flood Wave Propagation： A Case Study on Big Piney River, Missouri USA

Flood wave propagation modeling is of critical importance to advancing water resources management and protecting human life and property. In this study, we investigated how the advection-diffusion routing model performed in flood wave propagation on a 16 km long downstream section of the Big Piney River, MO. Model performance was based on gaging station data at the upstream and downstream cross sections. We demonstrated with advection-diffusion theory that for small differences in watershed drainage area between the two river cross sections, inflow along the reach mainly contributes to the downstream hydrograph＇s rising limb and not to the falling limb. The downstream hydrograph＇s falling limb is primarily determined by the propagated flood wave originating at the upstream cross section. This research suggests the parameter for the advectiondiffusion routing model can be calibrated by fitting the hydrograph falling limb. Application of the advection diffusion model to the flood wave of January 29, 2013 supports our theoretical finding that the propagated flood wave determines the downstream cross section falling limb, and the model has good performance in our test examples.

Identification of Candidate Transcription Factor Binding Sites in the Cattle Genome

A resource that provides candidate transcription factor binding sites （TFBSs） does not currently exist for cattle. Such data is necessary, as predicted sites may serve as excellent starting locations for future omics studies to develop transcriptional regulation hypotheses. In order to generate this resource, we employed a phylogenetic footprinting approach--using sequence conservation across cattle, human and dog and position-specific scoring matrices to identify 379,333 putative TFBSs upstream of nearly 8000 Mammalian Gene Collection （MGC） annotated genes within the cattle genome. Comparisons of our predictions to known binding site loci within the PCKI, ACTA1 and G6PC promoter regions revealed 75% sensitivity for our method of discovery. Additionally, we intersected our predictions with known cattle SNP variants in dbSNP and on the lllumina BovineHD 770k and Bos 1 SNP chips, finding 7534, 444 and 346 overlaps, respectively. Due to our stringent filtering criteria, these results represent high quality predictions of putative TFBSs within the cattle genome. Impact of community public health care on treatment effect,health cognition,and self-management in patients with type 2 diabetes

Endoscopic therapy in acute recurrent pancreatitis

Endoscopic retrograde cholangiopancreatography (ERCP) has evolved from a largely diagnostic to a largely therapeutic modality. Cross-sectional imaging, such as computed tomography (CT) and magnetic resonance imaging (MRI), and less invasive endoscopy, especially endoscopic ultrasound (EUS), have largely taken over from ERCP for diagnosis. However, ERCP remains the "first line" therapeutic tool in the management of mechanical causes of acute recurrent pancreatitis, including bile duct stones (choledocholithiasis), ampullary masses (benign and malignant), congenital variants of biliary and pancreatic anatomy (e.g. pancreas divisum, choledochoceles), sphincter of Oddi dysfunction (SOD), pancreatic stones and strictures, and parasitic disorders involving the biliary tree and/or pancreatic duct (e.g Ascariasis, Clonorchiasis).

Characterization and transcriptional analyses of cDNAs encoding three trypsin- and chymotrypsin-like proteinases in CrylAb-susceptible and CrylAb-resistant strains of sugarcane borer, Diatraea saccharalis

Diatraea saccharalis is a major corn borer pest. Midgut serine proteinases are essential for insect growth and development. Alteration of midgut proteinases is re- sponsible for Bt resistance development in some species. To clone midgut trypsin and chymotrypsin cDNAs and to test if the CrylAb resistance in D. saccharalis is associ- ated with changes in midgut proteinases, total midgut tryptic and chymotryptic activities, cDNA sequences, and gene expressions of three trypsin and three chymotrypsin genes were comparatively examined between Cry 1 Ab-susceptible （Cry 1 Ab- S S） and Cry 1 Ab-resistant （Cry 1 Ab-RR） strains. Full-length cDNAs encoding three trypsin- and three chymotrypsin- like proteinases were sequenced from CrylAb-SS and CrylAb-RR larvae. These cDNAs code for active forms of midgut serine proteinases with all fimctional motifs, includ- ing signal peptide, conserved His-Asp-Ser for the catalytic triad, three pairs of cysteines for disulfide bridge configurations, and conserved substrate specificity determination residues. In general, cDNA and putative protein sequences are highly similar between CrylAb-SS and CrylAb-RR strains, except for a few nucleotide and predicted amino acid substitutions, whose function need to be further clarified. Total trypsin and chymotrypsin activities were also similar between CrylAb-SS and CrylAb-RR strains. Transcriptional levels of the trypsin and chymotrypsin genes had numerical difference between Cry 1 Ab-SS and CrylAb-RR strains, but the difference was not statistically significant. Data suggest that the development of CrylAb resistance in D. saccharalis was not significantly as- sociated with these trypsins and chymotrypsins. Results clarified the role of six midgut proteinases and provided a foundation for continuing examination of potential involvement of other midgut proteinases in Bt resistance development and other important biochemical processes.

Time lag characteristics of sap flow in seed-maize and their implications for modeling transpiration in an arid region of Northwest China

Plant capacity for water storage leads to time lags between basal stem sap flow and transpiration in various woody plants. Internal water storage depends on the sizes of woody plants. However, the changes and its influencing factors in time lags of basal stem flow during the development of herbaceous plants including crops remain unclear. A field experiment was conducted in an arid region of Northwest China to examine the time lag characteristics of sap flow in seed-maize and to calibrate the transpiration modeling. Cross-correlation analysis was used to estimate the time lags between stem sap flow and meteorological driving factors including solar radiation（R_s） and vapor pressure deficit of the air（VPD_（air））. Results indicate that the changes in seed-maize stem sap flow consistently lagged behind the changes in R_s and preceded the changes in VPD_（air） both on hourly and daily scales, suggesting that light-mediated stomatal closures drove sap flow responses. The time lag in the maize＇s sap flow differed significantly during different growth stages and the difference was potentially due to developmental changes in capacitance tissue and/or xylem during ontogenesis. The time lags between stem sap flow and R_s in both female plants and male plants corresponded to plant use of stored water and were independent of total plant water use. Time lags of sap flow were always longer in male plants than in female plants. Theoretically, dry soil may decrease the speed by which sap flow adjusts ahead of shifts in VPD_（air） in comparison with wet soil and also increase the speed by which sap flow adjusts to R_s. However, sap flow lags that were associated with R_s before irrigation and after irrigation in female plants did not shift. Time series analysis method provided better results for simulating seed-maize sap flow with advantages of allowing for fewer variables to be included. Synthesis of a Novel Imidazole Ionic Liquid Modified Mesoporous Silica SBA15 for Selective Separation and Determination of Inorganic Arsenic in Rice

A novel 1-methylimidazole ionic liquid modified SBA15 mesoporous silica(1-MIIL@SBA15) was synthesized and applied to selective separation of inorganic arsenic(iAs) in rice by dispersive solid phase extraction(DSPE), followed by hydride generation-atomic fluorescence spectrometric(HG-AFS) quantification. The prepared sorbent was characterized by FTIR, FESEM, BET and Zeta potential. Key parameters of adsorption and desorption in DSPE were optimized using standard reference material 1568 b rice flour. Under optimal conditions, the limit of detection was 8.776 ng/kg, relative standard deviation was ≤2.0%, and recoveries of iAs were in the 92.3～94.4% range. This method was successfully applied to the determination of iAs in rice. Under acidic condition, the electrostatic interaction between the positively charged 1-MIIL@SBA15 and anionic iAs played an important role in selective iAs separation, rendering this method suitable for iAs analysis.
