What Has Been Overlooked on Study of Chinese Materia Medica in the West?

Chinese materia medica （CMM）, including Chinese herbal, animal, and mineral medicine, has been widely researched in the past century for their biological and pharmacological activities. However, their mechanism and clinical efficacy studies did not always give expected results. For example, the most commonly used Chinese herb for menstrual disorders, Radix Angelicae sinensis, showed neither estrogenic nor progesteronic activity in laboratory and clinical studies. Its efficacy should not be denied simply based on such results, because it is mostly used together with other herbs in formulae. Moreover, its regulation on menstruation may take effect through other mechanisms, such as regulation of blood circulation. The key difference of Chinese medicine （CM） from conventional medicine is its unique holistic view on human body and diseases. CMM is mostly applied in clinic in the form of formulae. Study on individual CMM, simply using methods for development of conventional drugs, is unable to thoroughly reveal the power of CMM formulae. The reason may partly result from improper design due to the lack understanding about application principle of CMMs in CM, and/or to current lack of knowledge about the causes of some symptoms and diseases. This paper will introduce the importance of qi and blood in CM etiology and pathology, Zheng differentiation, formulation of CMMs, and explain why one formula can treat different diseases and one disease can be treated with different formulae. Examples in the paper will demonstrate that proper studies on Zheng and its corresponding clinically proven formulae could help scientists find new direction to explain and treat symptoms and diseases that so far modern medicine has been unable to, provided that the designer properly understands CM theories, etiology and pathology of CM, as well as modern medicine. Strategy suggestions about research methods for CMM and its formulae will be given at the end.

OBJECTIVE： The purpose of this research is to study the effect of Roucongrong （Herba Cistanches Deserticolae） on reproductive toxicity in mice in- duced by a glycoside extracted from Leigongteng （Radix et Rhizoma Tripterygii）（GRT）. METHODS： Forty-eight BALB/c mice were random-ly divided into two groups in the ratio of I ： 3, 12 in one group and 36 in the other. The 12-mouse group was the control group that was intragastrical- ly administered physiological saline for 3 weeks. The 36 mice in the other group were given 30 mg- kg ·^-1 d i GRT for 3 weeks, then randomly divided into 3 subgroups： the model group, GRI group and Roucongrong （Herba Cistanches Deserticolae） group, with 12 mice in each group. In the model group, 0.25 mL physiological saline was intragastri- cally administered; in the GRT group, GRT, 0.25 mL at 30 mg · kg ^-1· d ^-1 was intragastrically adminis- tered once a day; in the Roucongrong （Herba Cis- tanches Deserticolae） group, mice were adminis- tered Roucongrong （Herba Cistanches Deserticolae） decoction equivalent to 0.25 mL at a final dose of *** 1.d.1 crude drug （calculated as per 20 times of 0.5 g· kg^-1. d ^-1 for adults）, and GRT 0.25 mL at 30 mg- kg^-1· d^- 1 daily. After another 3 weeks of exposure, expression levels of the reproduction-re- lated genes DEAD （Asp-Glu-Ala-Asp） box polypep- tide 3, Y-linked, B-cell CLL/lymphoma 6 and Signal transducer and activator of transcription 3 were evaluated. RESULTS： After 6 weeks of GRT treatment, the sper- matogenic cell population in the convoluted tu- bule of testis was in disorder and the tubule cavity expanded. Objective： To observe the clinical effects of acupuncture at Jiaji （EX-B 2） points plus tuina manipulation for thoracic facet joint disorder, and get new clinical evidence for treatment of thoracic facet joint disorder. Methods： Totally 106 patients with thoracic facet joint disorder were randomly allocated into an observation group and a control group based on the random number table. Patients in the control group were treated by tuina manipulation, while those in the observation group were treated by acupuncture at Jiaji （EX-B 2） points before tuina manipulation. Patients in the two groups were treated once a day. The improvements of signs and symptoms and the efficacy were observed after 3 treatments. Results： After treatment, there were intra-group statistical differences in scores of 8 signs and symptoms in both groups （P〈0.01}; the score of each item in the observation group was lower than that in the control group, and there was statistical significance in the inter-group difference （P〈0.05）. The cure rate of the observation group was 50.9%, versus 26.4% in the control group, and there was a significant difference between the two groups （P〈0.05）. Conclusion： In treating thoracic facet joint disorder, acupuncture plus tuina manipulation can restore the biomechanical balance of thoracic vertebrae, fully maximized the combined effect, and significantly improve the clinical efficacy.

Objective: To explore the association of the platelet-activating factor receptor(PAFR) gene rs5938, rs313152 and rs76744145 polymorphisms with coronary heart disease(CHD) and blood stasis syndrome(BSS) of CHD in Chinese Han population. Methods: A total of 570 CHD patients(299 with BSS and 271 with non-BSS) and 317 controls were enrolled. The PAFR gene rs5938, rs313152 and rs76744145 polymorphisms were genotyped using the multiplex SNaP shot technology. The statistical analysis was conducted using a multiple variable logistic regression model. Results: Significant differences were detected in the genotypes frequency distributions of the rs5938(P0.05), between the controls and CHD patients. Individuals with an rs5938 or rs313152 mutated allele had a low risk for CHD [adjusted odds ratio(aOR)=0.35, 95% confidence interval(CI): 0.23 to 0.56, P0.05). Furthermore, the interaction effect of the rs5938 and rs313152 polymorphisms for BSS of CHD was significantly based on an aOR value associated with the combination of the rs5938 GT genotype with the rs313152 TC genotype of 0.27(95% CI: 0.1 to 0.7, P<0.01). Conclusion: The PAFR gene rs5938 or rs313152 polymorphisms might be a potential biomarker for susceptibility to CHD, especially to BSS of CHD in Chinese Han population.
