Mycophenolate mofetil (MMF) is a pro-drug for mycophenolic acid (MPA) which reduces the likelihood of allograft rejection after renal transplantation. It was known that post-dose 12-hour trough concentration of MMF will be influenced by several factors including renal function and cyclosporine (CsA) co-administration and need adjustment when used in clinical renal transplanted patients,but there is still few data in dog. The objective of this study is to investigate Post-dose 12-hour trough concentration and determine the optimal dose of MMF in renal transplanted dog. Study 1: Investigate the relationship between oral dose and Post-dose 12-hour trough concentration of MPA in healthy dogs. Study 2: Investigate the changes of post-dose 12-hour trough concentration of MPA in healthy dogs when co-administrating with CsA. Study 3: Investigate the relationship between dose and Post-dose 12-hour trough concentration of MPA in low glomerular filtration rate (GFR) model dogs with CsA co-administration. 8 Adult female beagle dogs aged 1-2 years old,weight in 1. 5-13. 5kg. Study 1: After 7 days of prier oral administration of MMF (10 mg/kg,n = 4; 30 mg/kg,n = 4),MMF was oral administrated in the same dose in day 8,and MPA blood concentrations were measured at 0,20,40,60,90,120, 240,360,480,and 720 min after oral dose. Study 2: After 7 days of prier administration of 10 mg/kg MMF with (n = 4) or without (n = 4) 10 mg/kg CsA,MMF was oral administrated in the same dose with or without CsA in day 8,and post-dose 12-hour trough concentrations were measured. Study 3: Low GFR model dogs were prepared with renal artery ligation before this study. After 7 days of prier co-administration of 10 mg/kg (n = 4) or 20 mg/kg (n = 4) MMF with 10 mg/kg CsA,MMF was oral administrated in the same dose in day 8 and 12-hour trough concentrations of MPA were measured. Over half of solar energy consists of near-infrared and a wide range of preventative mechanisms have been evolutionarily maintained in organisms to protect against effects of near-infrared. However, the biological effects of near-infrared have not been investigated in detail. Despite the essential requirement of a water-filter to imitate solar near-infrared filtered by atmospheric water, previous studies used near-infrared resources without a water-filter or a cooling system. With these methods, near-infrared energy is primarily absorbed in the superficial tissues, thus these approaches are unable to sufficiently evaluate the biological effects of solar near-infrared that reaches human tissue. We have elucidated that near-infrared (1100 - 1800 nm together with a water-filter that excludes wavelengths 1400 - 1500 nm) non-thermally affects the skin into the deeper tissues. The biological effects of near-infrared have both beneficial applications and deleterious effects. Near-infrared induces collagen and elastin stimulation, which achieves skin rejuvenation and skin tightening, and induces long-lasting vasodilation that may prevent vasospasm and be beneficial for ischemic disorders. Near-infrared also relaxes and weakens dystonic and hypertrophic muscles to reduce wrinkles and myalgia. Nearinfrared is an essential tool in cancer detection and imaging, and induces drastic non-thermal DNA damage of mitotic cells, which may be beneficial for treating cancer. Activation of stem cells by near-infrared may be useful in regenerative medicine. However, continuous near-infrared exposure induces photoaging and potentially photocarcinogenesis. Humans have protective mechanisms against near-infrared on multiple levels, including perspiration, blisters, vasodilation, hair, skin, adipose tissue, and cotton or wool clothing. Photoprotective Ability of Sunscreens against Ultraviolet, Visible Light and Near-Infrared Radiation

Despite the widespread prevalence of daily sunscreen usage, solar-induced skin damage continues to occur. We have previously reported that solar visible light and near-infrared, in addition to ultraviolet radiation, perform as aging factors and induce deleterious effects such as photoaging, vasodilation, muscle thinning, skin ptosis, photoimmunosupression and photocarcinogenesis. Despite this, most commonly used sunscreens only block ultraviolet radiation. To evaluate the complete solar-spectrum blocking ability of sunscreens produced by internationally well-known companies, a double-beam spectrophotometer was used to optically measure the transmission spectra. The spectrophotometer utilizes a unique, single monochromatic design covering a wavelength range of 240 to 2600 nm. Sunscreens (thickness, 0.1 mm, SPF50+, PA+++ or ++++) from internationally well-known companies blocked 78.8% - 99.9% of ultraviolet, 33.4% - 99.6% of visible light, and 27.0% - 76.4% of near-infrared. It can be concluded that while most commercially available sunscreens filter ultraviolet radiation, they are not effective at blocking visible light and near-infrared radiation. The results of this study imply that sunscreens that provide comprehensive photoprotection from ultraviolet through to near-infrared should be considered to prevent skin photodamage.

AIM: To investigate the clinical efficacy of leukocytapheresis (LCAP) in patients with active ulcerative colitis (UC), and to elucidate the mechanisms by determining the changes in the cytokine levels in the peripheral blood and of the functions of the peripheral blood leukocytes in these patients. METHODS: The subjects were 19 patients with active UC, with a mean clinical activity index (CAI) of 9.2. The LCAP was conducted using Cellsorba E. In each session of LCAP, 2-3 L of blood at the flow rate of 30-50 mL/min was processed. The treatment was carried out in approximately 1-h sessions, once a week, for 5-10 wk. Blood samples for determination of the cytokine levels were collected from the inflow side of the column (site of dehematization; at the start of LCAP) and outflow side of the column (at the end of LCAP). Blood samples for the determination of reactive-oxygen-producing cells were collected from the peripheral blood before and after LCAP. RESULTS: LCAP resulted in clinical improvement in all the 19 patients of UC recruited for this study. Remission (CAI: ≤4) was noted in 15 (79%) of the 19 patients. The blood level of the pro-inflammatory cytokine IL-6 was found to be decreased following treatment by LCAP, and the level of the anti-inflammatory cytokine IL-10 at the outflow side of the LCAP column was found to be significantly elevated as compared to that at the inflow side of the column. The reactive-oxygen-producing granulocytes in the peripheral blood of UC patients was increased as compared to that in healthy persons and the increase was found to be decreased following treatment by LCAP. CONCLUSION: LCAP exerted a high therapeutic efficacy in patients with active UC. Our findings suggest that LCAP is associated with enhanced production of the inhibitory cytokine IL-10 to indirectly inhibit the functions of the inflammatory leukocytes, and that inflammation is also considerably attenuated by the direct removal of reactive-oxygen-producing neutrophils fro

Protection from Near-Infrared to Prevent Skin Damage

Over half of the solar energy consists of near-infrared, and in addition to natural near-infrared, humans are increasingly exposed to artificial near-infrared from electrical appliances. Thus, we are exposed to tremendous amounts of near-infrared. Despite the wide prevalence of a variety of ultraviolet blocking materials, the necessity to protect against near-infrared has not been well recognized. To clarify the necessity to protect against near-infrared, we assessed cell viability of human fibroblast cells after water-filtered broad-spectrum near-infrared (1100 - 1800 nm together with a water-filter that excludes wavelengths 1400 - 1500 nm) treatment using 2 sets of transparent polycarbonate plates, one to block ultraviolet and the other to block both ultraviolet and near-infrared. The cell viability was significantly decreased after 10 rounds of near-infrared irradiation at 20 J/cm2 in near-infrared treated cells without a protective polycarbonate plate and near-infrared treated cells using the polycarbonate plate to block only ultraviolet. Assuming that the cell viability of the non-irradiated control to be 100, the cell viability of the near-infrared treated cells without any protection was 0.2. The cell viability of the near-infrared treated cells with the polycarbonate plate to block only ultraviolet was 0.3, whereas both ultraviolet and near-infrared protected cells retained a viability of 85.1. The results of this study indicate that protection from not only ultraviolet but also near-infrared should be considered to prevent skin damage.

0前言激光切割机是从1964年发明二氧化碳气体激光（即CO2激光）之后，以金属加工领域为中心开始得以使用，但是由于激光的输出功率较小的缘故，在薄板加工为主的钣金行业作为替代冲床的技术而发展起来的：发明CO2激光约25年后的1989年，推出了搭载千瓦级高输出功率CO2激光发生器的大型激光切割机（搭载型激光切割机）。同时在中厚板的碳钢切割领域也开始使用起来。之后，随着激光发生器及切割机、外围装置的不断改进，目前已达到25mm左右的切割板厚，从而使激光切割机与气体切割机、等离子弧切割机一样，在造船、桥梁、建筑和剪切加工等行业广泛使用。

In Vivo Improvements in Facial Appearance and in Vitro Changes in Gene Expression Using a Topical Formulation Designed to Repair Environmentally Induced DNA Damage

Background: While sunscreen has been accepted as a mainline defence against photodamage from ultraviolet, visible light and near-infrared radiation, there appears to be a lack of research into photorepair. The concept of protecting the skin during the day and repairing cellular damage at night is intuitive, yet specific strategies revolving around combinations of proven reparative active ingredients remain unelucidated. Purpose: To investigate the efficacy of a solar repair Formulation following ultraviolet and environmental exposure in order to improve overall skin health and appearance through three hypotheses: The Formulation increases expression of DNA repair mechanisms markers;The Formulation enhances overall skin appearance through reducing signs of inflammation, elevating hydration, reinforcing skin firmness and amplifying radiance;In-Vivo efficacy test results are aligned with measured gene expression changes. Methods: The Formulation (#6NIC1.V1.1-1) was tested for: In-vitro LDH cytotoxicity activity, In-vitro qPCR gene expression with and without ultraviolet exposure on a reconstructed 3-dimensional skin model, and In-Vivo efficacy study on a panel of 22 participants objectively and subjectively. Results: Skin radiance, firmness, hydration, redness, and inflammation are significantly improved after In-Vivo skin exposure to the Formulation and environmental challenges such as ultraviolet radiation. These outcomes were confirmed by in-vitro genetic testing on a reconstructed human skin model. Conclusion: The studies allowed us to identify and group results in four main skin functions that were significantly enhanced following the application of the Formulation: firmness, hydration, radiance and soothing.
