Background sARs-CoV-2 continues to mutate over time,and reports on children infected with Omicron BA.5 are *** aimed to analyze the specific symptoms of Omicron-infected children and to improve patient *** We selected 315 consecutively hospitalized children with Omicron BA.5 and 16,744 non-Omicron-infected febrile children visiting the fever clinic at our hospital between December 8 and 30,*** convulsions and body temperatures were compared between the two *** analyzed potential associations between convulsions and vaccination,and additionally evaluated the brain damage among severe Omicron-infected *** Convulsion rates(97.5%vs.4.3%,Ps(median:2.0 vs.1.6,Psignificantly differed between Omicron-infected and non-Omicron-infected febrile *** body temperatures of Omicron-infected children were significantly higher during convulsions than when they were not convulsing and those of non-Omicron-infected febrile children during convulsions(median:39.5 vs.38.2 and 38.6℃,both Psubgroups,the temperature during convulsions was proportional to the percentage of patients and significantly differed(Psubgroups(P=0.244).The convulsion frequency was lower in the 55 vaccinated children compared to the 260 non-vaccinated children(average:1.8 vs.2.1,Pse and convulsion frequency in Omicron-infected children were significantly correlated(Psevere Omicron cases had brain *** Omicron-infected children experience higher body temperatures and frequencies during convulsions than those of non-Omicron-infected febrile *** additionally found evidence of brain damage caused by infection with omicron *** and prompt fever reduction may relieve symptoms.

Novel PLCZ1 mutation caused polyspermy during in vitro fertilization

Failure of oocyte activation,including polyspermy and defects in pronuclear(PN)formation,triggers early embryonic developmental *** studies have shown that phospholipase C zeta 1(PLCZ1)mutations cause failure of PN formation following intracytoplasmic sperm injection(ICsI);however,whether PLCZ1 mutation is associated with polyspermy during in vitro fertilization(IVF)remains ***-exome sequencing(WEs)was performed to identify candidate mutations in couples with primary *** sequencing was used to validate the *** PLCZ1-mutated sperm were injected into human and mouse oocytes to explore whether PN formation was *** oocyte activation(AOA)after ICsI was performed to overcome the failure of oocyte *** identified three PLCZ1 mutations in three patients who experienced polyspermy during IVF cycles,including a novel missense mutation c.1154C>T,*** formation failure was observed during the ICsI ***,injection of multiple PLCZ1-mutated sperm induced PN formation,suggesting that the Ca2+oscillations induced by the sperm exceeded the necessary threshold for PN *** after ICsI enabled normal fertilization,and all patients achieved successful *** findings expand the mutational spectrum of PLCZ1 and suggest an important role for PLCZ1 in terms of blocking ***,this study may benefit genetic diagnoses in cases of abnormal fertilization and provide potential appropriate therapeutic measures for these patients with sperm-derived polyspermy.

Respiratory syncytial virus(RsV)is the main pathogen that causes hospitalization for acute lower respiratory tract infections(ALRIs)in *** the reopening of communities and schools,the resurgence of RsV in the COVID-19 post-pandemic era has become a major *** understand the circulation patterns and genotype variability of RsV in tianjin before and during the COVID-19 pandemic,a total of 19,531 nasopharyngeal aspirate samples from hospitalized children in tianjin from July 2017 to June 2022 were *** immunoﬂuorescence and polymerase chain reaction(PCR)were used for screening RsV-positive samples and subtyping,*** analysis of mutations in the second hypervariable region(HVR2)of the G gene was performed through sanger *** results showed that 16.46%(3215/19,531)samples were RsV positive and a delayed increase in the RsV infection rates occurred in the winter season from December 2020 to February 2021,with the average RsV-positive rate of 35.77%(519/1451).The ON1,with H258Q and H266L substitutions,and the BA9,with T290I and T312I substitutions,are dominant strains that alternately circulate every 1–2 years in tianjin,China,from July 2017 to June *** addition,novel substitutions,such as N296Y,K221T,N230K,V251A in the BA9 genotype,and L226I in the ON1 genotype,emerged during the COVID-19 *** of clinical characteristics indicated no signiﬁcant differences between RsV-A and RsV-B *** study provides a theoretical basis for clinical prevention and ***,further studies are needed to explore the regulatory mechanism of host immune responses to different lineages of ON1 and BA9 in the future.

Binary prodrug nanoassemblies combining chemotherapy and ferroptosis activation for efficient triple-negative breast cancer therapy
