Luteolin promotes neuronogenesis in hippocampus of chronic unpredictable mild stress rats and primary hippocampus of fetal rats

OBJECTIVE: To investigate the effects of luteolin on chronic unpredictable mild stress(CUMS)-induced depressive rats and corticosterone(CORT)-induced depressive primary hippocampal neurons, and to elucidate the mechanism behind the ***: The antidepressant mechanism of luteolin was studied by using CUMS rat model and primary hippocampal neurons in fetal rats. In vivo, novelty suppressed feeding, open-field and sucrose preference tests as well as Morris water maze were evaluated. The content of brain derived neurotrophic factor(BDNF), 5-hydroxytryptamine(5-HT), norepinephrine(NE), and dopamine(DA) in serum were detected by enzyme-linked immunosorbent assay. The mechanisms of luteolin were explored based on neurotrophin and hippocampal neurogenesis, and proliferation. Survival of the septotemporal axis in hippocampus was assayed using the 5-bromo-2-deoxyuridine(BrdU), the expression of BDNF,neurotrophin-3(NT-3), and nerve growth factor(NGF) in hippocampus dentate gyrus region were measured by Western-blotting. In vitro, BDNF, NT-3, tropomyosin receptor kinase B(TrkB), and phosphorylated cyclic adenosine monophosphate responsive element binding protein(p-CREB) were detected through the high content analysis(HCA) to investigate neurotrophin and ***: Induction of CUMS in rats induced depressive symptoms, while luteolin significantly enhanced sucrose consumption, decreased feeding latency, increased locomotor activity, escape latency, distance of target quadrant and regulated the content of depressive-like biomarkers. Histology analysis revealed that luteolin increased the abundance of new born neurons that had been labeled with BrdU, BrdU + neuronal nuclear antigen,and BrdU + doublecortin in septo-temporal axis of S2(mid-septal) and T3(mid-temporal). Moreover,expression of BDNF, NT-3, and NGF increased significantly in the septo-temporal axis of S2 and T3. Heart failure(HF)is a highly morbid syndrome that seriously affects the physical and mental health of patients and generates an enormous socio-economic *** addition to cardiac myocyte oxidative stress and apoptosis,which are considered mechanisms for the development of HF,alterations in cardiac energy metabolism and pathological autophagy also contribute to cardiac abnormalities and ultimately *** information regulator 1(Sirt1)and adenosine monophosphate-activated protein kinase(AMPK)are nicotinamide adenine dinucleotide(NAD+)-dependent deacetylases and phosphorylated kinases,*** play similar roles in regulating some pathological processes of the heart through regulating targets such as peroxisome proliferator-activated receptorγcoactivator 1α(PGC-1α),protein 38 mitogen-activated protein kinase(p38 MAPK),peroxisome proliferator-activated receptors(PPARs),and mammalian target of rapamycin(mTOR).We summarized the synergistic effects of Sirt1 and AMPK in the heart,and listed the traditional chinese medicine(TCM)that exhibit cardioprotective properties by modulating the Sirt1/AMPK pathway,to provide a basis for the development of Sirt1/AMPK activators or inhibitors for the treatment of HF and other cardiovascular diseases(CVDs).

OBJECTIVE: To summarize the evidence from traditional chinese medicine(TCM) practice in the treatment of acute primary headache and provide clinical practice guidance. METHODS: The guidelines were developed in accordance with the World Health Organization guideline development manual. After the establishment of steering committee, panel and the registration and protocol formulation, the evidence on TCM for acute primary headache from published guidelines, clinical evidence, and expert experience and consensus were collected. The grading of recommendations assessment, development and evaluation method was used to grade the evidence and make the recommendations. RESULTS: Based on the available evidence, the guidelines recommended three TCM herbal decoctions, six chinese patent medicines, and two kinds of external application of chinese herbal medicines. Diagnostic recommendations based on the expert experience and consensus were also included in the guidelines. CONCLUSION: TCM diagnosis and treatment of decoction, chinese patent medicine and external application for treating acute primary headache were recommended. We hope these guidelines will be helpful in standardize the TCM acute treatment of primary headache.

Clinical efficacy and safety evaluation of Buzhongyiqi pills(补中益气丸) on appetite improvement in patients with colorectal cancer receiving chemotherapy: a pilot randomized cross-over clinical trial

OBJECTIVE: To evaluate the clinical efficacy and safety of Buzhongyiqi pills (BZYQP，补中益气丸) in improving the appetite of patients with colorectal cancer (CRC) receiving *** DESIGN: A pilot， randomized， single-blind cross-over clinical trial was conducted on diagnosed stage II-IV CRC patients receiving ***: Patients were randomly assigned to either the BZYQP-placebo or placebo-BZYQP groups. The BZYQP-placebo group received BZYQP for 1-2 days before the first cycle of chemotherapy and continued until the end of the third cycle. A 7-day washout followed， after which they received a placebo until the end of the sixth cycle. The placebo-BZYQP group followed the opposite treatment order. The oral dose of BZYQP and placebo was ten pills three times daily. A total of 12 visit points were scheduled in this study， with each visit point carried out before and after each of the six cycles of chemotherapy. The Simplified Nutrition Appetite Questionnaire (SNAQ)， the European Organization for research and Treatment of Cancer Quality of Life Questionnaire (EORTC QLQ-C30， version 3.0)， and the National Cancer institute Common Terminology Criteria for Adverse Events (CTCAE， V5.0) were used to evaluate patient appetite， quality of life， and drug ***: Totally 62 patients completed the study， and baseline characteristics were balanced between the BZYQP-placebo and placebo-BZYQP groups. The primary outcome， as assessed by SNAQ scores， demonstrates a statistically significant difference between the two groups during the first three cycles of chemotherapy， with the mean SNAQ score of the BZYQP-placebo group consistently higher than that of the placebo-BZYQP group from V1 (P < 0.001). After the washout period， the SNAQ score of the BZYQP-placebo group decreased from V7， and the difference in SNAQ scores between the two groups gradually became more significant after the intersection at V9. OBJECTIVE: To investigate the efficacy of Longteng Tongluo recipe (龙藤通络方， LTTL) combined with three-step analgesia for the treatment of lung cancer pain， and the changes in serum miRNA expressions before- and after treatment with LTTL and its correlation with lung cancer pain. The possible mechanism underlying LTTL effects on the treatment of lung cancer pain was conducted. METHODS: The pilot study was conducted at the oncology ward of the Yueyang hospital and the Longhua hospital between March 2018 and October 2019. A prospective， single-blind， placebo controlled， randomized clinical trial of LTTL or placebo combined with three-step analgesia treatments were administered to 24 cancer pain patients diagnosed with lung cancer. Analgesic efficacy was investigated as the primary outcome. Equivalent morphine consumption and numerical rating scale (NRS) scores were used as the secondary outcome. In the present study， we utilized deep sequencing techniques to compare the differential miRNA expressions in serum samples obtained from two groups: the lung cancer pain treatment group (LTTL + three-step analgesia) and the control group (placebo + three-step analgesia). Next， we employed the target prediction database to investigate the target genes for differential miRNA expressions and Gene Ontology (GO) analysis along with Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis to examine the roles and the major biochemical and signaling pathways related to the differentially expressed target genes， respectively. RESULTS: LTTL treatment significantly reduces the NRS score (P = 0.021) as compared to those before treatment， along with significant reductions in the total morphine equivalent consumption (P = 0.007) and the average daily equivalent morphine consumption (P = 0.003) as opposed to the control group. The expressions of 31 miRNAs differed considerably between the two groups of patients (≥ 2 times up-modulated or down-regulated between these groups

Highly efficient discovery of the covalent Minhibitors from crude Pu-erh tea by integrating biochemical and chemoproteomic approaches

The main proteases (Mpros) are hydrolases playing essential roles in the replication of β-coronaviruses including SARS-CoV-2. Herein， a highly efficient strategy was developed for discovering the Mproinactivators from crude plant extract integrating target-based biochemical assay and chemoproteomic approaches. Firstly， Pu-erh tea was found to potently suppress SARS-CoV-2 Mproin a time-dependent manner. Next， global chemical analysis coupling with peptide-modification profiling were used to identify the cysteine-modified constituents in Pu-erh tea. The results suggested that seven constituents in Pu-erh tea could modify SARS-CoV-2 Mpro， which turned out that epigallocatechin， gallocatechin and gallic acid were the most efficacious Mproinactivators. Further investigations demonstrated that epigallocatechin and gallocatechin could inactivate SARS-CoV-2 Mprovia blocking the formation of the homodimers. Collectively， this work proposed a novel and practical strategy for highly efficient discovery of time-dependent inhibitors of SARS-CoV-2 Mprofrom plant extracts， while three constituents in Pu-erh tea have emerged as robust SARS-CoV-2 Mproinactivators.
