Characterization of Fe-containing particles in chengdu,southwest China,using single-particle aerosol mass spectrometry

Single-particle aerosol mass spectrometry was used to study the characteristics of Fecontaining particles during winter in chengdu,southwest *** mass concentrations of PM_(2.5)and PM_(10)during the study period were 64±38 and 89±49μg/m~3,respectively,and NO_(2)and particulate matter were high compared with most other regions of *** Fecontaining particles were divided into seven categories with different mass spectra,sources and aging *** highest contribution was from Fe mixed with carbonaceous components(Fe-C,23.1%)*** was more mixed with sulfate than nitrate and therefore the contribution of Fe mixed with sulfate(Fe-S,20.7%)particles was higher than that of Fe mixed with nitrate(Fe-N,12.5%)*** contributions from Fe-containing particles related to primary combustion were high in the small particle size range,whereas aged Fecontaining particles and dust-related particles were mostly found in the coarse particle size *** air masses mainly originated from the west and east of chengdu,and the corresponding PM_(2.5)concentrations were 79±36 and 55±36μg/m~3,*** west and east air masses showed stronger contributions of Fe-containing particles related to biomass burning(Fe-B)and fossil fuel combustion(Fe-C and Fe-S)particles,*** southwest area contributed the most Fe-containing *** assessments of the effects of Fe-containing particles during heavy pollution period should pay more attention to Fe-C and Fe-S ***-reduction of Fe-containing particles should consider both local emissions and short-distance transmission from the surrounding areas.

A strategy for mechanically integrating robust hydrogeltissue hybrid to promote the anti-calcification and endothelialization of bioprosthetic heart valve

Bioprosthetic heart valve(BHV)replacement has been the predo-minant treatment for severe heart valve diseases over *** clinically available BHVs are crosslinked by glutaraldehyde(GLUT),while the high toxicity of residual GLUT could initiate calcification,severe thrombosis,and delayed ***,we construed a mechanically integrating robust hydrogel-tissue hybrid to improve the performance of *** particular,recombinant humanized coilagen type Ⅲ(rhCOLⅢ),which was precisely customized with anti-coagulant and pro-endothelialization bioactivity,was first incorporated into the polyvinyl alcohol(PVA)-based hydrogel via hydrogen bond ***,tannic acid was introduced to enhance the mechanicalperfo of PVA-based hvdrogel and interfacial bonding between the hydrogel layer and bio-derived tissue due to the strong affinity for a wide range of *** vitro and in vivo experimental results confirmed that the GLUT-crosslinked BHVs modified by the robust PVA-based hydrogel embedded rhCOLII and TA possessed long-term anti-coagulant,accelerated endothelialization,mild inflammatory response and anti-calcification ***,our mechanically integrating robust hydrogel-tissue hybrid strategy showed the potential to enhance the service function and prolong the service life of the BHVs after implantation.

Immunogenic death is a form of programmed cell death that is common in the development and progression of ***,its impact on tumor progression differs depending on the location of the ***,it also plays an important role in antitumor immunity because the process of immunogenic death can release substances that activate immunity.1 Therefore,in this study,the key mechanism of mediating immunogenic death in hepatocellular carcinoma(HCC)was found from the excavation of the immunogenic death characteristics of pan-cancer and the in-depth investigation of its effects on different tumor types(Fig.1A).

Norovirus(NoV)infection is a major cause of gastroenteritis *** virus poses great challenges in developing vaccines with broad immune protection due to its genetic and antigenic *** date,there are no approved NoV vaccines for clinical ***,we aimed to develop a broad-acting quadrivalent NoV vaccine based on a chimpanzee adenovirus vector,AdC68,carrying the major capsid protein(VP1)of noroviral GI and GII *** to intramuscular(i.m.),intranasal(i.n.),or other prime-boost immunization regimens(i.m.t i.m.,i.m.t i.n.,i.n.t i.m.),AdC68-GI.1-GII.3(E1)-GII.4-GII.17(E3),administered via i.n.t *** higher titers of serum IgG antibodies and higher IgA antibodies in bronchoalveolar lavage fluid(BALF)and saliva against the four homologous VP1s in *** also significantly stimulated the production of blocking antibodies against the four *** response to re-stimulation with virus-like particles(VLP)-GI.1,VLP-GII.3,VLP-GII.4,and VLP-GII.17,the quadrivalent vaccine administered according to the i.n.t *** effectively triggered specific cell-mediated immune responses,primarily characterized by IFN-γ***,the preparation of this novel quadrivalent NoV vaccine requires only a single recombinant adenovirus to provide broad preventive immunity against the major GI/GII epidemic strains,making it a promising vaccine candidate for further development.

Transdermal treatment for malignant melanoma by aptamer-modified tetrahedral framework nucleic acid delivery of vemurafenib

Melanoma is one of the most malignant skin tumors, whose high invasion is generally associated with BRAF gene mutation. Although new chemotherapeutic drugs, such as vemurafenib, have been developed to inhibit the growth of melanoma, these drugs are usually administered intravenously or orally, resulting in toxic side effects on major tissues and organs. Tetrahedral framework nucleic acids(tFNAs) are a novel type of DNA nanostructures with excellent biocompatibility and versatility which have been proven to penetrate through skin barrier with ease. In this study, we prepared t FNAs with vemurafenib and connected DNA aptamer AS1411 at the apex of t FNAs(AS1411-tFNAs/vemurafenib). On one hand, AS1411-tFNAs/vemurafenib could kill melanoma cells by blocking the mutated BRAF gene in vitro. Compared with free vemurafenib, AS1411-tFNAs/vemurafenib had no obvious toxicity to normal cells. On the other hand,AS1411-tFNAs could transfer vemurafenib to cross through the skin barrier and permeate into tumor tissues. In vivo, transdermal delivery of AS1411-t FNAs/vemurafenib could inhibit the growth of human A375melanoma, whose inhibiting effect was stronger than intravenous administration of vemurafenib. These results demonstrated the application prospects of tFNAs combined with chemotherapeutic drugs in skin tumors.

Since the outbreak of coronavirus disease 2019(COVID-19),the entire world has been affected by the epidemic[1,2].On November 9,2021,a variant of the novel coronavirus B.1.1.529 was detected for the first time from case samples in South *** Omicron strain spread and mutated significantly *** variant has the characteristics of escape immunity,which can induce relatively extreme disease and reduce the neutralization of antibodies in vaccinators,and is more likely to be reinfected[3,4].The clinical presentation of severe acute respiratory syndrome coronavirus 2(SARSCoV-2)in children has mild symptoms and a lower risk of hospitalization and death.

Introduction:To examine the recent trends in child injury mortality in ***:Injury mortality data of 2010–2021 for children and adolescents aged 0–19 years were from the China health Statistics *** mortality disparities across urban *** locations,gender,and age groups were *** percent change(APC),average annual percent change(AAPC),and their 95%confidence intervals(95%CI)were estiamted usimg Joinpoint regression ***:The age-standardized injury mortality significantly dropped from 21.87 to 9.41 per 100,000 population among children and youth aged 0–19 years during 2010–2021,with an AAPC of−6.7%(95%CI:−8.2%,−5.2%).The urban-rural disparity and gender gap in injury mortality reduced *** 2021,drowning and road traffic crashes were the top two causes of child injury deaths,explaing 31.1%and 27.9%of total injury deaths,*** accounted for 62.3%of injury deaths among infants younger than a ***,the suicide mortality rate rose from 2.16 to 3.42 per 100,000 population between 2010 and 2021 among teenagers aged 15–19 *** analyses yielded similar ***:During 2010–2021,the injury mortality decreased significantly among Chinese children and adolescents,and the responding urbanrural disparities narrowed.

Vaccines have proven effective in protecting populations against COVID-19,including the recombinant COVID-19 vaccine(Sf9 cells),the first approved recombinant protein vaccine in *** this positive-controlled trial with 85 adult participants(Sf9 cells group:n=44;CoronaVac group:n=41),we evaluated the safety,immunogenicity,and protective effectiveness of a heterologous boost with the Sf9 cells vaccine in adults who had been vaccinated with the inactivated vaccine,and found a post-booster adverse events rate of 20.45%in the Sf9 cells group and 31.71%in the CoronaVac group(p=0.279),within 28 days after booster *** group reported any severe adverse *** the Sf9 cells vaccine booster,the geometric mean titer(GMT)of binding antibodies to the receptor-binding domain of prototype SARS-CoV-2 on day 28 post-booster was significantly higher than that induced by the CoronaVac vaccine booster(100,683.37 vs.9,451.69,p<0.001).In the Sf9 cells group,GMTs of neutralizing antibodies against pseudo SARS-CoV-2 viruses(prototype and diverse variants of concern[VOCs])increased by 22.23–75.93 folds from baseline to day 28 post-booster,while the CoronaVac group showed increases of only 3.29–10.70 ***,neutralizing antibodies against live SARS-CoV-2 viruses(prototype and diverse VOCs)increased by 68.18–192.67 folds on day 14 post-booster compared with the baseline level,significantly greater than the CoronaVac group(19.67–37.67 folds).A more robust Th1 cellular response was observed with the Sf9 cells booster on day 14 post-booster(mean IFN-γ+spot-forming cells per 2×105 peripheral blood mononuclear cells:26.66 vs.13.59).Protective effectiveness against symptomatic COVID-19 was approximately twice as high in the Sf9 cells group compared to the CoronaVac group(68.18%vs.36.59%,p=0.004).Our study findings support the high protective effectiveness of heterologous boosting with the recombinant COVID-19 vaccine(Sf9 cells)against symptomatic COVID-19 of dive

Oral squamous cell carcinoma(OSCC)is known as one of the most malignant tumors with high recurrence and fatality *** poor tumor-targeting ability of traditional chemotherapeutic drugs has been a grand challenge for anti-OSCC *** that,a large quantity of tumor associated macrophages in OSCC tissues further diminish the anti-tumor effects of these ***,we produced a therapeutic nano drug delivery system(FA-PEG-PLA-JQ1)through encapsulating JQ1[a small-molecule inhibitor of bromodomain containing protein 4(BRD4)]into the folic acid(FA)-modified nanoparticle(PEG-PLA),which could prolong the half-life of JQ1 and target the tumor *** then,JQ1 released from this nanoparticle could prevent OSCC growth inducing tumor cell apoptosis,inhibiting tumor angiogenesis and the polarization of M2 type *** conclusion,our date demonstrated the therapeutic benefits of FA-PEG-PLA-JQ1 against OSCC in vivo or in vitro,which could be a novel treatment strategy for OSCC in coming days.
