Spinal cord injury is a severe insult to the central nervous system that causes persisting neurological *** currently available treatments involve surgical,medical,and rehabilitative ***,none of these techniques can markedly reverse neurological ***,extracellular vesicles from various cell sources have been applied to different models of spinal cord injury,thereby generating new cell-free therapies for the treatment of spinal cord ***,the use of extracellular vesicles alone is still associated with some notable shortcomings,such as their uncertainty in targeting damaged spinal cord tissues and inability to provide structural support to damaged ***,this paper reviews the latest combined strategies for the use of extracellular vesicle-based technology for spinal cord injury,including the combination of extracellular vesicles with nanoparticles,exogenous drugs and/or biological scaffold materials,which facilitate the targeting ability of extracellular vesicles and the combinatorial effects with extracellular *** also highlight issues relating to the clinical transformation of these extracellular vesicle-based combination strategies for the treatment of spinal cord injury.

Relationship between dietary niacin intake and erectile dysfunction:a population-based study

Existing research on the precise link between dietary niacin intake and erectile dysfunction(ED)is ***,this study aimed to investigate the potential association between dietary niacin intake and the risk of *** logistic regression and restricted cubic splines(RCSs)were used to examine the relationship between dietary niacin intake and *** interaction analysis was performed to assess the impact of different subgroups on the study *** addition,1:1 propensity score matching(PSM)was employed to adjust for potential confounding factors,ensuring the reliability of the *** analyzed data were collected from the 2001–2004 National Health and Nutrition Examination Survey(NHANES)in the *** study encompassed 3184 adults,among whom 863 participants were identified as having *** adjustments for potential confounders,the findings revealed that higher niacin intake,specifically in the highest tertile,was associated with a decreased risk of ED compared to that in the lowest tertile,showing an odds ratio(OR)of 0.56(95%confidence interval[CI]:0.37–0.85).Analysis of dose–response curves illustrated a negative correlation between dietary niacin intake and the risk of *** and interaction analyses fortified the consistency of these ***,PSM corroborated the validity of the *** study suggests an inverse association between dietary niacin intake and the risk of ***,establishing a cause-and-effect relationship remains elusive,and defining the safe threshold of niacin intake to prevent ED requires further investigation.

BACKGROUND The neutrophil to lymphocyte ratio(NLR)has been reported as a novel predictor for atherosclerosis and car-diovascular *** study aimed to determine the effects of NLR on long-term clinical outcomes of chronic total occlusion(CTO)*** A total of 670 patients with CTO who met the inclusion criteria were included at the end of the follow-up *** were divided into tertiles according to their baseline NLR levels at admission:low(n=223),intermediate(n=223),and high(n=224).The incidence of major adverse cardiac events(MACEs)during the follow-up period,including all-cause death,nonfatal myocardial infarction(MI),or ischemia-driven revascularization,were compared among the three *** Major adverse cardiac events were observed in 27 patients(12.1%)in the low tertile,40(17.9%)in the intermediate tertile,and 61(27.2%)in the high NLR tertile(P<0.001).Kaplan-Meier analysis demonstrated a significantly higher incidence of MACE,ischemia-driven coronary revascularization,non-fatal MI,and mortality in patients within the high tertile than those in the low and intermediate groups(all P<0.001).Multivariable COX regression analysis showed that the high tertile of baseline NLR level showed a strong association with the risk of MACE(hazard ratio[HR]=2.21;95%confidence interval[CI]:1.21-4.03;P=0.009),ischemia-driven coronary revascularization(HR=3.19;95%CI:1.56-6.52;P=0.001),MI(HR=2.61;95%CI:1.35-5.03;P=0.043)and mortality(HR=3.78;95%CI:1.65-8.77;P=0.001).CONCLUSION Our findings suggest that NLR is an inexpensive and readily available biomarker that can independently pre-dict cardiovascular risk in patients with CTO.

Effect of Tanshinone IIA on High Glucose-induced Apoptosis and Oxidative Stress Damage in Vascular Endothelial Cells

Objective: The primary cause of microvascular disease in diabetic complications is long-term hyperglycemia, wherein the damage and apoptosis of vascular endothelial cells play a significant role. Sodium tanshinone IIA sulfonate(STS) has been found to have beneficial effects on cardiovascular health. This study aimed to investigate the impact of STS on high glucose-induced apoptosis and oxidative stress damage in vascular endothelial cells, as well as its potential protective mechanisms. Methods:Human umbilical vein endothelial cells(HUVECs) were divided into five groups:low-glucose group, high-glucose group, and three STS groups(STS-a, STS-b, and STS-c). The low-glucose group was incubated with DMEM low-sugar medium containing 5.5 mmol·L-1glucose, while the high-glucose group was treated with 33.3mmol·L-1glucose. The STS groups were exposed to 10, 30, and 50 μg·m L-1of STS,respectively. Each group was cultured for 72 h, and the MTT method was utilized to assess cell proliferation. Additionally, flow cytometry was employed to monitor changes in cell apoptosis and cellular oxidative stress indicators at 24, 48, and 72 h of cell culture in each group. Results: As time went on, the cell proliferation ability and apoptosis rate of each group gradually increased. The high-glucose group exhibited lower proliferation ability compared to the other groups. The STS-c group demonstrated the highest OD value for proliferation ability(24 h: 1.19±0.12; 48 h: 1.20±0.13; 72 h:1.25 ±0.12), but it was still lower than that of the low-sugar group. Notably, the high-glucose group had the highest cell apoptosis rate, while the low-glucose group had the lowest. The apoptosis rate of the STS-c group(24 h: 8.02±0.13; 48 h: 10.10±0.12;72 h: 13.18 ±0.11)% was between that of the low-glucose group and the high-glucose group, and lower than the STS-a and STS-b groups. The extremely poor prognosis of patients is largely due to hepatocyte growth factor(HGF)/MET signaling,which promotes migration and invasion of glioblastoma(IDH wildtype;GBM;WHO grade 4).1,2 Clinical trials targeting MET,the only receptor of HGF,have yielded unimpressive results in GBM.3,4 Here we found that HGF induced strong chemotaxis on GBM cells,but MET expression was extremely ***,therefore,used single-cell RNA sequencing(scRNA-seq)coupled with label-free proteome profiling to identify membrane proteins associated with HGF/MET signaling amplification in GBM and to provide a novel modulator,MPZL1,for HGF/MET-targeted therapy.
