Role of ferroptosis in fibrosis:From mechanism to potential therapy

Fibrosis,which is a manifestation of the physiological response to injury characterized by excessive accumulation of extracellular matrix components,is a ubiquitous outcome of the repair ***,in cases of repetitive or severe injury,fibrosis may become dysregulated,leading to a pathological state and organ *** recent years,a novel form of regulated cell death,referred to as ferroptosis,has been identified as a possible contributor to fibrosis;it is characterized by iron-mediated lipid *** has garnered attention due to the growing body of evidence linking ferroptosis and fibrogenesis,which is believed to be driven by underlying inflammation and immune *** the increasing interest in the relationship between ferroptosis and fibrosis,a comprehensive understanding of the precise role that ferroptosis plays in the formation of fibrotic tissue remains *** review seeks to synthesize previous research related to the *** categorized the different direct and indirect mechanisms by which ferroptosis may contribute to fibrosis into three categories:(1)iron overload toxicity;(2)ferroptosis-evoked necroinflammation,with a focus on ferroptosis and macrophage interplay;and(3)ferroptosis-associated pro-fibrotic factors and ***,the review considers the potential implications of these findings and highlights the utilization of ferroptosis-targeted therapies as a promising strategy for mitigating the progression of *** conclusion,novel anti-fibrotic treatments targeting ferroptosis could be an effective treatment for fibrosis.

Targeted protein degradation(TPD)represented by proteolysis targeting chimeras(PROTACs)marks a significant stride in drug discovery.A plethora of innovative technologies inspired by PROTAC have not only revolutionized the landscape of TPD but have the potential to unlock functionalities beyond ***-small-molecule-based approaches play an irreplaceable role in this field.A wide variety of agents spanning a broad chemical spectrum,including peptides,nucleic acids,antibodies,and even vaccines,which not only prove instrumental in overcoming the constraints of conventional small molecule entities but also provided rapidly renewing *** we summarize the burgeoning non-small molecule technological platforms inspired by PROTACs,including three major trajectories,to provide insights for the design strategies based on novel paradigms.

The concept of enhanced recovery after surgery(ERAS)has been practiced for decades and has been implemented in numerous surgical *** is a global surgical quality improvement initiative,and it is an element in the field of perioperative *** had shown significant clinical outcomes,patientreported satisfaction,and improvements in medical service *** has been developed for specific surgical procedures,but with the fast progress of newly introduced surgical procedures,the original ERAS have been developed and *** appearing Topics and future research trends encompass ERAS protocols for other types of surgery and the enhancement of perioperative status,including but not limited to pediatric surgery,laparoscopic and robotic assisted surgery,bariatric surgery,thoracic surgery,and renal *** elements and pathways of ERAS have been developed with the introduction of up-to-date methodologies in the pre-operative,operative,and post-operative *** costs are higher than traditional care,but the patient’s clinical outcome and satisfaction are *** is in progress in the fields of anesthetic tasks,pediatric surgery,and organ *** ERAS has shown significant clinical outcomes,there are needs to modify the protocol for specific cases,hospital facilities,resources,and nurses training on elements of *** challenges and limitations exist in the implementation of ERAS that deserve consideration,it includes:Frailty,maximizing nutrition,prehabilitation,treating preoperative anemia,and enhancing ERAS adoption globally are all included.

How Agent Orange impacts prostate cancer risk,pathology,and treatment outcomes

Between 2.6 and 3.8 million veterans served in Vietnam while the US military dispersed Agent Orange(AO),although the exact number of exposed individuals is *** Orange,an herbicide,is a known risk factor for various cancers,including sarcoma and leukemia,but less is known about its link with prostate cancer(PC).Prostate cancer is the most commonly diagnosed malignancy in men and the fifth most common cause of cancer-related death in men *** 2023,approximately 288,300 patients will be given a diagnosis of PC,and an estimated 34,700 fatalities will occur in the United ***,whether the pathologic characteristics of PC among those exposed to AO differ from those in the general population remains *** review synthesizes the literature regarding the impact of AO exposure on PC incidence and disease course.A comprehensive PubMed literature search of articles published beginning in 1950 was performed using the primary search terms“Agent Orange,”“TCDD,”and“tetrachlorodibenzodioxin”and the secondary search terms“prostate cancer”or“prostate neoplasm.”The search was limited to studies that focused on human participants and were published in *** authors thoroughly reviewed the retrieved articles for relevancy to the study aims:discussion of PC diagnosis,prognosis,or management among patients exposed to *** 108 studies identified in our search,13 were included in this systematic *** within studies concerning AO exposure with relation to PC incidence,age at diagnosis or treatment initiation,and PC severity seemed to be mixed and generally ***,the literature seems to indicate that there are no significant differences in survivorship between exposed and unexposed veterans who are given a diagnosis of *** these heterogeneous outcomes,the evidence does not encourage a significantly different approach to the diagnosis and management of PC for veterans exposed to *** shouldmake case-by-case decision

Updated prevalence of latent prostate cancer in Chinese population and comparison of biopsy results: An autopsy-based study

Prostate cancer detected by autopsy is named latent prostate *** the repertoire of clinical prostate cancer,latent cancer may better reflect the disease *** clinical prostate specimens,which are obtained exclusively from biopsy-positive cases,prostate specimens obtained through autopsy provide information on biopsy-negative cases,helping calculate the true sensitivity of prostate *** 2014 to 2021,we collected autopsy specimens of the prostate from body donors in China and performed transperineal and transrectal biopsies on specimens before step-sectioning and pathological *** found that the crude prevalence of latent prostate cancer in middle-aged and elderly men was 35.1%(81/231),which was higher than previous estimates for Chinese *** overall per-patient sensitivities of transperineal and transrectal biopsies were not significantly different(33.3%vs.32.1%,p=0.82),but the two approaches differed in preferential sampling area along the proximal-distal axis of the *** biopsy had a higher sensitivity for detecting clinically significant lesions in the distal third(34.7%vs.16.3%,p=0.02)and distal half(30.6%vs.18.1%,p=0.04),while transrectal biopsy had a higher sensitivity for lesions in the proximal half(25.0%vs.13.9%,p=0.046).Both transperineal and transrectal methods of biopsy missed most small lesions(0.5 mL).In conclusion,the prevalence of latent prostate cancer in China has increased over the past 2 *** transperineal and transrectal methods of biopsy had comparable sensitivities but had different preferential sampling *** approaches miss one-third of large lesions.
