Transcriptomic analysis identifies a pan-cancer association of IL27 expression with cancer prognosis and immune microenvironment

Nowadays great progress has been made in targeted therapy and comprehensive therapy for a few cancers. But the prognosis of the vast majority of cancer patients is still weakness. It has been proven that inflammation can affect the development and treatment of cancer. Some previous studies have suggested that inflammation-related interleukins (ILs) play a significant role in the development of some cancers.1,2 ILs act as a considerable role in immunity,3 and inflammation affects the development of cancer. therefore, the expression profile of IL gene may also be one of the vital markers of early pathological changes, occurrence and progression of cancer.4,5 Currently, studies on the expression of IL27 in cancer are still extremely limited. Our work comprehensively and systematically studied the IL27 gene expression in the generic cancer when compared with the existing IL27 gene related research experiment. We screened out cancer species linked with poor prognosis, and explored the correlation between IL27 gene expression and immune cell infiltration and immune microenvironment in these cancers further, providing an increasing number of the direct basis for subsequent studies. Our findings showed that IL27 can be used to assess prognosis and immune infiltration in patients with colon adenocarcinoma (COAD), glioblastoma multiforme (GBM), head and neck squamous cell carcinoma (HNSC), skin cutaneous melanoma (SKCM) and testicular germ cell tumors (TGCT) as a biomarker.

AIM:To evaluate the SF-36, Diabetes Specificity Quality of Life Scale (DSQL) and anxiety and depression symptoms and investigate its changes in proliferative diabetic retinopathy (PDR) by vitrectomy interventions. MEthODS:the present study included 108 diabetic retinopathy (DR) patients:54 with PDR and 54 with non-proliferative diabetic retinopathy (NPDR). Each healthy control group (n =54) sociodemographically matched to DR groups was established respectively. the quality of life, anxiety and depression symptoms were evaluated and analyzed on preoperative and postoperative month 1 using SF-36, DSQL and hospital Anxiety and Depression Scale (HADS). · RESULTS:DR patients described impaired HRQL (Health Related Quality of life, SF -36) in 6 out of 8 subscales, including ‘Body Health’, ‘Body RoleFunction’,‘General Health’,‘Society Function’,‘Emotion Role Function’and‘Mental Health’. Compared with controls, DR patients (NPDR and PDR) suffered from statistically significantly impaired HRQL (SF-36 Summary score) (Pthe anxiety and depression score were significantly reduced, while the health and quality of life (SF-36 Summary scores and DSQL scores) was improved in patients with PDR (P <0.05). CONCLUSION:DR patients were affected in mentation and quality of life. Surgery interventions can improve SF-36, DSQL, anxiety and depression in PDR patients.

Epithelial ovarian carcinoma （EOC） is the most common form of ovarian malignancies and the most lethal gynecologic malignancy in the United States. To date, in spite of treatment to it with the extensive surgical debulking and chemotherapy, the prognosis of EOC remains dismal. Recently, it has become increasingly clear that in many instances, the signaling and molecular players that control development are the same, and when inappropriately regulated, drive tumorigenesis and cancer development. Here, we discuss the possible involvement of Hedgehog （Hh） pathway in the cellular regulation and development of cancer in the ovaries. Using the in vitro and in vivo assays developed has facilitated the dissection of the mechanisms behind Hh-driven ovarian cancers formation and growth. Based on recent studies, we propose that the inhibition of Hh signaling may interfere with spheroid-like structures in ovarian cancers. the components of the Hh signaling may provide novel drug targets, which could be explored as crucial combinatorial strategies for the treatment of ovarian cancers.

Objective:To determine the neuroprotective effects of apigenin against streptozotocin(STZ)-induced diabetic neuropathy(DN).Methods:To induce DN,Wistar rats(150-200 g)were administered with STZ(55 mg/kg,i.p.).then they were randomly assigned to various groups,viz.,normal,diabetic control,insulin(10 IU/kg,s.c.),apigenin(5,10,and 20 mg/kg,p.o.),and insulin(10 IU/kg)plus apigenin(20 mg/kg,p.o.).Various behavioral,biochemical,and molecular markers[tumor necrosis factor-alpha(TNF-α),interleukin(IL)-1β,IL-6,Toll-like receptor 4(TLR4),myeloid differentiation primary response 88(MyD88),and nuclear factor erythroid 2-related factor 2(Nrf2)]were ***:Apigenin(10 and 20 mg/kg,p.o.)substantially reduced plasma glucose levels,lipid profile,aspartate transaminase,alanine transaminase,glycated hemoglobin,and neural advanced glycation end products in STZ-induced DN rats(Pthione)and Na+K+ATPase activity were also considerably elevated(Pthe level of lipid peroxidation was diminished following apigenin therapy(Pthermore,apigenin markedly upregulated the Nrf2 mRNA level while downregulating the mRNA expressions of TNF-αand ILs and the protein expressions of TLR4 and MyD88(Pthe sciatic nerve were also improved by apigenin ***:Apigenin exerts its neuroprotective effect by modulating the inflammatory and oxidative stress pathways via regulating the TLR4-MyD88 signaling pathway.

Background:Nasogastric tube(NGT)placement is part of the post-operative management of upper gastrointestinal perforation,but its routine use in esophageal perforation(EP)caused by foreign bodies remains *** purpose of this research was to investigate the necessity for routine NGT placement in patients with EP after endoscopic foreign body ***:A total of 323 patients diagnosed with EP caused by foreign bodies at the First affiliated hospital of nanchang university between January 2012 and December 2021 were included in this retrospective *** were divided into the NGT group and the non-NGT group according to whether or not NGT placement was *** perforation healing rate,post-operative adverse events,hospital stay,and death rate were analysed using a 1:1 propensity score matching ***:Before matching,there were 263 patients in the NGT group and 60 patients in the non-NGT *** were significant differences in the time to treatment,infection,albumin,and types of endoscopy between the two groups,while the length of hospital stay in the NGT group was significantly longer than that in the non-NGT *** 1:1 propensity score matching,48 pairs of patients were matched between the two *** perforation healing rate,post-operative adverse events,length of hospital stay,and death rate did not show significant differences between the two ***:For patients with small EP caused by foreign bodies,routine NGT placement after endoscopic foreign body removal may be unnecessary.

AIM: To determine the effect of different concentrations of the acetylcholinesterase (AChE) inhibitors tacrine and donepezil on retinal protection in AChE(+/-) mice (AChE knockout mice) of various ages. MEthODS: Cultured ARPE -19 cells were treated with hydrogen peroxide (H2O2) at concentrations of 0, 250, 500, 1000 and 2000 mu mol/L and protein levels were measured using Western blot. Intraperitoneal injections of tacrine and donepezil (0.1 mg/mL, 0.2 mg/mL and 0.4 mg/mL) were respectively given to AChE4- mice aged 2mo and 4mo and wild-type S129 mice for 7d; phosphate buffered saline (PBS) was administered to the control group. the mice were sacrificed after 30d by cardiac perfusion and retinal samples were taken. AChE(+/-) deficient mice were identified by polymerase chain reaction (PCR) analysis using specific genotyping protocols obtained from the Jackson Laboratory website. H&E staining, immunofluorescence and Western blot were performed to observe AChE protein expression changes in the retinal pigment epithelial (RPE) cell layer. RESULTS: Different concentrations of H2O2 induced AChE expression during RPE cell apoptosis. AChE(+/-) mice retina were thinner than those in wild -type mice (Pthe retinal structure was still intact at 2mo but became thinner with increasing age(P thermore, AChE'l- mice developed more slowly than wild-type mice (P the protection of the retina function and morphology (P >0.05). CONCLUSION: in viva, tacrine and donepezil can inhibit the expression of AChE; the decrease of AChE expression in the retina is beneficial for the development of the retina.
