AIM： To demonstrate the changes in ultrastructure and histopathology of the cornea in acute corneal alkaline burns after femtosecond laser-assisted deep lamellar keratoplasty.·MEthODs： the New Zealand white rabbits treated with alkaline corneal burn were randomized into two groups,Group A（16 eyes） with femtosecond laser-assisted deep lamellar keratoplasty 24 h after burn and Group B（16 eyes）without keratoplasty as controls. All eyes were evaluated with transmission electron microscopy（TEM） at 1, 2, 3,and 4wk follow-up, then all corneas were tested by hematoxylin and eosin staining histology.· REsULTs： the corneal grafts in Group A were transparent, while those in Group B showed corneal stromal edema and loosely arranged collagen fibers. One week after treatment, TEM revealed the intercellular desmosomes in the epithelial layers and intact non-dissolving nuclei in Group A. At week 4, the center of the corneas in Group A was transparent with regularly arranged collagen fibers and fibroblasts in the stroma. In Group B, squamous cells were observed on the corneal surface and some epithelial cells were detached.· CONCLUsION： Femtosecond laser-assisted deep lamellar keratoplasty can suppress inflammatory responses, prevent toxic substance-induced injury to the corneal endothelium and inner tissues with quicker recovery and better visual outcomes.

OBJECTIVE: To study the anti-tumor effects of the extracts from Huangqi(Radix Astragali Mongolici)and Ezhu(Rhizoma Curcumae Phaeocaulis) on the growth of Lewis lung carcinoma(LLC) in a xenograft mouse model and to investigate the possible underlying ***: LLC tumor-bearing C57 BL/6 mice were treated with normal saline, cisplatin(2 mg/kg intraperitoneally every other day), or Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis)(1∶1, 2∶1, or 3∶1 ratio;5, 8, or 11 g/kg crude drug intragastrically every day) for 15 *** weights and tumor volumes were measured every other day. Tumors were excised on day 15 and analyzed. Tumor microvessel density(MVD)was assessed by immunohistochemical staining of CD34;and expression of vascular endothelial cell growth factor(VEGF), the mitogen-activated protein kinases p38 mitogen-activated protein kinase(MAPK), extracellular signal-regulated kinases 1 and 2(ERK1/2), and Jun N-terminal kinase(JNK)and their phosphorylated forms were assessed by Western ***: Treatment with cisplatin caused a significant loss of body weight compared with controls,whereas Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) extract combinations had no effect. Extracts from Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) significantly decreased tumor weight and tumor MVD compared with controls,and at the 3∶1 treatment group had similar efficacy to cisplatin in reducing MVD. AIM: To investigate the effect of biliary drainage on inducible nitric oxide synthase (iNOs), CD14 and TGR5 expression in rats with obstructive jaundice (OJ). MEthODs: Male adult sprague-Dawley rats were randomly assigned to four groups: OJ, sham operation (sH), internal biliary drainage (ID) and external biliary drainage (ED). Rat models were successfully established by two operations and succumbed for extraction of Kupffer cells (KCs) and liver tissue collection on the 8 th and 15 th day. KCs were isolated by in situ hepatic perfusion and digested with collagen Ⅳ, density gradient centrifuged by percoll reagent and purified by cell culture attachment. the isolated KCs were cultured with the endotoxin lipopolysaccharide (LPs) with and without the addition of ursodeoxycholic acid (UDCA). the expression of iNOs, CD14 and bile acid receptor-TGR5 protein in rat liver tissues was determined by immunohistochemistry. the expression of iNOs and CD14 messenger RNA (mRNA) on the isolated KCs was detected by reverse transcription polymerase chain reaction (PCR) and the TGR5 mRNA level in KCs was measured by real-time quantitative PCR. REsULTs: the iNOs protein was markedly expressed in the liver of OJ rats, but rare expressed in sH rats. After relief of OJ, the iNOs expression was decidedly suppressed in the ID group (ID vs OJ, P sly increased in rats of ED (ED vs OJ, P=0.004). When interfered only with LPs, the expression of iNOs mRNA by KCs was increased in the OJ group compared with the sH group (P=0.004). After relief of biliary obstruction, the iNOs mRNA expression showed slight changes in the ED group (ED vs OJ, P=0.71), but dropped in the ID group (ID vs OJ, P=0.001). Compared with the simple intervention with LPs, the expressions of iNOs mRNA were significantly inhibited in all four groups after interfered with both LPs and UDCA (P spectively). OBJECTIVE: To investigate the effect of optimal combination(E) of Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) on proliferation and apoptosis of A549 lung cancer cells and the possible mechanism underpinning the ***: A uniform design method was used to optimize the E of Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) in A549 lung cancer cells. MTs assay was applied to analyze the effect of the component formula ofHuangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) on A549 cells viability in various uniform design groups. A549 cells with exponential growth in routine culture were exposed to Co Cl_2(200 μmol/L) to mimic hypoxic conditions. Group 0 was treated with RPMI-1640, the group Co Cl_2 was treated with Co Cl_2(200 μmol/L),the group DDP + Co Cl_2 was treated with 4 mg/L Cisplatin injection(DDP) + Co Cl_2(200 μmol/L), and the drug group was treated with various dose of E(0.5 E, 1 E, 2 E) + Co Cl_2(200 μmol/L). All groups were cultured for 24 h. Cell apoptosis was measured by Annexin V-FITC/propidium iodide double staining and flow cytometry. Western blot assay and quantitative real-time polymerase chain reaction(q RT-PCR) were employed to detect the protein and m RNA expression of B-celllymphoma-2(Bcl-2),Bcl-2-associated X protein(Bax) and cysteinyl aspartate specific proteinase-3(caspase-3).REsULTs: the E obtained by the uniform design was comprise of 200 mg/L Astragalus polysaccharide(X1) and 32 mg/L Curcumin(X3). Group DDP+Co Cl_2, group 1 E + Co Cl_2 and group 2 E + Co Cl_2 promoted the apoptosis of A549 cells(P statistically significant differences compared with the group DDP + Co Cl_2(P > 0.05). BACKGROUND Adrenocorticotropic hormone(ACth)-independent Cushing's syndrome(Cs) is mostly due to unilateral tumors, with bilateral tumors rarely reported. Its common causes include primary pigmented nodular adrenocortical disease,ACth-independent macronodular adrenal hyperplasia, and bilateral adrenocortical adenomas(BAAs) or carcinomas. BAAs causing ACthindependent Cs are rare; up to now, fewer than 40 BAA cases have been reported. the accurate diagnosis and evaluation of BAAs are critical for determining optimal treatment options. Adrenal vein sampling(AVs) is a good way to diagnose ACth-independent *** sUMMARY A 31-year-old woman had a typical appearance of Cs. the oral glucose tolerance test showed impaired glucose tolerance and obviously increased insulin and Cpeptide levels. Her baseline serum cortisol and urine free cortisol were elevated and did not show either a circadian rhythm or suppression with dexamethasone administration. the peripheral 1-deamino-8-D-arginine-vasopressin(DDVAP)stimulation test showed a delay of the peak level, which was 1.05 times as high as the baseline level. Bilateral AVs results suggested the possibility of *** computed tomography showed bilateral adrenal adenomas with atrophic adrenal glands(right: 3.1 cm × 2.0 cm × 1.9 cm; left: 2.2 cm × 1.9 cm × 2.1 cm). Magnetic resonance imaging of the pituitary gland demonstrated normal findings. A left adenomectomy by retroperitoneoscopy was performed first,followed by resection of the right-side adrenal mass 3 mo later. Biopsy results of both adenomas showed cortical tumors. Evaluations of ACth and cortisol showed a significant decrease after left adenomectomy but could still not be suppressed, and the circadian rhythm was absent. Radiotherapy is a common strategy in treating lung *** evidence suggested that radiotherapy has the potential to promote the metastasis and invasion of carcinoma *** this study,we aimed to testify the role of vascular endothelial growth factor(VEGF)and C-C chemokine receptor-7(CCR-7)in the metastasis of human adenocarcinoma A549 cells in vivo and in *** mice were injected with A549 cells irradiated by 0,2 and 4 Gy X-rays,*** detections of VEGF-A/C and CCR-7 mRNA from lung sample were performed by real-time *** interfering RNA(siRNA)transfection technology was further used to testify the role of the genes in the metastasis of A549 *** and CCR-7mRNA could only be detected 10 weeks post injection in vivo when visible tumor foci scattered in *** addition,VEGF-A/C mRNA expressed significantly higher in mice injected with A549 cells irradiated by 2 and 4 X-rays than those with 0 Gy X-rays *** the other hand,A549 cells with or without X-rays irradiation transfected with VEGF siRAN and CCR-7 siRNA showed a dramatic decrease of invasiveness compared to normal A549 cells with or without *** migration indexes were?0.7,?0.48,?0.34 and?0.32 for A549 cells with CCR-7 siRNA,VEGF siRNA,X-rays combined CCR-7 siRNA and X-rays combined VEGF-siRNA *** results demonstrated that X-rays could promote the metastasis of A549 cells,and VEGF-A/C and CCR-7 mRNA were closely related to the metastasis of A549 cells in vivo and in vitro.

Risk of cardiac rupture after acute myocardial infarction is related to a risk of hemorrhage

Although cardiac rupture (CR) is a fatal mechanical complication of acute myocardial infarction (AMI), to date no predictive model for CR has been described. CR has common pathological characteristics with major bleeding. We aimed to investigate the relationship between the risk factors of major bleeding and CR. A total of 10 202 consecutive AMI patients were recruited, and mechanical complications occurred in 72 patients. AMI patients without CR were chosen as control group. Clinical characteristics including bleeding-related factors were compared between the groups. the incidences of free wall rupture (FWR), ventricular septal rupture (VsR), and papillary muscle rupture (PMR) were 0.39%, 0.21%, and 0.09%, respectively, and the hospital mortalities were 92.5%, 45.5%, and 10.0%, respectively. Female proportion and average age were significantly higher in the groups of FWR and VsR than in the control group (Ps (Pthe control group, patients with CR were more likely to receive an administration of thrombolysis [26.39% vs. 13.19%, Pss likely to be treated with primary percutaneous coronary intervention (PCI) [41.67% vs. 81.60%, Pthe major bleeding scores (integer scores) of FWR, VsR, and PMR were (17.70±7.24), (21.91±8.33), and (18.60±7.88), respectively, and were significantly higher than that of the control group (11.72±7.71) (Pssion analysis identified age, increased heart rate, anemia, higher white blood cell count, and thrombolysis as independent risk factors of CR, most of which were major bleeding-related factors. the patients with CR have a significantly higher risk of hemorrhage compared to the group without CR. Risk of CR after AMI is related to the risk of hemorrhage.

Background Left ventricular(LV)remodeling is the most common target organ damage in ***,our study found that plasma microRNA-29a(miR-29a)level was associated with the LV remodeling in hypertensive ***,the causal relationship between miR-29a and LV remodeling remains ***,the aim of this study was to investigate the regulation mechanism of miR-29a in LV ***&Results Overexpression and knockdown miR-29a mice were generated by tail-intravenous injection of miR-29a-mimic and inhibitor lentivirus for one week *** the mice were subjected to angiotensin-II(AngII)induced LV remodeling by subcutaneous AngII capsule osmotic pumping into AngII for four ***-induced LV remodeling mice as the model group(n=9).Age-matched male sPF C57/BL6J mice(6–8 weeks old)were treated with the pumping of saline as a vehicle(n=6).In vivo,overexpression miR-29a ameliorated AngII-induced LV remodeling,while knockdown miR-29a deteriorated LV ***,we observed that overexpression miR-29a mice inhibited but knockdown miR-29a mice increased cardiac cross-sectional area,indicating that miR-29a has an antagonistic effect on cardiac *** studies found that overexpression miR-29a inhibited the content of the LV collagen including collagen I and ***,the expression of transforming growth factor-β(TGF-β)and phosphorylated sMAD2/3 decreased with the down-regulation of collagen I and III in overexpression miR-29a *** Our finding indicates that overexpression miR-29a attenuates LV remodeling by inhibiting collagen deposition,TGF-β,and phosphorylated sMAD2/3 ***,intervention miR-29a may be a therapeutic target for attenuating LV remodeling.

OBJECTIVE: To evaluate the effect of heat stimulation via scar-producing moxibustion at the acupoints Zusanli(sT 36) and Feishu(BL 13) on the neutrophil-to-lymphocyte ratio(NLR) and quality of life in patients with non-small-cell lung cancer(NsCLC).MEthODs: seventy patients with NsCLC were randomly assigned into two groups: group A received scar-producing moxibustion at the acupoints Zusanli(sT 36) and Feishu(BL 13) every day for 6 weeks, while group B received no intervention(control group). Outcome measures were the NLR and the scores from the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire(EORTC QLQ-C30). the NLR and the EORTC QLQ-C30 were assessed at baseline and at the end of 6 ***: Five participants dropped out, leaving afinal total of 65 participants who completed the trial. Groups A and B had a similar mean NLR at baseline. After the treatment course, the NLR in group A was significantly lower than that in group B(P th group B, the EORTC QLQ-C30 scores in group A were significantly greater in terms of global health status or quality of life(P significantly lower in terms of symptoms(P sION: the present study suggests that performing scar-producing moxibustion by heat-stimulating the acupoints Zusanli(sT 36) and Feishu(BL13) effectively decreases the NLR and improves the quality of life in patients with NsCLC.
