Genetic diversity and phylogenetic analysis of EG95 sequences of Echinococcus granulosus:Implications for EG95 vaccine application

Objective:To analyse the genetic variability of EG95 sequences and provide guidance for EG95 vaccine application against Echinococcus granulosus(E. granulosus). Methods:We analysed EG95 polymorphism by collecting total 97 different E. granulosus isolates from 12 different host species that originated from 10 different countries. Multiple sequence alignments and the homology were performed by Lasergene 1(DNASTAR Inc.,Madison,WI),and the phylogenetic analysis was performed by using MEGA5.1(CEMI,Tempe,AZ,USA). In addition,linear and conformational epitopes were analysed,including secondary structure,NXT/S glycosylation,fibronectin type ecoⅢ(Fnndary Ⅲ) domain and glycosylphosphatidylinositol anchor signal(GPIanchor). The s structure was predicted by PSIPRED method. Results:Our results indicated that most isolates overall shared 72.6-100% identity in EG95 gene sequence with the published standard EG95 sequence,X90928. However,EG95 gene indeed has polymorphism in different isolates. Phylogenetic analysis showed that different isolates could be divided into three subgroups. Subgroup 1 contained 87 isolates while Subgroup 2 and Subgroup 3 consisted of 3 and 7 isolates,respectively. Four sequences cloned from oncosphere shared a high identity with the parental sequence of the current vaccine,X90928,and they belonged to Subgroup 1. However,in comparison to X90928,several amino acid mutations occurred in most isolates besides oncosphere,which potentially altered the immunodominant linear epitopes,glycosylation sites and secondary structures in EG95 genes. All these variations might change their previous antigenicity and thereby affecting the efficacy of current EG95 vaccine. Conclusions:This study reveals the genetic variability of EG95 sequences in different E. granulosus isolates,and proposed that more vaccination trials would be needed to test the effectiveness of current EG95 vaccine against distinct isolates in different countries.

Heterodera avenae （cereal cyst nematode, CCN） infects many cereal crops and causes serious yield losses worldwide. Interaction studies investigating H. avenae and its hosts are still in their infancy. In this study, a barley model plant, the Hordeum vulgare cultivar Golden Promise, was investigated for its potential as a candidate model host to study its inter- action with H. avenae. CCN-infective juveniles were attracted by the root tips and gathered around the root elongation zones of Golden Promise on 0.7% water agar plates. The juveniles invaded the roots and developed successfully until maturation at 40 days after inoculation in sterile sand soil. The cryotomy and syncytium measurements indicated that the syncytia enlarged gradually throughout the development of the nematodes and caused the corresponding root regions to swell obviously. Quantitative real-time PCR analysis showed that the down-regulation of defence-related barley genes and up-regulation Of development-related barley ger^es contribute to the understanding of compatible interaction between H. avenae and Golden Promise. Barley stripe mosaic virus （BSMV） virus-induced gene silencing （VIGS） can be used in the roots of Golden Promise. In conclusion, the Hordeum vulgare cultivar Golden Promise is a suitable candidate model host for interaction studies with Heterodera avenae. The studies presented above document the first CCN host that no.t only has published genome context but also be compatible to BSMV VIGS.

Systems model-guided rice yield improvements based on genes controlling source, sink, and flow

A large number of genes related to source, sink,and flow have been identified after decades of research in plant genetics. Unfortunately, these genes have not been effectively utilized in modern crop breeding. This perspective paper aims to examine the reasons behind such a phenomenon and propose a strategy to resolve this situation. Specifically, we first systematically survey the currently cloned genes related to source, sink, and flow;then we discuss three factors hindering effective application of these identified genes, which include the lack of effective methods to identify limiting or critical steps in a signaling network, the misplacement of emphasis on properties, at the leaf, instead of the whole canopy level,and the non-linear complex interaction between source,sink, and flow. Finally, we propose the development of systems models of source, sink and flow, together with a detailed simulation of interactions between them and their surrounding environments, to guide effective use of the identified elements in modern rice breeding. These systems models will contribute directly to the definition of crop ideotype and also identification of critical features and parameters that limit the yield potential in current cultivars.

Earliest fossil fruit record of the genus Paliurus(Rhamnaceae) in Eastern Asia

The genus Paliurus is characterized by its distinctive orbicular-winged fruits and a trilocular(or bilocular) ovary. Macrofossil records suggested this genus was distributed widely in North America, Europe, and Asia during the Paleogene and Neogene, before its present limitation to Eurasia. In this study, we describe some Paliurus fossil winged fruits from the lower part of Youganwo Formation(middle Eocene) and Huangniuling Formation(late Eocene) of the Maoming Basin, South China. These fruits are recognized as Paliurus favonii Unger based on a detailed study on the fossil morphology and cuticle characteristics. This represents the earliest occurrence of Paliurus fossil fruits in eastern Asia, and to date is the world's lowest latitude appearance for this genus. The most ancient records of Paliurus throughout the world are from the middle Eocene of North America and the late Eocene of Japan, leading some researchers to conclude the Paliurus may have originated in North America. Molecular data, however, do not support this hypothesis. The present Paliurus fossil fruits provide new evidence for the origin and phytogeographic history of this genus.

Targeting of anti-microbial proteins to the hyphal surface amplifies protection of crop plants against Phytophthora pathogens

Phytophthora pathogens are a persistent threat to the world's commercially important agricultural crops,including potato and *** strategies aim at reducing crop losses rely mostly on disease-resistance breeding and chemical pesticides,which can be frequently overcome by the rapid adaptive evolution of *** crops with intrinsic disease resista nee offer a promising alternative and con tinue to be ***,we explored Phytophthora-derived PI3P(phosphatidylinositol 3-phosphate)as a novel control target,using proteins that bind this lipid to direct secreted anti-microbial peptides and proteins(AMPs)to the surface of Phytophthora *** transgenic Nicotiana benthamiana,soybean,and potato plants,significantly enhanced resistance to different pathogen isolates was achieved by expression of two AMPs(GAFP1 or GAFP3 from the Chinese medicinal herb Gastrodia elata)fused with a PI3P-specific binding domain(FYVE).Using the soybean pathogen *** as an example,we demonstrated that the FYVE domain could boost the activities of GAFPs in multiple independent assays,including those performed in vitro,in vivo,and in *** analysis of *** PI3K1 and PI3K2 genes of this pathogen confirmed that the enhanced activities of the targeted GAFPs were correlated with PI3P levels in the ***,our study provides a new strategy that could be used to confer resistance not only to Phytophthora pathogens in many plants but also potentially to many other kinds of plant pathogens with unique targets.

GS6, A Member of the GRAS Gene Family,Negatively Regulates Grain Size in Rice

Grain size is an important yield-related trait in rice. Intensive artificial selection for grain size during domestication is evidenced by the larger grains of most of today＇s cultivars compared with their wild relatives. However, the molecular genetic control of rice grain size is still not well characterized. Here, we report the identification and cloning of Grain Size 6 （GS6）, which plays an important role in reducing grain size in rice. A premature stop at the ＋348 position in the coding sequence （CDS） of GS6 increased grain width and weight significantly. Alignment of the CDS regions of GS6 in 90 rice materials revealed three GS6 alleles. Most japonica varieties （95%） harbor the Type I haplotype, and 62.9% of indica varieties harbor the Type II haplotype. Association analysis revealed that the Type I haplotype tends to increase the width and weight of grains more than either of the Type II or Type III haplotypes. Further investigation of genetic diversity and the evolutionary mechanisms of GS6 showed that the GS6 gene was strongly selected in japonica cultivars. In addition, a ＂ggc＂ repeat region identified in the region that encodes the GRAS domain of GS6 played an important historic role in the domestication of grain size in rice. Knowledge of the function of GS6 might aid efforts to elucidate the molecular mechanisms that control grain development and evolution in rice plants, and could facilitate the genetic improvement of rice yield.

Rodent biology and management: Current status, opinion and challenges in China

China experiences serious agricultural damages from rodents, and, correspondingly, researchers in China are actively studying rodent biology and management. Following the popularization of ecologically-based rodent pest management and extension of the basic state policy of Ecological Civilization Construction in China, recent efforts in rodent control technologies are mainly focused on eliminating side effects of rodenticides and developing environmentally friendly techniques. Considering ecological functions of rodents to test their interaction with the environment is a promising design for understanding rodent populations in local ecosystems and for developing ecologically-based rodent management. Using ecological phenotypes and applying advanced techniques of molecular biology, genetics, and epigenetics might be useful for illustrating the deep regulatory mechanisms of fluctuations in rodent populations. However, facing the dual characteristics of rodents as pests and their roles as keystone species in many ecosystems, it is still a challenge to establish reasonable local thresholds to balance rodent management with the negative effects of rodent control on the environment in the future.

SENP6 restricts the IFN-I-induced signaling pathway and antiviral activity by deSUMOylating USP8

Warming decreases photosynthates and yield of soybean [Glycine max(L.) Merrill] in the North China Plain

Understanding the responses of field crops such as soybean to climate warming is critical for economic development and adaptive management of food security. A field warming experiment was conducted using infrared heaters to investigate the responses of soybean phenology, photosynthetic characteristics, and yield to climate warming in the North China Plain. The results showed that 0.4 °C and 0.7 °C increases in soybean canopy air and soil temperature advanced anthesis stage by 3.8 days and shortened the length of entire growth stage by 4.5 days. Warming also decreased the leaf photosynthetic rate by 6.6% and 10.3% at the anthesis and seed filling stages, respectively, but increased the leaf vapor pressure deficit by 9.4%, 15.7%, and 14.1% at the anthesis, pod setting, and seed filling stages,respectively. However, leaf soluble sugar and starch were decreased by 25.6% and 20.5%,respectively, whereas stem soluble sugar was reduced by 12.2% at the anthesis stage under experimental warming. The transportation amount of leaf soluble sugar and contribution rate of transportation amount to seed weight were reduced by 58.2% and 7.7%, respectively,under warming. As a result, warming significantly decreased 100-seed weight and soybean yield by 20.8% and 45.0%, respectively. Our findings provide better mechanistic understanding of soybean yield response to climate warming and could be helpful for forecasting soybean yield under future climate warming conditions.
