Determination of 14 Organophosphorus Pesticide Residues in Mutton by Gel Permeation Chromatography-Gas Chromatography-Mass Spectrometry(GPC-GC-MS)

[Objectives]This study was conducted to purify mutton samples by gel permeation chromatography(GPC).[Methods]Fourteen organophosphorus pesticide residues in samples were qualitatively and quantitatively analyzed by gas chromatography-mass spectrometry(GC-MS)in selective ion scanning mode(SIM).[Results]The organophosphorus pesticide standard solutions showed good linearity in the mass concentration range of 0.1-10.0μg/ml with correlation coefficients(r)not lower than 0.999,and the detection limits(S=3 N)ranged from 0.01 to 0.05 mg/*** average recovery values were in the range of 80.2%-99.7%,with relative standard deviations(RSDs,n=3)in the range of 1.8%-6.3%,at the addition levels of 0.5,1.0 and 2.0 mg/kg.[Conclusions]The method is simple,sensitive and accurate,and can be used for the determination of organophosphorus pesticide residues in mutton.

Chestnut leaves and bran were evenly mixed according to different mass ratios,and the mixtures were sprayed and inoculated with a certain amount of prepared EM microbial liquid,and then compressed into 70 cm×40 cm×30 cm blocks by a fully automatic yellow-storage block compression and packaging *** obtained blocks were packaged and sealed with plastic film,and placed in a freely ventilated place for more than 15 d of anaerobic fermentation,so as to obtain compressed fermented chestnut leaf block feed.[Results]Through the detection and analysis of nutritional components in the compressed fermented chestnut leaf block feed in the laboratory,the protein content was equal to or greater than 10.2%,which was 44.68%higher than that of unfermented chestnut leaf feed;the tannin content was equal to or greater than 638 mg/kg,which was 18.41%lower than that of the original feed;and the crude fiber content was equal to or greater than 19.5%,which was 14.09%lower than that of the original feed.[Conclusions]This study improves the palatability of chestnut leaf feed,increases the use efficiency of feed and reduces feeding *** is worth popularizing.

UPLC-QTOF/MS Method for Screening and Determination of 59 Non-labeled Components in Veterinary drug Preparations

[Objectives]This study was conducted to provide an accurate and reliable method for rapid high-throughput screening of veterinary drug preparations.[Methods]For the matrixes of veterinary drug preparations,high-performance liquid chromatography-high resolution quadrupole time-of-flight mass spectrometry(UPLC-QTOF/MS)was used to establish a fast screening method for 59 non-standard components in five categories of antiviral agents,aminoglycosides,quinolones,sulfonamides,and tetracyclines in veterinary *** target drugs were separated by a Waters ACQUITY UPLC HSS T3 chromatographic column(50 mm×2.1 mm,1.8μm),and data were collected in the positive ion *** separation of the 59 drugs was achieved within 7 min.[Results]In the concentration range of 0-100 ng/ml,each drug showed a good linear relationship,and the correlation coefficients were all greater than *** detection limits of the 59 drugs were in the range of 0.1-0.5 mg/ml,and the recovery under the addition concentration of 5 mg/ml was in the range of 85.2%-103.8%.[Conclusions]The method is fast,simple,accurate,and highly sensitive,and is suitable for high-throughput screening and qualitative identification of non-standard components in veterinary drug preparations.

Optimization of Molecular Distillation for Removal of Limonene from Ponkan Essential Oil Using Response Surface Methodology

Ponkan (Citrus reticulate Blanco) is one of the most popular citrus fruits for daily consumption and processing in China. In this study, limonene was removed from ponkan essential oil (PEO) by using an effective methodology to estimate the optimum levels of three independent variables of molecular distillation (MD) in order to minimize the limonene content and (b) maximize the removal rate. A three-factor Box-Behnken design (BBD) was employed to evaluate the effect of distilling temperature (45.00~55.00 ℃), wiper rolling speed (150.00~250.00 r/ min) and distilling pressure (25.00~35.00 Pa). The optimum parameters within the experimental range of the variables were extrapolated to be: distilling temperature 47.00℃, wiper rolling speed 203.00 r/min and distilling pressure 30.00 Pa. Optimal values extrapolated by response surface methodology (RSM) for the limonene content and removal rate of the desirable PEO were 47.82% and 84.67%, respectively.

[Objectives]This study was conducted to understand the effects of microbial inocula on the biological yield and nutritional components of corn.[Methods]Silage corn varieties suitable for planting in Hebei Province were selected,and set with an experimental group and a control check group ***,at the late stage of milk ripening of silage corn growth cycle,the nutritional components and indexes of silage corn were determined and analyzed by a near-infrared analyzer and the NIRS ***,the biological yield of silage corn was determined at the maturation stage of its growth period,aiming at comprehensively evaluating the regulation effects of the microbial inoculum tested on silage corn from the aspects of basic nutritional components,mineral element contents,energy and related indexes of corn,combined with agronomic characters and biological yield.[Results]The microbial inoculum improved the biological yield and nutritional indexes of silage corn,and had a positive regulation effect on the growth of silage corn.[Conclusions]The results of this study provide a theoretical basis for popularizing the planting methods and cultivation methods of silage corn with high yield and high quality.
