Unlocking the future:Mitochondrial genes and neural networks in predicting ovarian cancer prognosis and immunotherapy response

BACKGROUND Mitochondrial genes are involved in tumor metabolism in ovarian cancer(OC)and affect immune cell infiltration and treatment *** To predict prognosis and immunotherapy response in patients diagnosed with OC using mitochondrial genes and neural *** Prognosis,immunotherapy efficacy,and next-generation sequencing data of patients with OC were downloaded from The Cancer Genome Atlas and Gene Expression *** genes were sourced from the MitoCarta3.0 *** discovery cohort for model construction was created from 70% of the patients,whereas the remaining 30% constituted the validation *** the expression of mitochondrial genes as the predictor variable and based on neural network algorithm,the overall survival time and immunotherapy efficacy(complete or partial response)of patients were *** In total,375 patients with OC were included to construct the prognostic model,and 26 patients were included to construct the immune efficacy *** average area under the receiver operating characteristic curve of the prognostic model was 0.7268[95% confidence interval(CI):0.7258-0.7278]in the discovery cohort and 0.6475(95%CI:0.6466-0.6484)in the validation *** average area under the receiver operating characteristic curve of the immunotherapy efficacy model was 0.9444(95%CI:0.8333-1.0000)in the discovery cohort and 0.9167(95%CI:0.6667-1.0000)in the validation *** The application of mitochondrial genes and neural networks has the potential to predict prognosis and immunotherapy response in patients with OC,providing valuable insights into personalized treatment strategies.

In non-small cell lung cancers,the non-squamous and squamous subtypes（nsqNSCLC and sqNSCLC） exhibit disparities in pathophysiology,tumor immunology,and potential genomic correlates affecting responses to immune checkpoint inhibitor（ICI）-based *** our inhouse training cohort（n=85）,the presence of the LRP1B deleterious mutation（LRP1B-del） was associated with longer and shorter progression-free survival（PFS） on ICIs alone in nsqNSCLCs and sqNSCLCs,respectively(Pinteraction=0.008).These results were validated using a larger public ICI cohort(n=208,Pinteraction<0.001).Multiplex immunofluorescence staining revealed an association between LRP1B-del and increased and decreased numbers of tumor-infiltrating CD8+T cells in nsqNSCLCs（P=0.040） and sqNSCLCs（P=0.014）,*** the POPLAR/OAK cohort,nsqNSCLCs with LRP1B-del demonstrated improved PFS benefits from atezolizumab over docetaxel(hazard ratio（HR）=0.70,P=0.046),whereas this benefit was negligible in those without LRP1B-del（HR=1.05,P=0.64）.Conversely,sqNSCLCs without LRP1B-del benefited more from atezolizumab（HR=0.60,P=0.002） than those with LRP1B-del（HR=1.30,P=0.31）.Consistent results were observed in the in-house CHOICE-01 cohort,in which nsqNSCLCs with LRP1B-del and sqNSCLCs without LRP1B-del benefited more from toripalimab plus chemotherapy than from chemotherapy alone(Pinteraction=0.008).This multi-cohort study delineates the antithetical impacts of LRP1B-del in nsqNSCLCs and sqNSCLCs on predicting the benefits from ICI alone or with chemotherapy over chemotherapy *** findings highlight the distinct clinical utility of LRP1B-del in guiding treatment choices for nsqNSCLCs and sqNSCLCs,emphasizing the necessity for a detailed analysis based on pathological subtypes when investigating biomarkers for cancer therapeutics.

Evaluation of serum and pleural levels of endostatin and vascular epithelial growth factor in lung cancer patients with pleural effusion

Objective:To evaluate the diagnostic value of endostatin(ES),vascular endothelial growth factor (VEGF) and careinoembryonie antigen(CEA) in both serum and pleural effusion of lung cancer ***:Levels of ES,VEGF and CEA in 52 malignant pleural effusion due to lung cancer and 50 patients with non-malignant disease were measured by using sandwich enzymelinked immunosorbent assay and microparticle enzyme ***:The *** and CEA levels in pleural effusion and serum,and their ratio(F/S) were higher in lung cancer group than that in benign group,and the differences were statistically significant(P<0.05).The diagnostic efficiency of ES+VEGF for lung cancer was superior to either single *** diagnostic efficiency of ES+VEGK+CEA was superior to either ES+VEGF or ES+***: The results suggest that ES,VEGF and CEA might be useful in the differentiation between benign and malignant pleural effusion due to lung *** comparison with either single determination of concentration in serum or pleural fluid,the couiljined detection of two or three markers is of important clinical significance in the diagnosis of lung cancer.

ESTABLISHMENT AND CHARACTERIZATION OF HUMAN MALIGNANT PLEURAL MESOTHELIOMA CELL LINE SMC-1

A cell line （SMC-1） has been established from the resected tissue of a patient with malignant pleural mesothelioma which has been pathologically confirmed to be the mixed type. The cell line is characterized by the pleomorphy and multilaycred growth of the cells with the population-doubling time of approximately 32h, the highest mitotic index of 46—50‰ and the modal chromosome number of 69—70. Transplantation of the cells into the anireals can produce tumors bearing histological resemblance to the original material. The cultured cells, xenografted tumor cells and the cells from the host specimen show similar histochemical expressions. These data meet the requirements of a human malignant pleural mesothelioma cell line. The establishment of this cell line will serve as a useful tool in clinical research of diagnosis and treatment of this devastating disease.

Efficacy of Moxifloxacin against Mycobacterium abscessus in Zebrafish Model in vivo

The Relationship Between THE PATHOLOGICAL CHANGES AND Response Rate (RR) IN Non-small Cell Lung Cancer Treated by Neoadjuvant Chemotherapy with Mitomycin (MMC), Vindesine (VDS) and Cisplatin (DDP) Combination

Objective: To explore the change of pathology and the clinical response rate treated by neoadjuvant chemotherapy with MVP regimen for non-small cell lung cancer. Methods: This is a randomized study in patients with stage I-lIIa. Among them, 46 patients enrolled in neoadjuvant chemotherapy treated by 1-2 course MVP regimen. MMC was given 6 mg/M2 by intravenous (I.V.) infusion on day1, VDS 2.5-3 mg/M2 I.V. on day1, 8 and/or day15, DDP 90 mg/M2 I.V. on day1. The treatment was recycled every 28 days. The clinical RR evaluated with WHO criteria. All surgical samples were classified with pathology. Results: The overall response rate in 2 courses chemotherapy is better than that in 1 course (P<0.01). The number of patient with pathology grade I-II in 2 course chemotherapy is higher than that in 1 course (P<0.01). But the RR can not completely translated into pathology grade I-II. The pathology grade I-II is closely related with tumor involvement (T) (P<0.01) but not closely related with regional lymph node metastasis (N). It is reasonable to use RR together with PCR to judge the chemotherapy response. NR patients can not be regard as chemotherapy failure. No serve toxicities and surgical mortality were observed. Conclusion: MVP regimen is an effective neoadjuvant treatment regimen for I-IIIa NSCLC.

On the Detection of COVID-19 from chest X-Ray Images Using CNN-Based Transfer Learning
