Acyl-CoA:cholesterol acyltransferase(ACAT) is an intracellular exclusive enzyme which utilizes long-chain fatty acyl-CoA and free cholesterol as substrates to form cholesteryl esters. It is a key enzyme involved in maintaining cellular cholesterol homeostasis, and plays important roles in some serious human diseases including atherosclerosis, Alzheimer’s disease(AD) as well as cancer. In mammals, two ACAT genes encoding ACAT1 and ACAT2 have been identified. In this work, we report the impact of human ACAT1 on the dynamic free cholesterol in plasma membrane(PM-FC) of neural cell line SK-N-SH. First, the Filipin staining and PM-FC determining assays indicate that the increase of the PM-FC content of neural cell line SK-N-SH is in dose and time dependent manners by the ACAT1 inhibition, while the western blot analyses display that the α-processing of amyloid precursor protein(APP) mainly on the plasma membrane can be inversely decreased. More importantly, we have observed that the decreased α-processing of APP by the ACAT1 inhibition can be conversely enhanced when depleting the increased PM-FC directly by methyl-β-cyclodextrin, meaning that the ACAT1 inhibition can impact the dynamic PM-FC which leads to the change of APP α-processing on the plasma membrane of neural cell line SK-NSH. Then, under the different levels of cellular cholesterols by inhibiting the absorption, efflux, synthesis of cholesterolsand/or increasing cholesterols by adding LDL or mevalonate in culture media, the results show that the ACAT1 inhibition can evidently impact the dynamic PM-FC. Finally, further evidences confirm that the α-processing of APP affected by the dynamic PM-FC through the ACAT1 activity change can be mimicked by the RNAi knockdown and overexpression of ACAT1 in neural cell line SK-N-SH. These data suggest that ACAT1 impacting the dynamic PM-FC may play important role in neural cells.

The large-scale synthesis of platinum-free electrocatalysts for the oxygen reduction reaction(ORR)remains a grand *** report the large-scale production of stable and active ORR electrocatalysts based on iron,an earth-abundant element.A core–shell zeolitic imidazolate framework–tannic acid coordination polymer composite(ZIF-8@K-ta)was utilized as the catalyst precursor,which was transformed into iron atoms dispersed in hollow porous nitrogen-doped carbon capsules(H-Fe-N_(x)-C)through ion exchange and pyrolysis.H-Fe-N_(x)-C fea-tures site-isolated single-atom iron centers coordinated to nitrogen in graphitic layers,high levels of nitrogen doping,and high permeability to incoming *** from these characteristics,H-Fe-N_(x)-C demonstrated efficient electrocatalytic activity(E_(1/2)=0.92 V,***)and stability towards the ORR in both alkaline and acidic *** ORR performance,it surpassed the majority of recently reported Fe-N-C catalysts and the standard Pt/C *** addition,H-Fe-N_(x)-C showed outstanding tolerance to methanol.

The Inner Split in Genetics of Wu Colloquial Expression“[hÃɲ^(33) paʔ^(3) lÃɲ^(33) tãɲ^(31)]”

“[ã^(44) kã^(44) paʔ^(5) lã44 tã^(53)]”(Chinese characters transcription“昂更八冷打”)is an colloquial expression in Tongxiang dialect of Suzhou-Jiaxing-Huzhou Sub-branch,taihu Branch,*** describes an awkward state with unfinished *** similar expressions can be found in other regions of taihu Branch,including Yixing,Changzhou,Shaoxing and Ningbo,usually serving as“all”,while the expression“[hÃɲ^(33) paʔ^(3) lÃɲ^(33) tãɲ^(31)]”(“亨八冷打”)in shanghai Sub-branch indicates“the remaining parts”when used alone.“[hÃɲ^(33) paʔ^(3) lÃɲ^(33) tãɲ^(31)]”stands for“all”only when followed by a word with meaning of“adding together”.In this paper,it is concluded and deduced from other similar morphemes that the contradictory connotations are related to an“inner split”process of an original *** similar morphemes in other Chinese dialects(including Cantonese,Hakka,Jianghuai and Min),the following analysis proposes three possible original expressions and correlating trajectories of this inner phonetic-semantic split process,and selects a most credible way based on the rhyming dictionary and previous reconstructive models of ancient Chinese *** presumptive analysis indicates that a broader“split and derivate”process generating new morphemes exists widely in Sinitic dialects as a natural company of phonetic features in Chinese language flow.

寻找量子色动力学(Quantum Chromodynamics,QCD)相变信号是当前重离子碰撞实验的一个基础科学目标,对理解强相互作用物质的性质、大爆炸初期的宇宙演化、超新星的爆发机制、致密星的内部结构以及双中子星并合产生的引力波等重要前沿问题都具有重要意义。当重离子碰撞中发生非连续QCD相变时,引发的强相互作用物质的密度涨落与关联可以通过轻原子核的产额观察量来探测。核子间的多体关联决定了重离子碰撞中轻核的产额。特别地,基于核子并合模型对于轻核产额的计算表明质子(p)、氘核(d)和氚核(t)的产额比值N_(t)N_(p)/Nd_(2)敏感于核子密度涨落与关联,是一个较好的寻找QCD相变信号的可观测量。进一步,基于输运模型对于重离子碰撞中的手征相变的模拟发现,当系统的相轨迹经过一阶相变区域时,轻核产额比值N_(t)N_(p)/Nd_(2)有明显增加。这些结果为利用重离子碰撞中的轻核产生寻找QCD相变提供了理论依据。
