The“harmful algae and algal toxins in coastal waters of China:investigation and database”project(HAATC)is funded(US$2.3 million)by the Ministry of Science and Technology of China for 60 months starting on the 1st Jan *** the 1990s,national projects and other related work have greatly improved harmful algal bloom(HAB)research in ***,the existing research on HABs is often limited to specific sea areas or specific algal bloom disasters,which is insufficient for understanding the long-term evolution of HAB occurrence under the influence of intensified human activity and global *** HAATC project intends to determine the overall distribution of marine harmful microalgae and macroalgae,and marine algal toxins(in phytoplankton and shellfish)through a large-scale investigation in the coastal waters of China and continuous investigation in key sea *** also aims to observe biotic and abiotic processes during harmful red tides and green tides,and establish an open database and query platform of HABs in *** research consortium,led by chief scientist *** YAN,includes approximately 100 scientists and *** than 2 years of work have shown the increased distribution of HAB species(including cysts)and algal toxins in China’s coastal waters,the increased occurrence of macroalgae blooms(mainly by Ulva prolifera and Sargassum horneri)in the Southern Yellow Sea,increased amount of causative species being identified due to the use of molecular technologies in taxonomy,and new records of algal toxins such as azaspiracid,cycloimines and neurotoxinsβ-N-methylamino-l-alanine have been detected,which complicates the research in algal toxins and ***,the project will provide systematic basic data and techniques for the rapid and accurate identification of harmful algae species to support the research,monitoring,and control of harmful algal blooms,as well as seafood safety management in China.

Synthesis and Characterization of Eu+++Doped Y2O3(Red Phosphor) and Tb+++Doped Y2O3(Green Phosphor) by Hydrothermal Processes

Eu+++ and Tb+++ doped Y2O3 nanoparticles have been synthesized by hydrothermal process using yttrium oxo-isopropoxide Y5O(OPri)13 as precursor (OPri = isopropxy). X-ray diffraction (XRD), transmission electron microscopy (TEM), nanoparticle size analyzer and photoluminescence (PL) spectroscopy have been used to characterize these powders. The as synthesized powders gave very sharp peak in the X-ray diffraction suggesting crystalline particles with average particle size between 28 - 51 nm for Eu+++ doped Y2O3 nanoparticles and 43 - 51 nm for Tb+++ doped Y2O3 nanoparticles annealed at 300℃ for 3 h, 4 h and 5 h, which could be unique in comparison to other reports. Transmission electron micrograph investigation of the particles shows single dispersed particles along with agglomerates. The ratio of intensities of transitions in the europium and terbium emission spectrum have been used as structural probe to indicate the local environment around Eu+++ and Tb+++ in the Y2O3 particles.

Three deep-sea black corals belonging to the family Schizopathidae,are reported from two seamounts on the Caroline Ridge in the tropical Western Pacific:Umbellapathes parva *** depth of 1488-1766 m,Telopathes *** MacIsaac&Best,2013 with depth of 937-1016 m,and Stauropathes ***(Roule,1905)with depth of 942-1353 *** latter two species are the first records in the Western Pacific *** *** characterized by monopodial corallum,relatively long unpinnulated stalk with complex pinnulated branche s developing from the lowermost primary pinnules and small spine *** differs from two known congeners by much smaller spine s and the present of secondary *** MacIsaac&Best,2013 has sparsely branched corallum,relatively long and simple pinnules arranged in subopposite or alternate ***(Roule,1905)is characterized by the corallum with almost planar pinnulated branches,and pinnules arranged in subopposite *** results indicate that the corallum size and shape are variable in conspecific specimens particularly those at different growth *** contrast,the size of the polypar spines as well as the abpolypar spines have little variation and can be served as a main differing feature for schizopathid *** phylogenetic analyses using nuclear internal transcribed spacer region(spanning partial 18 S rDNA,ITS 1,5.8 S,ITS2,and partial 28 S rDNA)and two mitochondrial fragments cox3-IGR-coxl(COI)and trnW-IGR-nad2(NAD2)showed Telopathes,Stau ropathes,and Bathypathes had close relationships,and Umbellapathes formed a sister clade with Alternatipathes.

Benchmarking Intrinsic Promoters and Terminators for Plant Synthetic Biology Research

The emerging plant synthetic metabolic engineering has been exhibiting great promise to produce either value-added metabolitesor therapeutic proteins. However, promoters for plant pathway engineering are generally selected empirically. The quantitativecharacterization of plant-based promoters is essential for optimal control of gene expression in plant chassis. Here, we used *** leaves and BY2 suspension cells to quantitatively characterize a library of plant promoters by transient expressionof firefly/Renilla luciferase. We validated the dual-luciferase reporter system by examining the correlation between reporterprotein and mRNA levels. In addition, we investigated the effects of terminator–promoter combinations on gene expressionand found that the combinations of promoters and terminators resulted in a 326-fold difference between the strongest andweakest performance, as reflected in reporter gene expression. As a proof of concept, we used the quantitatively characterizedpromoters to engineer the betalain pathway in N. benthamiana. Seven selected plant promoters with different expressionstrengths were used orthogonally to express CYP76AD1 and DODA, resulting in a final betalain production range of 6.0–362.4 μg/g fresh weight. Our systematic approach not only demonstrates the various intensities of multiple promoter sequencesin N. benthamiana and BY2 cells but also adds to the toolbox of plant promoters for plant engineering.
