Lymphocyte-to-monocyte ratio predicts survival of patients with hepatocellular carcinoma after curative resection

AIM: To investigate the prognostic value of preoperative lymphocyte-to-monocyte ratio(LMR) in patients with hepatocellular carcinoma(HCC) undergoing curative hepatectomy.METHODS: Clinicopathological data of 210 hepatitis B virus(HBV)-associated HCC patients who were treated by radical hepatic resection between 2003 and 2010 were retrospectively analyzed. None of the patients received any preoperative anticancer therapyor intraoperative radiofrequency ablation. The diagnosis was confirmed by pathological examination after surgery. Absolute peripheral blood lymphocyte and monocyte counts were derived from serum complete blood cell count before surgery,and LMR was calculated by dividing lymphocyte count by monocyte count. The best cutoff was determined by receiver operating characteristics(ROC) curve analysis. Correlations between LMR levels and clinicopathological features were assessed using the χ2 test. Survival outcomes were estimated using the Kaplan-Meier method and compared by the log-rank test. Univariate and multivariate analyses were performed to evaluate the prognostic impact of LMR and other clinicopathological factors on overall survival(OS) and recurrence-free survival(RFS),using the Cox proportional hazards model.RESULTS: The optimal cutoff value of LMR for survival analysis was 3.23,which resulted in the most appropriate sensitivity of 55.3% and specificity of 74.7%,with the area under the curve(AUC) of 0.66(95%CI: 0.593-0.725). All patients were dichotomized into either a low(≤ 3.23) LMR group(n = 66) or a high(> 3.23) LMR group(n = 144). A low preoperative LMR level was significantly correlated with the presence of cirrhosis,elevated levels of total bilirubin and larger tumor size. Patients with a low LMR level had significantly reduced 5-year OS(61.9% vs 83.2%,P < 0.001) and RFS(27.8% vs 47.6%,P = 0.009) compared to those with a high LMR level. Objective:To explore the value of transrectal ultrasonography(TRUS)for tumor node metastasis(TNM)restaging for patients with locally advanced rectal cancer after neoadjuvant chemoradiotherapy(neo-CRT).Methods:One hundred and forty-nine patients with locally advanced rectal cancer(cT3-4 or cN+)who underwent TRUS after neo-CRT were retrospectively reviewed.TRUS restaging was compared with the results of post-operative pathological TNM findings.Results:After neo-CRT,the accuracy of TRUS for diagnosing T-staging was 30.9%,with 60.4%(90/149)of cases overestimated.The sensitivity of TRUS for T-staging(T0 vs T1 vs T2 vs T3 vs T4)were 16.3%,0%,12.5%,42.6%and 75.0%,respectively.The accuracy of TRUS for diagnosing N-staging after neo-CRT was 81.2%,with the sensitivities of N0 and N+were 93.3%and 31.0%,respectively.After neo-CRT,27.5%(41/149)of patients achieved pathologically complete response(pCR).The sensitivity,specificity,positive predictive value and negative predictive values of TRUS for pCR were 17.1%,99.1%,87.5%and 75.9%,respectively.Conclusions:TRUS can be applied for restaging T4 and N0,and has potential for screening out patients with pCR in those with locally advanced rectal cancer after neo-CRT,although some stages are overestimated for T-staging and its sensitivity for predicting pCR is low.

Optical coherence tomography angiography(OCTA),an extent function of traditional optical coherence tomography(OCT),is a non-invasive,high-resolution imaging system designed to display vascular networks.The fundamental principle of OCTA is to achieve the signal of blood flow based on the analysis of complex OCT signal,amplitude of OCT signal or phase of OCT signal.OCTA can display and monitor the vascular abnormalities in patients with diabetic retinopathy(DR),including microaneurysms,vessel density(VD),nonperfusion,neovascularization,and other lesions.OCTA offers a new and potential horizon in the monitor of the DR progress and evaluation of DR treatment.

GNSS is an essential source of information for daily life,providing positioning and timing data.However,due to the low power of satellite navigation information at the receiving end,the open signal structure,as well as with the development of spoofing technology,the problem of spoofing and jamming at the receiving end has become increasingly severe.The GNSS spoofing detection technology based on DOA offers robust detection performance,which can adapt to various scenarios.To address the issues of low angular resolution and poor detection performance in a low SNR environment,we propose a CNNDOA-based spoofing detection method.Firstly,we use the Toeplitz matrix reconstruction algorithm to estimate the DOA of the coherent signal.Then,we use the DOA spectrum generated by random angle distribution and random SNR as samples,with the DOA value and authenticity of the signal used as labels for CNN network training.When the power ratio of the spoofing signal to the authentic signal is greater than 1dB(SNR=0dB),the detection accuracy of the spoofing signal is nearly 90%.Compared to the PI algorithm and the ADBF algorithm,the proposed model has higher resolution and robustness.

Background and Objective: Early diagnosis of nasopharyngeal carcinoma (NPC) is difficult due to the insufficient specificity of the conventional examination method. This study was to investigate potential and consistent biomarkers for NPC, particularly for early detection of NPC. Methods: A proteomic pattern was identified in a training set (134 NPC patients and 73 control individuals) using the surface-enhanced laser desorption and ionization-mass spectrometry (SELDI-MS), and used to screen the test set (44 NPC patients and 25 control individuals) to determine the screening accuracy. To confirm the accuracy, it was used to test another group of 52 NPC patients and 32 healthy individuals at 6 months later. Results: Eight proteomic biomarkers with top-scored peak mass/charge ratios (m/z) of 8605 Da, 5320 Da, 5355 Da, 5380 Da, 5336 Da, 2791 Da, 7154 Da, and 9366 Da were selected as the potential biomarkers of NPC with a sensitivity of 90.9% (40/44) and a specificity of 92.0% (23/25). The performance was better than the current diagnostic method by using the Epstein-Barr virus (EBV) capsid antigen IgA antibodies (VCA/IgA). Similar sensitivity (88.5%) and specificity (90.6%) were achieved in another group of 84 samples. Conclusion: SELDI-MS profiling might be a potential tool to identify patients with NPC, particularly at early clinical stages.

Genome sequencing and analysis of a granulovirus isolated from the Asiatic rice leafroller, Cnaphalocrocis medinalis

The complete genome of Cnaphalocrocis medinalis granulovirus(CnmeGV) from a serious migratory rice pest, Cnaphalocrocis medinalis(Lepidoptera: Pyralidae), was sequenced using the Roche 454 Genome Sequencer FLX system(GS FLX) with shotgun strategy and assembled by Roche GS De Novo assembler software. Its circular double-stranded genome is 111,246 bp in size with a high A+T content of 64.8% and codes for 118 putative open reading frames(ORFs). It contains 37 conserved baculovirus core ORFs, 13 unique ORFs, 26 ORFs that were found in all Lepidoptera baculoviruses and 42 common ORFs. The analysis of nucleotide sequence repeats revealed that the CnmeGV genome differs from the rest of sequenced GVs by a 23 kb and a 17 kb gene block inversions, and does not contain any typical homologous region(hr) except for a region of non-hr-like sequence. Chitinase and cathepsin genes, which are reported to have major roles in the liquefaction of the hosts, were not found in the CnmeGV genome, which explains why CnmeGV infected insects do not show the phenotype of typical liquefaction. Phylogenetic analysis,based on the 37 core baculovirus genes, indicates that CnmeGV is closely related to Adoxophyes orana granulovirus. The genome analysis would contribute to the functional research of CnmeGV,and would benefit to the utilization of CnmeGV as pest control reagent for rice production.

EZH2 is over-expressed in human colon cancer and is closely associated with tumor proliferation,metastasis and poor prognosis.Targeting and inhibiting EZH2 may be an effective therapeutic strategy for colon cancer.3-Deazaneplanocin A(DZNep),as an EZH2 inhibitor,can suppress cancer cell growth.However,the anti-cancer role of DZNep in colon cancer cells has been rarely studied.In this study,we demonstrate that DZNep can inhibit the growth and survival of colon cancer HCT116 cells by inducing cellular senescence and apoptosis.The study provides a novel view of anti-cancer mechanisms of DZNep in human colon cancer cells.

The influencing mechanism of O_(2),H_(2)O,and CO_(2)on the H_(2)S removal of food waste digestate-derived biochar with abundant minerals

Hydrogen sulfide(H_(2)S)removal has been a significant concern in various industries.In this study,food waste diges-tate-derived biochar(DFW-BC),a by-product of food waste treatment with abundant minerals,was assessed for removing H_(2)S from different simulated biogas containing oxygen(O_(2))and carbon dioxide(CO_(2))and under differ-ent moisture(H_(2)O)contents(0%and 20%)of biochar.The influencing mechanisms of the gas conditions combined with the moisture contents were also investigated.The results showed an H_(2)S removal of 1.75 mg g^(−1)for dry bio-char under pure H_(2)S,4.29 mg g^(−1)for dry biochar under H_(2)S+O_(2),5.29 mg g^(−1)for humid biochar under H_(2)S,and 12.50 mg g^(−1)for humid biochar under H_(2)S+O_(2).For dry DFW-BC,the high Fe content was responsible for the O_(2)enhancement.In contrast,O_(2)+H_(2)O activated the catalytic H_(2)S oxidation of the less reactive minerals(mainly Ca).The inhibition of CO_(2)on H_(2)S adsorption was not obvious for dry DFW-BC;the specific pore structure may have provided a buffer against the physisorption competition of CO_(2).However,when H_(2)O was present on DFW-BC,the changes in critical biochar properties and sulfur speciation as opposed to that without H_(2)O implied an evident occurrence of CO_(2)chemisorption.This CO_(2)chemisorption partially hindered O_(2)+H_(2)O enhancement,decreasing the H_(2)S removal capacity from 12.50 to 8.88 mg g^(−1).The negative effect was ascribed to mineral carbonation of CO_(2),neutralizing the alkaline surface and immobilizing metal oxides,which thus reduced the acceleration in H_(2)S dissociation and activa-tion in catalytic H_(2)S oxidation by O_(2)+H_(2)O.
