Cough is one of the most common symptoms observed in patients presenting with COVID-19,persisting for an extended duration following SARS-CoV-2 *** aim to describe the distribution of airway microbiota and explore its role in patients with post-CovID-19 chronic cough.A total of 57 patients experiencing persistent cough after infection were recruited during the Omicron wave of SARS-CoV-2 in *** microbiota profiling is assessed in nasopharyngeal swab,nasal lavage,and induced sputum samples at 4 and 8 weeks after SARS-CoV-2 *** findings reveal that bacterial families Staph-ylococcaceae,Corynebacteriaceae,and Enterobacteriaceae are the most prevalent in the upper airway,while Streptococcaceae,Lachnospiraceae,and Prevotellaceae emerge as the most prevalent bacterial families in the lower *** increase in the abundance of Staphylococcus in nasopharyngeal swab samples and of Streptococcus in induced sputum samples is observed after one ***,the abundance of Staphylococcus identified in nasopharyngeal swab samples at the baseline period emerges as an insightful predictor for improvement in cough *** conclusion,dynamic alterations in the airway microbial composition may contribute to the post-coviD-19 chronic cough progression,while the compositional signatures of nasopharyngeal microbiota could reflect the improvement of this disease.

Airborne transmission is an important mechanism of spread for both viruses and bacteria in hospitals,with nosocomial infections putting a great burden on public *** this study,we designed and manufactured a bed for pediatric clinic consultation rooms providing air isolation to protect patients and medical personnel from pathogen *** pediatric isolation bed has several primary efficiency filters and a high-efficiency particulate air filter in the bedside *** air circulation between inlet and outlet forms negative pressure to remove the patient's exhaled air timeously and effectively.A computational fluid dynamics model was used to calculate the speed of the airflow and the angle of *** this,we conducted purification experiments using cigarette smoke,Staphylococcus albus(***)and human adenovirus type 5(HAdV-5)to demonstrate the isolation *** results showed that the patient's head should be placed as close to the air inlet hood as possible,and an air intake wind speed of 0.86 m/s was *** isolation efficacy of the pediatric isolation bed was demonstrated by computational fluid dynamics *** isolation efficiency against cigarette smoke exceeded 91.8%,and against *** was greater than 99.8%,while the isolation efficiency against HAdV-5 was 100%.The pediatric isolation bed could be used where isolation wards are unavailable,such as in intensive care units and primary clinical settings,to control hospital acquired infections.

Human adenoviruses(HAdVs)can cause acute hepatitis in immunocompromised ***,it is unclear whether HAdVs are contributors to hepatitis in immunocompetent *** this study,the liver function test(LFT)results were retrospectively analyzed among children hospitalized(agerespiratory infection caused by *** transaminase(ALT)and aspartate aminotransferase(AST)levels were elevated in 7.74%and 46.89%of patients,*** patients with>2 folds of the upper limit of ALT or AST levels were infected with HAdV-7 or *** higher levels of ALT,AST,γ-glutamyl transpeptidase(γ-GT),and lower albumin levels were observed in the HAdV-7 infection group than in the HAdV-3 infection ***-55 infection led to significantly higherγ-GT,total bilirubin,and direct bilirubin levels than the other infection *** records of four patients with serial monitoring of the LFT results were further *** indicators remained abnormal during the entire hospitalization in these *** results indicate that HAdV infection is often accompanied by abnormal liver function,and HAdV-7 and HAdV-55 might be under-recognized contributors to hepatitis among children.

A Sensitive and High-Throughput Flow Cytometry-Based Assay for Measuring Antibody Neutralization of Human Adenovirus Type 3

The assessment of neutralization activity is an important step in the evaluation of neutralizing antibodies (NAbs).The traditional methods for measuring the antibody neutralization of human adenovirus type 3 (HAd V-3) are the microneutralization (MN) assay,which has insufficient sensitivity,and the plaque reduction neutralization test (PRNT),which is not suitable for high-throughput ***,we describe the development of a flow cytometry-based neutralization(FCN) assay for measuring the neutralization of sera,cell culture supernatants,and chimeric antibodies against HAd V-3 on the basis of a recombinant HAd V-3 (r HAd V-3) construct expressing the enhanced green fluorescent protein (EGFP).For flow cytometry-based assays,the optimal cell confluence was determined as 90%,and the virus was titrated using the *** established FCN assay follows the percentage law and an optimal MOI of not less than 5 9 10~(-4)was determined by using a purified chimeric *** addition,comparison of the anti-HAd V-3 NAb titers of 72 human serum samples by the MN and FCN assays,showed that both assays correlated strongly with each *** FCN assay was an improvement over the MN assay because the observation period was reduced from 3 to 1 days and data analysis could be performed objectively and ***,the newly established FCN assay allows measurement of the neutralization activity of chimeric antibodies expressed in cell culture ***,this sensitive and highthroughput FCN assay is a useful alternative to the MN assay for measuring the antibody neutralization of HAd V-3 and for screening anti-HAd V-3 NAbs in cell culture supernatants.
